Project description:Study of the impact of 4-oxo-RA on LSK (bone marrow and spleen) cell characteristics post-myocardial infarction in C57BL/6J mice. Myocardial infarction was induced in female C57BL/6J mice (aged 6 to approximately 12 weeks) through LAD occlusion. Post-surgery, mice were intraperitoneally injected with 30 mg/kg 4-oxo-RA (MI+4-oxo-RA) or the equivalent DMSO in PBS (MI+vehicle) on the 1st and 2nd days after MI. Three days post-MI, mice were euthanized. ScRNA-seq was performed on 25k LSK cells (Lin-, Sca1+, c-Kit+) that were facs sorted.

Plasma small extracellular vesicles of ischemic cardiomyopathy aggravate ventricular remodeling post-myocardial infarction through miR-223-3p-mediated dysfunction in regulatory T cells

Project description:Ischemic cardiomyopathy (ICM), caused by acute myocardial infarction (AMI), is the most common type of chronic heart failure (HF). Treg cells, maintaining peripheral immune tolerance, are dysfunctional in ICM mice. The mechanism of dysfunctional Treg cells in ICM remains unclear. Furthermore, small extracellular vesicles (sEVs) are significant in intracellular communication and tissue homeostasis. This study tends to illustrate whether plasma sEVs of ICM promote the dysfunctional state of Treg cells through miRNA and thus aggravate cardiac remodeling.In ICM patients, the number of Treg cells was increased while Foxp3 mean fluorescence intensity (MFI) was decreased in peripheral blood, indicating a dysfunctional phenotype of Treg cells. miR-223-3p, identified by sequencing using plasma sEVs of ICM patients, was upregulated and negatively correlated with blood Treg cell number. Additionally, the number of cardiac Treg cells of ICM mice was increased while Foxp3 MFI was decreased. Furthermore, plasma sEVs from ICM mice aggravate ventricular remodeling in mice post-AMI. Besides, ICM-sEVs decreased the number of Treg cells while miR-223-3p knockdown increased the number of them and promoted the expression of Foxp3 in vitro.Plasma sEVs of ICM aggravate ventricular remodeling post-AMI through miR-223-3p-mediated Treg cells dysfunction in ICM.

Gene expression signatures of CD11b+ cells in mice after myocardial infarction

Project description:To further search for potential gene targets which are involved in the pathological process of myocardial infarction, we have employed whole genome microarray expression profiling as a discovery platform to identify the expression of STAT6, β1-adrenergic receptors and inflammatory cytokines. The CD11b+ cells were isolated from spleen and bone marrow of mice by magnetic beads, and the experimental mice were divided into four time points (control, MI 1week, MI 2 weeks, and MI 4 weeks). We found the β1-AR related sympathetic signal, the STAT6 signal and inflammatory cytokines (IL-1α, IL-18 and TGF-β) are involved in the activation of CD11b+ immune cells and cardiac fibrogenesis after myocardial infarction.

Knockdown of miR-192 and miR-432 in progenitor cell-derived small extracellular vesicles enhances tissue repair in myocardial infarction

Project description:Small extracellular vesicles (sEVs) play a critical role in cardiac cell therapy by delivering molecular cargo and mediating cellular signaling. Among sEV cargo molecule types, microRNA (miRNA) is particularly potent and highly heterogenous. However, not all miRNAs in sEV are beneficial. Two previous studies utilizing computational modeling identified miR-192-5p and miR-432-5p as potentially deleterious in cardiac function and repair. Here, we show that knocking down miR-192-5p and miR-432-5p in cardiac c-kit+ cell-derived sEVs enhances the therapeutic capabilities of sEVs in vitro and in a rat in vivo model of cardiac ischemia reperfusion. miR-192-5p and miR-432-5p depleted CPC-sEVs enhance cardiac function by reducing fibrosis, enhancing mesenchymal stromal cell-like cell mobilization, and inducing macrophage polarization to the M2 phenotype. Knocking down deleterious miRNAs from sEV could be a promising therapeutic strategy for treatment of chronic myocardial infarction.
