The identification of VAMP8 interactive protein under normal and EBSS condition

ABSTRACT: VAMP8 was first identified as an endosomal SNARE that participates in diverse biological functions, including autophagy fusion. We investigated the protein interaction and modification of VAMP8 upon starvation condition compared with normal condition, we thus excised the GFP-VAMP8 band from the Coomassie staining gels that observed in approximately (25-55Kda) and sent the excised gels for mass spectrum analysis, and the same time, we identified the potential candidate of E3 ligase for targeting VAMP8.

Autophagy classically functions to protect cells and organisms during stressful conditions by catabolizing intracellular components to maintain energy homeostasis. Lysosome-autophagosome fusion is a critical step in emptying degraded unwanted contents. However, the mechanism of autophagosome fusion with lysosomes is still not fully understood. Here, we report that DNA Damage-Regulated Autophagy Modulator 1 (DRAM1) interacts with Vesicle Associated Membrane Protein 8 (VAMP8) to mediate the fusion  ...[more]
