Identification of PEDV NSP12-interacting proteins by IP-MS screening

ABSTRACT: To investigate and identify that the host proteins interacting with PEDV NSP12, we transfected the p3×Flag-NSP12 and p3×Flag-CMV-10 plasmids into 293T cells, respectively. Then the proteins were immunoprecipitated with anti-Flag antibody. PEDV NSP12 interacting proteins and identified the precipitated proteins by Liquid Chromatography-Mass Spectrometer/Mass Spectrometry (LC-MS/MS).

Screening of host proteins interacting with African swine fever virus MGF360-9L

Project description:To identify the potential host protein binding to African swine fever virus MGF360-9L, the 3 × FLAG tag MGF360-9L plasmid and empty FLAG were transfected into PK-15 cells for co-immunoprecipitation and LC-MS analysis. The cell lysates were immunoprecipitated using anti-FLAG antibody. Immunoprecipitation samples of cells transfected with empty FLAG were used as negative controls to eliminate non-specific interactions. LC-MS identified the interaction between MGF360-9L and PK-15 cell proteome. The final MGF360-9L binding protein data eliminated the nonspecific interaction through empty FLAG control. The interactions that remained were analyzed by significance analysis of interactome. Finally, 268 cellular proteins interacting with MGF360-9L were identified.

Mass spectrometry runs and analysis of host proteins co-immunoprecipitated with RABV M protein

Project description:To explore the role of the matrix protein in RABV infection of host cells, we looked for host proteins that interact with the RABV M protein. We performed Flag-tag-based immunoprecipitation of lysates of HEK293T cells transfected with plasmids expressing either N- or C-terminally Flag-tagged M protein (M-NF or M-CF, respectively), and identified host proteins that interacted with the M proteins by using mass spectrometry (MS). Proteins were detected in lysates of cells transfected with the plasmid expressing Flag-tagged M proteins but not in those of cells transfected with the control plasmid. We identified a total of 2,672 host proteins that co-immunoprecipitated with the M protein; 492 proteins co-immunoprecipitated to both M-NF and M-CF, 1,761 proteins co-immunoprecipitated with M-NF only, and 419 proteins co-immunoprecipitated with M-CF only.

Project description:In this study, we performed a ChIP-chip experiment to determine the respective regulons of RpoHI and RpoHII in Rhodobacter sphaeroides. We grew R. sphaeroides in aerobic conditions and induced either proteins ectopically and immuno-precipitated the regions of the genomic DNA interacting with the sigma factors. DNA immunoprecipitated with antibodies against RpoHI or RpoHII was labelled with Cy5 and hybridized on two-color tilling arrays (triplicates for each) with genomic DNA as an input control labelled with Cy3.
