Project description:Global gene expression patterns associated with early stage endometrial cancer have been reported, but changes in molecular expression associated with tumor grade, depth of myometrial invasion, and non-endometrioid histology have not been previously elucidated. Our group hypothesized there are unique genetic events underlying early endometrial carcinogenesis. Ninety-two samples of pathologically reviewed stage I endometrial cancers (80 endometrioid and 12 serous) with a heterogeneous distribution of grade and depth of myometrial invasion (i.e. 9 IAG1, 14 IAG2, 7 IAG3, 14 IBG1, 12 IBG2, 13 IBG3, 7 ICG1, 10 ICG2, and 6 ICG3) were examined in relation to 12 samples of atrophic endometrium from postmenopausal women. Specimens were analyzed using oligonucleotide microarray analysis and a subset of the differentially expressed transcripts was validated using quantitative PCR. Comparison of early stage cancers with normal endometrium samples by the univariate t-test with 10,000 permutations identified 900 genes that were differentially regulated by at least 4-fold at a p value of <0.001. Unsupervised analysis revealed that when compared to normal endometrium, serous and endometrioid stage I cancers appeared to have similar expression patterns. However, when compared in the absence of normal controls, they were distinct. Differential expression analysis revealed a number of transcripts that were common as well as unique to both histologic types. This data uncovers previously unrecognized, novel pathways involved in early stage endometrial cancers and identifys targets for prevention strategies that are inclusive of both endometrioid and serous histologic subtypes. Project description:Endometrium-specific Cre-Lox deletion of both Pten and Tp53 via regulation of the progesterone receptor promoter is known to generate a spontaneous carcinogenic mouse model of endometrial cancer. A mouse endometrial cancer cell line, mECC, was established from the uterine carcinoma tumor tissue of its pure strain model generated by backcrossing in C57BL/6 mice. A high-grade endometrial cancer cell line (Highly Progressive mECC: HPmECC) was established by further introducing c-Myc in mECCs. RNA sequencing analysis was performed using cultured HPmECCs and mECCs, as well as primary cultured C57BL/6 normal endometrial cells for assessing the genetic characteristics of HPmECCs.

Project description:Uterine NK cells (uNK) play a role in the regulation of placentation but their functions in non-pregnant endometrium are not understood. We have previously reported suppression of endometrial bleeding and alteration of spiral artery morphology in women exposed to asoprisnil, a progesterone receptor modulator (PRM). We now compare global endometrial gene expression in asoprisnil-treated versus control women and demonstrate a statistically significant reduction of genes in the IL-15 pathway, known to play a key role in uNK development and function. Suppression of IL-15 by asoprisnil was also observed at mRNA level (p<0.05), and immunostaining for NK cell marker CD56 revealed a striking reduction of uNK in asoprisnil-treated endometrium (p<0.001). IL-15 levels in normal endometrium are progesterone-responsive. Progesterone receptor (PR) positive stromal cells transcribe both IL-15 and IL-15RA. Thus, the response of stromal cells to progesterone will be to increase IL-15 trans-presentation to uNK, supporting their expansion and differentiation. In asoprisnil-treated endometrium, there is a marked down-regulation of stromal PR expression and virtual absence of uNK. These novel findings indicate that the IL-15 pathway provides a missing link in the complex interplay between endometrial stromal cells, uNK and spiral arteries affecting physiological and pathological endometrial bleeding. 39 Samples

Project description:Endometrial cancer represents the most frequent gynecologic malignant disease. Although several genetic alterations have been associated with increased risk, to date diagnosis and prognosis still rely on morphological features of the tumor, such as histological type, grading and invasiveness. As molecular-based classification is desirable for optimal treatment and prognosis of these cancers, , we explored the potential of lncRNAs as molecular biomarkers. To this end, we first identified by RNA sequencing (RNA-Seq) a set of lncRNAs differentially expressed in cancer vs normal endometrial tissues, a result confirmed also by analysis of RNA-Seq data of normal and cancerous endometrium from The Cancer Genome Atlas (TCGA). A significant association of a subset of these differentially expressed lncRNAs with tumor grade was then determined in 405 TCGA endometrial cancer profiles. Integrating endometrial cancer-specific expression profiles of long and small non-coding RNAs a functional association network was then identified. These results described for the first time a functional ‘core’ network, comprising small and long RNAs, whose deregulation is associated with endometrial neoplastic transformation, representing a set of cancer biomarkers that can be monitored and targeted for diagnosis, follow-up and therapy of these tumors.

Project description:Transcriptional profiling of rat EAC - comparison of non-/pre-malignant endometrium with endometrial tumors Endometrial cancer develops from the endometrium of the uterus and is the most common pelvic malignancy diagnosed in women in the western countries. Similar to all cancer diseases, endometrial cancer is a genetic disorder that results from complex patterns of genetic and epigenetic alterations involved in the malignant transformation. The genetic heterogeneity inherent in the human population, differences in the environment and life styles poses enormous difficulties when analyzing the complex patterns of genetic alterations contributing to cancer etiology. Inbred animal models constitute unique experimental genetic tools as the genetic heterogeneity and the influence of environmental factors can be readily reduced. The BDII/Han rat model is unique for spontaneous hormonal carcinogenesis since more than 90% of the female virgins spontaneously develop endometrial cancer during their life span. The possibility to perform global gene expression profiling of tumor cells would likely provide important information of the genes and pathways that are involved in EAC susceptibility and carcinogenesis. Project description:The molecular explanation for tamoxifen serving as a breast cancer treatment but displaying partial estrogenic in the uterus is not known. Previously, we reported that differential promoter context and cofactor recruitment contribute to the tissue specificity of tamoxifen. Here, we investigated the genomic basis for the partial oestrogenic activity of tamoxifen in the endometrium. We showed that tamoxifen not only affects the rate of transcription of oestrogen target genes but also targets a unique set of genes. Since oestrogen and tamoxifen are both able to bind to oestrogen receptors (ERs) and because both promote endometrial carcinogenesis, we hypothesized that the molecular effectors for ERs in endometrial carcinogenesis most likely reside in genes that are commonly targeted by oestrogen and tamoxifen. Among those target genes, we identified a paired-box gene PAX2 that is critically involved in cell proliferation and carcinogenesis in the endometrium. Our experiments also demonstrated that PAX2 is activated by oestrogen and tamoxifen in endometrial carcinomas but not in normal endometrium, and this activation is associated with cancer-linked hypomethylation of the PAX2 promoter.
