Time-Series Multi-Omics Identifies ZG16 as a Prognostic Immunoregulatory Driver of Colitis-associated Colon Cancer Development and Checkpoint Blockade Response

ABSTRACT: This dataset contains quantitative proteomic profiles across five disease stages (W0, W2, W5, W8, W11) of colitis-associated colorectal cancer (CAC) using a mouse AOM/DSS model. Proteins were extracted from tumor tissues and processed for LC-MS/MS after labeling with iTRAQ 6-plex reagents. The goal was to identify temporal dynamics of protein expression underlying chronic inflammation-induced tumorigenesis.

Time-Series Multi-Omics Identifies ZG16 as a Prognostic Immunoregulatory Driver of Colitis-associated Colon Cancer Development and Checkpoint Blockade Response

Project description:Chronic inflammation is a known driver of colitis-associated cancer (CAC), yet the trajectory of immune suppression and its reversibility remain poorly defined. Here, we performed a time-series multi-omics analysis combining paired transcriptomic and proteomic profiling, flow cytometry, and cytokine measurements in an AOM/DSS-induced model, further supported by single-cell transcriptomic and epigenomic datasets. This roadmap delineates a stepwise progressive evolution of immune suppression, capturing coordinated changes in immune cell composition, cytokine dynamics, and inflammation-associated mutagenesis throughout CAC progression. We identified ZG16 as a previously undervalued immune modulator that is progressively downregulated during CAC and CRC development. Analysis of CRC cohorts in the TCGA database confirmed that ZG16 loss correlates with PD-L1 upregulation, suppression of T cell costimulatory/proliferative programs, and both poor prognosis and resistance to immunotherapy. Functionally, ZG16 suppresses PD-L1 expression and promotes CD8+ T/NK cell infiltration, thereby reshaping the tumor immune microenvironment. In both inflammation-driven and syngeneic tumor models, recombinant ZG16 reduces tumor burden and enhances the efficacy of PD-1 blockade. These findings position ZG16 as a rheostat-like immunoregulatory factor and translationally relevant biomarker in inflammation-driven cancers.

Colon gene expression signatures in wild type and Card9-/-mice exposed to DSS induced colitis

Project description:It is crucial to decipher the host-microbiota interactions as they are involved in intestinal homeostasis and diseases. Caspase Recruitment Domain 9 (Card9) is an inflammatory bowel disease (IBD) susceptibility gene coding for an adapter protein for innate immunity toward many microorganisms. Card9 mediates colitis recovery via interleukin 22 pathway activation and Card9-/- mice have enhanced susceptibility to colitis. To reveal the mechanisms responsible of this defect in Card9-/-mice, we compared colon transcriptomics in WT and Card9-/- mice before and during DSS-induced colitis. Mice transcriptomes clusterized according to the genotype supporting a pattern clearly different in WT and Card9-/- mice. The number of up-regulated genes at day 7 was largely higher in Card9-/- compared to WT mice. Pathway analyses of the induced transcripts showed a dominance of immune-related pathway with a stronger signal in Card9-/- mice. Interestingly, NOD-like receptor signaling pathway, in which CARD9 is involved, was an exception with weaker activation in Card9-/- than in WT mice. During the recovery period at day 12, pathways involved in cell proliferation and replication were significantly activated in WT compared to Card9-/- mice confirming the healing defect in Card9-/- mice. For the induction of colitis, mice were given drinking water supplemented with 2% (w/v) Dextran sulphate sodium (DSS) for 7 days, then allowed to recover by drinking water alone for 5 additional days. 3 mice of each groups (WT and Card9-/-) were sacrified before DSS administration. 5 WT mice and 4 Card9-/- mice were sacrified 7 days after DSS administration and 5 mice of each group were sacrified at day 12.

Colon epithelial cells gene expression data of Sphingomyelin synthase 2 knockout colitis mice

Project description:Sphingomyelin synthase (SMS) 2 is the synthetic enzyme of sphingomyelin (SM), which regulates the fluidity and microdomain structure of the plasma membrane. We investigated the effect of SMS2 deficiency on dextran sodium sulfate (DSS)-induced murine colitis, and found suppression of DSS-induced inflammation in SMS2 deficient (SMS2-/-) mice. Results provide insight into the role of SMS2 in inflammation.
