Heart resident macrophage proteome of WT and CCR-/- mice treated with DOX

ABSTRACT: Heart from WT and CCR-/- mice treated with DOX was harvest, then the resident macrophage was extract and subject to TMT labeling for LC-MS/MS analysis. Three samples from WT mice were labeled with labeling reagent 126,127N and 127C, 3 samples from CCR-/- mice were labeled with labeling reagent 128N,128C and 129N. MS/MS analysis was performed on a Q Exactive Plus mass spectrometer coupled with an EASY-nLC 1200 system

Project description:Mammary organoids harvested from ErbB3 DOX-KO mice, which utilize MMTV-Cre transgene expression in the LE to cause genomic recombination at floxed ErbB3 alleles in ErbB3FL/FL were cultured in the presence or absence of doxycycline to induce ErbB3 loss. The gene expression shift following DOX-induced ErbB3 loss in the 3D organoids was examined by microarray. Gene expression patterns were interrogated in mammary organoids from ErbB3 inducible-knockout mice cultured in the presence of absence of doxycycline. Three biological replicates of the experiment were performed, resulting in a total of 6 samples (3 treatment, 3 control).

Analysis of fluorophore-labeled human cytochrome b5

Project description:Cytochrome b5 (b5) is a small accessory protein involved in several reactions. The interaction of b5 with its partner proteins, e.g., cytochrome P450 enzymes, has received much attention and has been the subject of numerous studies. In order to obtain further insights into protein-protein interactions of b5, we modified it with a green fluorophore, Alexa 488, which has a maleimide linker. The labeling was applied to both WT (without Cys) and the T70C mutant. This study revealed labeled sites in WT b5 and confirmed the Cys-selective labeling in the T70C mutant.

Project description:6C secondary MEFs were treated with Dox in mES media to turn on the Oct4, Klf4, cMyc, Sox2. Total RNA was extracted at day 0 (no Dox), day2, 5, 8, 11, 16 and 21 (with Dox) and day 30 (Dox-independent secondary iPS). RNA from Parental MEFs and Primary-iPS cells were also extracted for reference. 1B secondary MEFs were Dox treated for 5-days followed by RNA extraction. Subsequently, a culture removed of Dox treatment for an additional 5days was also analyzed.
