Hepatic NIK Protects Against Ethanol-Induced Steatosis by Stabilizing INPP5B to Suppress PI3K/AKT/ACLY Lipogenesis

Project description:Alcohol-associated liver disease (ALD) imposes a major global health burden, yet lacks FDA-approved targeted therapies. While NF-κB-inducing kinase (NIK) is recognized for its dual roles in hepatic inflammation and lipid metabolism, its specific function in ALD pathogenesis remained incompletely defined. Contrary to its established protective role against high-fat diet-induced steatosis, this study reveals a paradoxical protective function of hepatic NIK in chronic ethanol-induced steatosis. We demonstrate that liver-specific NIK deficiency unexpectedly exacerbates hepatic steatosis in chronic ethanol-fed mice. This aggravation occurs via activation of the PI3K/AKT-ATP Citrate Lyase (ACLY) signaling pathway, promoting lipogenesis. Importantly, NIK overexpression reversed steatosis in deficient mice, independent of the non-canonical NF-κB effector p52. Mechanistically, proteomics and functional studies identified Inositol polyphosphate-5-phosphatase b (INPP5B), a negative PI3K/AKT regulator, as a key downstream target. NIK stabilizes INPP5B by suppressing its ubiquitination and enhancing its interaction with Adaptor protein APPL1, thereby inhibiting PI3K/AKT/ACLY signaling. These findings unveil a novel, kinase/scaffold-dependent role for NIK in mitigating ethanol-induced steatosis through INPP5B stabilization and highlight its context-dependent, double-sided effects in ALD progression, offering new insights for therapeutic strategies.

Hepatic CYP2B10 is highly induced by binge ethanol and contributes to acute-on-chronic alcohol-induced liver injury

Project description:We altered the NIAAA (Gao Binge Model) of 8 week chronic plus binge ethanol (E8G1) to have 3 total ethanol gavages (5 g/kg; 1 every 24h) of ethanol after 8 weeks of chronic ethanol feeding (E8G3) to determine how the liver responds to chronic plus consecutive binge ethanol. Surprisingly, E8G3 treatment induced lower levels of liver injury, steatosis, inflammation, and fibrosis as compared to mice after E8G1 treatment. Microarray analyses were performed, identifying several pathways that may contribute to the observed liver adaptation to binge ethanol. In addition, cytochrome P450 2B10 (Cyp2b10) was a top upregulated gene in the E8G1 group and further upregulated in the E8G3 group, but only moderately induced after chronic ethanol consumption, which was confirmed by RT-qPCR and western blot analyses. Genetic disruption of the Cyp2b10 gene worsened liver injury in E8G1 and E8G3 mice with higher blood ethanol levels compared to wild-type control mice, while in vitro experimentation revealed that CYP2b10 did not directly promote ethanol metabolism. Metabolomic analyses revealed significant differences in hepatic metabolites from E8G1-treated Cyp2b10 KO and WT mice, and these metabolic alterations may contribute to the reduced liver injury in Cyp2b10 KO mice.

GLP-1 receptor agonism results in reduction in hepatic ethanol metabolism

Project description:Glucagon-like peptide 1 receptor (GLP-1R) agonists are used along with ethanol consumption, but their interactions are not understood. Our aim was to determine the effects of GLP-1R agonism on the liver in mouse models of high ethanol consumption. We identified that GLP-1R agonism reduced ethanol consumption, mitigated ethanol-induced upregulation of several liver metabolizing enzymes, including Cyp2e1 and also reduced Cyp2e1 independent of ethanol intake. As expected from a reduction in Cyp2e1, GLP-1R agonism resulted in increased blood ethanol levels. This occurred after a single dose of ethanol when given by gavage, and by the intraperitoneal route. This suggests that GLP-1R agonism can reduce ethanol-mediated hepatotoxicity despite continued ethanol consumption and elevate blood alcohol levels

Genome-wide analysis of nucleus accumbens gene expression after ethanol consumption in rat

Project description:The study investigated ethanol-induced alterations in the transcriptome of adult male Wistar rats. The study was divided into two parts. Part 1: 10 animals were single housed during 8 weeks and exposed to 6% ethanol solution and water simultaneously or just water (24 hours/day, 7 days/week). Fluid intake and body weight were recorded twice weekly during 8 weeks. Animals were sacrificed immediately after the last consumption measurement and the nucleus accumbens were dissected and snap frozen until further processing. Part 2: 12 rats were single housed and exposed to continuous access to ethanol and water (24 hours/day, 7 days/week). After 4 weeks, animals were divided into 2 groups, equally balanced based on previous ethanol intake, and were given additional ethanol or water through a gavage tube daily during 3 weeks. This was followed by an additional 2 weeks of continuous ethanol access and then animals were immediately sacrificed and the nucleus accumbens were dissected and snap frozen until further processing. Next, the RNA was extracted from the cells using Qiagen RNeasy Lipid tissue mini kit and the RNA concentration and quality was measured using a BioAnalyzer. RNAseq was performed using standard Illumina protocols in collaboration with SciLifeLab, Stockholm, Sweden. No difference in gene expression was detected between any groups.
