Non-modified Molecules Strategy Enable Proteome-Wide Mapping of Catechol Derivatives Interactome

ABSTRACT: Here, we presented a modification-free strategy termed oxidative addition-boronate affinity enrichment (OABA), that leverages catechol oxidation to ortho-quinones for covalent capture of nucleophilic protein residues, followed by chemoselective enrichment via boronate affin-ity. This redox-enabled approach preserves native structure-activity relationships while enabling direct mapping of catechol-protein interactomes.
