Proteomics of chronic hepatitis B across distinct clinical stages

ABSTRACT: Chronic hepatitis B (CHB) is a global public health concern characterized by multiple disease stages. Existing CHB research is largely constrained by limited access to infection sites, with most studies relying on peripheral samples such as blood, which inadequately elucidate the interplay among liver parenchyma, the immune microenvironment, and viral dynamics. Only a few studies have employed multi-omics technologies to analyze liver biopsy tissues, revealing molecular differences at the hepatic level. This study represents the first large-scale proteomic analysis of different clinical stages of CHB in liver tissue.

Circulating microRNA profiles in patients with type-1 autoimmune hepatitis

Project description:Recent studies have demonstrated that micro (mi)RNA molecules can be detected in the circulation and can serve as potential biomarkers of various diseases. This study used microarray analysis to identify aberrantly expressed circulating miRNAs in patients with type 1 autoimmune hepatitis (AIH) compared with healthy controls. Patients with well-documented and untreated AIH were selected from the National Hospital Organization (NHO)-AIH-liver-network database. They underwent blood sampling and liver biopsy with inflammation grading and fibrosis staging before receiving treatment. To further confirm the microarray data, circulating expression levels of miR-21 and miR-122 were quantified by real-time quantitative polymerase chain reaction in 46 AIH patients, 40 patients with chronic hepatitis C (CHC), and 15 healthy controls. Consistent with the microarray data, serum levels of miR-21 were significantly elevated in AIH patients compared with CHC patients and healthy controls. miR-21 and miR-122 serum levels correlated with alanine aminotransferase levels. Circulating levels of miR-21 and miR-122 were significantly reduced in AIH patients with liver cirrhosis, and were inversely correlated with increased stages of fibrosis. By contrast, levels of circulating miR-21 showed a significant correlation with the histological grades of inflammation in AIH. We postulate that aberrantly expressed serum miRNAs are potential biomarkers of AIH and could be implicated in AIH pathogenesis. Untargeted proteomic analysis of simultaneously collected human liver tissue and plasma of patients with a histological diagnosis of metabolic dysfunction-associated steatotic liver disease.

Project description:The liver tissue and plasma were collected from patients when undergoing liver biopsy for a diagnosis of metabolic dysfunction-associated steatotic liver disease (MASLD), along with controls who had no histological abnormalities. The study group included seven patients with no liver histological abnormalities and 64 patients with MASLD across its spectrum, including those with metabolic dysfunction-associated steatotic hepatitis (MASH) and those with cirrhosis. The data were compared between liver tissue and plasma, across various stages of MASLD, and for the detection of biomarkers for different stages of MASLD.

Interferon signaling related gene expression and clinical outcome of chronic hepatitis C

Project description:Chronic hepatitis C (CH) frequently developed liver cirrhosis and hepatocellular carcinoma without suitable treatment. However, peginterferon(IFN) and ribavirin therapy (combination therapy), which is present standard therapy, is imperfect effect because of several side effect. We attempt to catalog IFN related gene expression profile according to the response of combination therapy in to predict the outcome of drug-response before administration and establish the adequate treatment according to the individual clinical information. Chip1 [GPL9960-related Samples]: We quantified expression profile of 200 IFN related genes by microarray in the liver biopsy specimen from 88 CH patients without history of anti-viral therapy, 10 fatty liver patients without HCV infection were also enrolled. Chip2 [GPL9962-related Samples]: We quantified expression profile of 37 IFN related genes by microarray in the liver biopsy specimen from 88 CH patients without history of anti-viral therapy, 10 fatty liver patients without HCV infection were also enrolled.

Early transcriptional programming links progression to hepatitis C virus-induced severe liver disease in transplant patients

Project description:This study determined key transcriptional signatures associated with HCV recurrence and eventual development of severe liver disease in infected transplant patient liver biopsies over time following transplant. We identified molecular signatures associated with severe fibrosis and liver injury prior to histologic evidence of disease progression. 111 liver biopsy specimens collected longitudinally from 57 HCV-infected liver transplant patients following organ transplant. Several batches of pooled normal liver RNA samples (Utah normal pool; UNP) are included.

We quantified expression profile of 474 mature miRNAs by microarray in the liver biopsy specimen from 99 CH patients without history of anti-viral therapy.

Transcriptome patterns for Acute Cellular Rejection in Recipients with Recurrent Hepatitis C after Liver Transplantation

Project description:The histological evaluation of liver via biopsy remains as the standard for the diagnosis of both acute cellular rejection (ACR) and recurrent hepatitis C (RHC) after liver transplantation. Nevertheless, it is often difficult to diagnose ACR in HCV-positive recipients because of common co-existing and overlapping morphological changes with RHC. The aim of the study was to identify potential target genes for ACR in recipients with RHC. We analyzed 22 liver biopsy samples obtained from 21 HCV-positive recipients. The clinicopathological diagnoses for the biopsies were ACR-predominant with superimposed RHC in 9 samples (ACR group) and RHC with no ACR (non-ACR group) in the remaining. We compared the transcriptional alterations between the two groups with oligonucleotide microarray and selected 2206 genes which were significantly modulated in ACR. Subsequently we analyzed the regulatory networks in ACR using Ingenuity Pathway Analysis, and focused on 5 genes (IFNAR1, IL-12RB2, NFATC3, BMP2, and CASP8) from the core network as the target genes for ACR. In conclusion, our results demonstrated that novel transcriptome patterns of ACR and concurrent RHC were present and distinct from recipients with only RHC, suggesting that gene expression profiling may have a role in the diagnosis of ACR in recipients with hepatitis C.
