Proteomics data of hepatocytes in metabolism dysfunction associated steatohepatitis.

ABSTRACT: In this study, metabolism dysfunction associated steatohepatitis (MASH) hepatocytes and normal hepatocytes were collected from three MASH mouse models and normal diet control fixed-frozen liver sections with laser capture microdissection (LCM), respectively. Protein was processed from these LCM samples using the HDMSP method. LC-MS/MS analysis was performed with Orbitrap Astral mass spectrometer coupled with Vanquish Neo UHPLC system.

Project description:Metabolic dysfunction-associated steatohepatitis (MASH) remains a significant health challenge. Herein, we identify sphingomyelin phosphodiesterase 3 (SMPD3) as a key driver of hepatic ceramide accumulation through increasing sphingomyelin hydrolysis at the cell membrane. Hepatocyte-specific Smpd3 gene disruption and pharmacological inhibition of SMPD3 alleviate MASH, whereas reintroducing SMPD3 reverses the resolution of MASH. While healthy livers express low-level SMPD3, lipotoxicity-induced DNA damage suppresses SIRT1, triggering an upregulation of SMPD3 during MASH. This disrupts membrane sphingomyelin-ceramide balance and promotes MASH progression by enhancing caveolae-dependent lipid uptake in hepatocytes and extracellular vesicle secretion from steatotic hepatocytes to worsen inflammation and fibrosis. Thus, SMPD3 acts as a central hub integrating key MASH hallmarks. Notably, we discovered a bifunctional agent that simultaneously activates SIRT1 and inhibits SMPD3, which shows significant therapeutic potential in MASH treatment. These findings suggest that inhibition of hepatic SMPD3 restores membrane sphingolipid balance and holds great promise for developing novel MASH therapies.

Role of ALDH18A1 in regulating the progression of Metabolic dysfunction-associated steatohepatitis

Project description:Metabolic dysfunction-associated steatohepatitis (MASH) continues to be a considerable health problem. We identify Aldehyde dehydrogenase 18 family, member A1 (ALDH18A1) as a key factor in MASH aggravation via promoting liver proline accumulation. Silencing of the liver-specific Aldh18a1 gene mitigates MASH, while overexpression of ALDH18A1 exacerbates the phenotype of MASH. Mechanistically, under MASH pathological conditions, ALDH18A1 may affect hepatic lipid metabolism through increasing proline synthesis in hepatocytes. Our results indicate that the overexpression of the enzyme-inactive mutant of ALDH18A1 at the cellular level does not promote lipid accumulation in hepatocytes, in contrast to the overexpression of the wild-type ALDH18A1. Moreover, our observations of inhibited cellular mitochondrial respiratory function due to ALDH18A1 overexpression demonstrated that the overexpression of ALDH18A1, which drives higher proline concentration, is associated with impaired mitochondrial respiratory function. The results indicate that the suppression of hepatic proline production may be beneficial for the advancement of new MASH therapeutics.

Project description:Astrocyte elevated gene-1/Metadherin (AEG-1/MTDH) plays an important role in regulating lipid metabolism and inflammation in hepatocytes, and promotes metabolic dysfunction-associated steatohepatitis (MASH). Myeloid cells, such as macrophages, play a key role in regulating inflammation. Here we investigated the role of AEG-1 in myeloid cells in regulating high fat/high sugar diet (HF/HSD)-induced MASH. Both male and female AEG-1MAC mice were significantly protected from MASH development compared to AEG-1fl/fl mice.

Changes in intestinal permeability and gut microbiota following diet-induced weight loss in patients with metabolic dysfunction-associated steatohepatitis and liver fibrosis

Project description:We explore whether a low-energy diet intervention for Metabolic dysfunction-associated steatohepatitis (MASH) improves liver disease by means of modulating the gut microbiome. 16 individuals were given a low-energy diet (880 kcal, consisting of bars, soups, and shakes) for 12 weeks, followed by a stepped re-introduction to whole for an additional 12 weeks. Stool samples were obtained at 0, 12, and 24 weeks for microbiome analysis. Fecal microbiome were measured using 16S rRNA gene sequencing. Positive control (Zymo DNA standard D6305) and negative control (PBS extraction) were included in the sequencing. We found that low-energy diet improved MASH disease without lasting alterations to the gut microbiome.

Immuno-laser capture microdissection of perfusion-fixed brain tissue coupled to RNA-seq

Project description:Normal mouse brain was fixed by cardiac perfusion with 4% paraformaldehyde in PBS, then post-fixed by immersion in the same solution for 2hr at RT. Brain was embedded in cryomatrix and frozen sections were cut at 7 um on a cryostat. Sections were immunostained with anti-CD31 antibody and anti-GFAP antibody to detect microvascular endothelial cells and surrounding astrocytes, respectively. Immediately after immunostaining, sections were subject to laser capture microdissection (LCM), using ultraviolet laser to ablate unwanted astrocytes and infrared laser to lift target endothelial tissue. Total RNA was isolated from LCM-retrieved endothelial tissue and processed for RNA-seq.
