Integrated Multi-omics Profiling of Adipose Tissue Response to Leucic Acid in a Mouse Model

ABSTRACT: This project employs mass spectrometry-based proteomics to characterize the tissue-specific molecular mechanisms underlying the anti-obesity effects of leucic acid, a gut microbiota-derived metabolite. The goal is to identify protein networks and pathways modulated by leucic acid across key metabolic organs, with a current focus on the gonadal white adipose tissue (gWAT).

Adipocyte autophagy-mediated signals in the control of macrophage phenotype during diet-induced obesity

Project description:A hallmark of obesity is a pathological expansion of white adipose tissue that is accompanied by an increase in local inflammation and fibrosis. Autophagy is increased during obesity in adipose tissue, however, its role remains incompletely understood. Here, we report that autophagy is a critical regulator of pathological white adipose tissue remodelling and inflammation in diet-induced obese mice. The absence of adipocyte autophagy substantially exacerbates pericellular fibrosis specifically in gonadal white adipose tissue, ameliorating metabolic syndrome. Notably, changes in tissue architecture correlate with increased infiltration of macrophages and autophagy-dependent rewiring of adipocyte metabolism.

MicroRNAs Expressed in Visceral Adipose Tissue from Mice Fed High-Fat Diets Supplemented with Eicosapentaenoic Acid (EPA)

Project description:The goal of this subproject is to identify microRNAs (miRNAs) expressed in visceral (VIS) adipose tissue (gonadal fat from male C57BL/6 mice) and regulated by eicosapentaenoic acid (EPA). This will provide insight into microRNAs regulated by EPA and their potential role in obesity-associated inflammation. We performed small RNA-sequencing of white (VIS) adipose tissue from high-fat diet (45% kcal from fat) supplemented with EPA (45% Kcal from fat, 6.75% EPA). Using the Gunaratne Next-Generation pipeline (published in Creighton et al. 2009), miRNA expression profiles were identified. Counts of each unique read were normalized to total usable reads, and had 40 counts added. We mapped about 13.8 million sequence reads per sample to the Mus musculus genome (build mm 10). We are specifically interested in those miRNAs expressed in VIS fat from EPA-fed mice compared to HF-fed mice.

Single-cell RNA sequencing (sc RNAseq) of white adipose tissue of aged mice

Project description:Aging and obesity are associated with pro-inflammatory changes in adipose tissue. Overlapping mechanisms, such as the infiltration of inflammatory macrophages and T cells into visceral adipose tissue, have been implicated in contributing inflammation. However, a comparative analysis from both states is needed to identify distinct regulatory targets. Here, we performed single-cell RNA sequencing of stromal vascular fractions (SVF) isolated from gonadal white adipose tissue (gWAT) of young mice fed a normal or a high-fat diet, and aged mice fed a normal diet. Our analysis revealed that physiological aging, more so than high-fat diet induced obesity, was associated with accumulation of phenotypically distinct CD8 T cells resembling virtual memory (VM) CD8 T cells.

Single-cell transcriptome of stromal vascular fraction isolated from adipose tissue of lean and obese mice

Project description:Visceral white adipose tissue (VAT) is susceptible to inflammation as a result of obesity, and this inflammation contributes to the development of obesity-related diseases. During the development of obesity, inflammatory immunocytes, including CD8+ T cells, Th1 cells, M1-like macrophages, neutrophils, and mast cells, accumulate in VAT. While these profound changes in the immunocyte composition of VAT lead to local inflammation, the triggers of VAT inflammation remain poorly understood. Therefore, a better understanding of the mechanisms underlying an early event of obesity-associated VAT inflammation will contribute to the development of novel therapies for obesity-related diseases. We utilized CellChat, which employs the expression of ligand-receptor pairs as indicators of intercellular communications, to examine the potential impact of these identified cell types within SVF on T cells. Our analysis revealed that ASCs exert the greatest influence on T cells in both lean and obese conditions. Finally, we demonstrated that ASCs promote Tcell infiltration into WAT during the early stages of obesity.

Comparison of white adipose tissue and bone marrow adipose tissue from New Zealand White rabbits

Project description:Microarray analysis of white adipose tissue (WAT) and bone marrow adipose tissue (BMAT) from 22-week-old or 13-week-old male New Zealand White rabbits. From both cohorts, BMAT was sampled from the distal tibia (dBMAT) and the radius and ulna (ruBMAT), while WAT was sampled from the inguinal (iWAT) and gonadal (gWAT) depots. From the 13-week-old cohort, BMAT was also sampled from the proximal tibia (pBMAT). Sufficient RNA could not be isolated from all tissues for all rabbits, so for some rabbits only a subset of tissues is included.
