Mass spectrometry: the association of LPHN2 with the MET complex

ABSTRACT: To delineate the mechanism by which LPHN2 participates in the MET process, we performed proteomic analyses to investigate the LPHN2 interactome in mouse cochleae. The Flag-tagged LPHN2-GAIN construct with deletion of the 523 N-terminal residues of LPHN2 showed mechanosensitive potential that was comparable to that of full-length LPHN2. We therefore purified the Flag-tagged LPHN2-GAIN protein to use as bait to identify potential candidate proteins that coordinate LPHN2 function in vivo.

Hepatocyte-specific MET deletion exacerbates acetaminophen-induced hepatotoxicity in mice

Project description:Despite the well-known role of MET in liver regeneration following partial-hepatectomy (PHx), its role in the clinically-relevant acetaminophen (APAP)-induced liver injury (AILI) model remains unexplored. AILI markedly differs from PHx because it is associated with massive liver necrosis. This study aims to delineate the role of MET specifically in AILI. Hepatocyte-specific MET-KO mice were given a toxic-dose of APAP and assessed for hepatotoxicity/regeneration parameters. MET deletion strikingly exacerbated initial hepatotoxicity and impaired subsequent proliferative response, culminating in significant mortality. Mechanistically, MET deletion enhanced JNK-activation and its mitochondrial translocation, resulting in excessive mitochondrial oxidative-damage, releasing cell-death inducer AIF into cytosol. Excess JNK-activation was attributed to reduced inhibitory activity of AKT on JNK in the absence of MET-signaling. Pharmacological activation of AKT reduced JNK-activation and hepatotoxicity in MET-KO mice. RNA-sequencing/immunoblotting not only showed repression of proliferative/survival signaling, but also activation of cell death/senescence pathways along with impaired unfolded-protein-response in MET-KO mice. Analysis of published single-nucleus RNA-sequencing data showed proliferation in livers from APAP-induced ALF patients was associated with strong activation of HGF/MET signaling in hepatocytes, with spatial-transcriptomics showing striking induction of HGF surrounding the necrotic-zones. Interestingly, 35% of the genes altered in human-ALF were regulated by MET in the mouse AILI-model. In conclusion, our study demonstrates that MET is crucial for restraining hepatotoxicity following APAP overdose via inhibiting mitochondrial cell-death signaling pathway.

Transcriptome analysis of MET-constitutively active versus MET-silenced colorectal cancer cells

We then silenced MET in LoVo cells using CRISPR/Cas9 knock-out (KO) protocol, and performed gene expression profiling analysis using data obtained from RNA-seq of cells with hyperactivated-MET versus MET-KO.

Molecule Glue That Stabilizes the LRPPRC-MET-G4 Interaction Complex to Drive MET Downregulation

Project description:Targeting the MET oncoprotein is an effective strategy in precision cancer therapy, whereas its clinical efficacy varies dramatically across tumor types. To address this challenge, we explored an alternative approach to downregulate MET expression at transcriptional levels. We identified a cis-regulatory element in the MET proximal promoter region that folds into a stable parallel DNA G-quadruplex (MET-G4). We further determined the high-resolution NMR solution structure of MET-G4 and demonstrated that MET-G4 recruits the cellular LRPPRC protein to promote MET transcription, thereby uncovering a previously unrecognized epigenetic mechanism that drives MET overexpression. Moreover, we characterized the specific G4-binding domain of LRPPRC and elucidated its structural basis for recognition of the MET-G4. Through screening an in-house natural product library, we identified nitidine alkaloid (NIT) as a potent MET-G4 stabilizer. Strikingly, nitidine acts as a molecular glue, strengthening the LRPPRC-MET-G4 interaction and inducing the formation of a stable LRPPRC-NIT-MET-G4 ternary complex. This complex likely alters the structure and function of LRPPRC, ultimately leading to MET downregulation and potent anticancer effects. Notably, this is the first report of a molecular glue that stabilizes a G4-protein complex to silence an oncogene. Additionally, comprehensive in vitro and in vivo experiments demonstrated that nitidine significantly inhibits tumor progression mainly through an LRPPRC-MET-G4-dependent mechanism. Collectively, our study reveals a novel epigenetic regulatory mechanism involving LRPPRC-MET-G4-mediated MET upregulation and suggests a promising therapeutic strategy for MET-driven tumors with molecular glues that target the LRPPRC-MET-G4 interface

Expression data of Met-low and Met-high B16-F10 malignant melanoma cells

Project description:B16-F10 malignant mouse melanoma cells have been frequently used as highly metastatic cells. Based on heterogenous cell surface expression of Met/HGF (hepatocyte growth factor) receptor in B16-F10 cells, the cells were divided into Met-low and Met-high cells by flow cytometry and these populations were subjected to microarray analysis. Met-low and Met-high cells showed different expression profiles in genes involved characteristics of tumors, including stem cell maintenance, pigmentation, and angiogenesis.
