microRNA analysis of supernatants from stromal vascular fraction (SVF) cells derived from healthy controls and lipedema patients

Project description:SVF cells were isolated from liposuction material (hips, outer thigh) of healthy controls and lipedema patients and seeded for the collection of cell supernatants after 24 hours. After several centrifugation steps, the supernatants were concentrated and enriched for small extracellular vesicles. Total RNA was extracted from the concentrated supernatants and the small extracellular vesicles fraction. 187 miRNAs were analyzed using qPCR reaction (miRCURY SYBR® Green mastermix, Quiagen).

Extracellular Vesicles From Liver Progenitor Cells Downregulates Fibroblast Metabolic Activity and Increase the Expression of Immune-Response Related Molecules

Project description:Extracellular vesicles are an important part of cell to cell crosstalk, and help to the acceptor cells to prepare for changes in their environment. MLP29 cells are mouse liver progenitor cells that release EVs loaded with cell fate signaling. In the current work, we have fed 3T3-L1 mouse fibroblast with those EVs, and then analyzed the cellular response by proteomics and transcriptomics. Results showed a downregulation of both proteins and transcripts related to proliferative and metabolic routes dependent on TGFb, and an increase on the ERBB2 inhibitor protein (ERBIN). An increase in the transcription of CXCL2, as well as the increase of ribosome biogenesis and interferon response molecules points toward the activation of immune system signaling, compatible with a response against tissue damage.

Genome-wide expression analysis of the effects of a parasitic nematode's secreted exosomes on mammalian cells.

Project description:We show that a gastrointestinal nematode that infects mice, Heligmosomoides polygyrus, secretes vesicles packaged with specific microRNAs (miRNAs) and Y RNAs as well as a nematode-specific Argonaute protein. The vesicles are of intestinal origin and are enriched for homologs of mammalian proteins found in exosomes, including heat shock proteins, tetraspanins and an ESCORT protein. The nematode-derived vesicles are internalized by mouse intestinal epithelial cells and mediate changes in expression of host genes involved in inflammation and immunity, including the receptor for the alarmin IL33 as well as a key regulator of MAPK signaling, DUSP1. A total of 12 samples were analyzed, from 4 different conditions: medium, exosome-treated, LPS and LPS+exosomes, each with 3 biological replicates. One of the 'medium' replicates was discarded due to outlier behaviour.

Synergistic activity of BET protein antagonist-based combinations in Mantle Cell Lymphoma cells sensitive or resistant to ibrutinib

Project description:To determine the global transcriptome changes in mantle cell lymphoma cells following treatment with the BET bromodomain antagonist, JQ1 Mantle Cell Lymphoma (MCL) cells exhibit increased B cell receptor and NFkB activities. The BET protein BRD4 is essential for the transcriptional activity of NFkB. Here, we demonstrate that treatment with the BET protein bromodomain antagonist (BA) JQ1 attenuates MYC and CDK4/6, inhibits the nuclear RelA levels and the expression of NFÎºB target genes including Brutonâs Tyrosine Kinase (BTK) in MCL cells. While lowering the levels of the anti-apoptotic BCL2 family proteins, BA treatment induces the pro-apoptotic protein BIM and exerts dose-dependent lethality against cultured and primary MCL cells. Co-treatment with BA and the BTK inhibitor ibrutinib synergistically induces apoptosis of MCL cells. Compared to each agent alone, co-treatment with BA and ibrutinib markedly improved the median survival of mice engrafted with the MCL cells. BA treatment also induced apoptosis of the in vitro isolated, ibrutinib-resistant MCL cells which overexpress CDK6, BCL2, Bcl-xL, XIAP and AKT, but lack ibrutinib resistance-conferring BTK mutation. Co-treatment with BA and panobinostat (pan-histone deacetylase inhibitor) or palbociclib (CDK4/6 inhibitor) or ABT-199 (BCL2 antagonist) synergistically induced apoptosis of the ibrutinib-resistant MCL cells. These findings highlight and support further in vivo evaluation of the efficacy of the BA-based combinations with these agents against MCL, including ibrutinib-resistant MCL. MO2058 cells treated with vehicle, 250 nM or 1000 nM JQ1 for 8 hours. Samples were acquired and analyzed in duplicate.
