Project description:To search for the functional “m6A readers” specifically in the context of Drosophila early embryos, we incubated m6A-modified or unmodified RNA probes with cytosolic lysates from early embryos, and performed immunoprecipitation experiments followed by mass spectrometry analysis. We found that Drosophila FMRP (FMR1) was highly abundant in complexes immuno-precipitated by the m6A-modified probe. We performed data-independent acquisition (DIA) quantitative proteomics on the wild type and fmr1 maternal mutant embryos at multiple developmental stages, including 0-1 h, 2-3 h, and 5-6 h stages. To screen the FMR1-associated partners, we collected the 2–3-hour embryos and performed immunoprecipitation using the anti-Drosophila FMR1 antibody, followed by mass spectrometric analysis.

Project description:Environmental factors, including diet, can influence gene expression and developmental outcomes through epigenetic modifications. While maternal diet is known to affect offspring DNA methylation and phenotypes, its impact on the oocyte and subsequent embryo epigenome remains less understood. Here, we investigated how maternal methionine supplementation affects DNA methylation patterns in oocytes and embryos of Polypay ewes. Whole genome bisulfite sequencing (WGBS) was performed on oocytes collected from 16 twin ewe pairs (8 methionine-treated and 8 control). These ewes were later bred to control rams, and embryos were flushed for WGBS as well. In oocytes, 2,056 differentially methylated cytosines (DMCs) were identified, with 2,034 hypomethylated and 22 hypermethylated. Additionally, 17 mitochondrial DMCs were identified, with 12 hypermethylated and 5 hypomethylated. In embryos, 113 DMCs were identified, with 112 hypermethylated and only one hypomethylated. Mitochondrial DNA analysis revealed 22 hypermethylated DMCs. To assess the inheritance of methyl marks, we compared DMCs between oocytes and embryos. While no direct overlaps were found in nuclear DNA, three CpGs exhibited opposite methylation trends—hypomethylated in oocytes but hypermethylated in embryos. In contrast, five mitochondrial DMCs overlapped between oocytes and embryos. To functionally assess the role of differentially methylated genes, we performed siRNA-mediated knockdown of two embryo DMC-associated genes: SCRIB and CERS3. Knockdown of SCRIB led to a 16.4% average decrease in blastocyst formation rate (p = 0.001), while CERS3 knockdown resulted in a 9.5% decrease (p = 0.005). These results demonstrate that maternal methionine supplementation alters both nuclear and mitochondrial DNA methylation in oocytes and embryos and that affected genes may play critical roles in early embryonic

Project description:The subcortical maternal complex (SCMC) is essential for the maintenance of female fertility in mammals. Most pathogenic mutations in SCMC-encoding genes result in female reproductive failure characterized by early embryonic arrest. Assembled in oocytes, the SCMC maintains the normal cleavage of early embryos, but the mechanism regulating this function remains unclear. Here, we report that 14-3-3, a multifunctional protein, is a component of the SCMC. By resolving the cryoelectron microscopy structure of the 14-3-3-containing SCMC, we revealed that specific phosphorylation sites in TLE6 contribute to the recruitment of 14-3-3, which is indispensable for the role of the SCMC in maintaining the cleavage of mouse early embryos. Mechanistically, during the maternal-to-embryo transition, the SCMC stabilizes the 14-3-3 protein, and an acute reduction in 14-3-3 protein expression in zygotes impairs their further development to blastocysts. The SCMC containing 14-3-3 contributes to the proper control of CDC25B, thus ensuring the activation of the maturation-promoting factor and mitotic entry in mouse zygotes. Notably, we proved that the recruitment of 14-3-3 to the SCMC and the molecular link between the SCMC and CDC25B are conserved in human oocytes/embryos. This study reveals the molecular mechanism through which the SCMC regulates the cell cycle in early embryos and represents a breakthrough in fully elucidating the functions of the SCMC in mammalian early embryogenesis.

Project description:Basonuclin, which is a zinc-finger protein found in abundance only in the keratinocytes of the stratified epithelium, male germ cells and oocytes, qualifies as a maternal-effect gene because the source of pre-implantation embryonic basonuclin is maternal. Using a transgenic-RNAi approach, we knocked-down basonuclin specifically in mouse oocytes, which led to female sub-fertility. Basonuclin deficiency in oocytes perturbed both RNA polymerase I- and II-mediated transcription and oocyte morphology was affected as evidenced by cytoplasmic and cell surface abnormalities. The affected oocytes, however, could still mature to and arrest at metaphase II and be ovulated, suggesting the impaired pathways were not essential for oocyte development and maturation. Nevertheless, an early embryonic failure in pre-implantation development was identified and likely accounted for the sub-fertility phenotype. These results suggest that basonuclin is a new member of the mammalian maternal-effect genes and interestingly, differs from the previously reported mammalian maternal-effect genes in that it also apparently perturbs oogenesis. Using detailed single cell cDNA microarray analysis of normal versus mutant oocytes, we found that a large proportion of maternal genes, including C-mos, are under the control of miRNAs. This study provides strong evidence that maternal miRNAs are essential for the earliest stages of mouse embryonic development. mRNA expression was compared with miRNA expression in dicer knockout and wild-type mouse mature oocyte.

Project description:We report the application of low input high-throughput profiling of histone modifications in mouse oocytes and 2-cell embryos. We used antibodies against H3K36me3 (190 wildtype and 93 knockout MIIcoocytes pooled as one replicate) and H3K9me3 ( around 118 control and MZ embryos / replicate; 1 biological replicates per genotype). In the case of H3K36me3, we find that its establishment and maintence is not affected in oocytes lacking Kdm4a. In the case of H3K9me3 in 2-cell embryos, we find that the histone mark aberrantly gained in Kdm4a deficient oocytes is also maintained in the MZ mutant 2-cell embryo. This shows that Kdm4a is important to keep open chromatin clear of H3K9me3 spreading in oocytes in order to transfer a healthy epigenome amenable for proper genome activation post fertilization.
