Different expressed proteins between the social transmission of depression sensitive and insensitive proteins in the medial prefrontal cortex

ABSTRACT: A social transmission of depression (STD) score was calculated based on all 6 different behavioral tests, including open field test, elevated plus maze, tail suspension test, forced swimming test, sucrose preference test, novelty-suppressed feeding test. social interaction ratio, sucrose preference, and forced swim immobility. All behavioral tests were performed during the light cycle. Mice were anesthetized by isoflurane and brains were removed. mPFC tissue was quickly hand dissected and frozen with liquid nitrogen. Three pairs of mPFC samples from STDs mice and STDi mice were used for proteomic analysis.

Project description:Experimental evidence suggests that environmental stress conditions can alter the expression of BDNF and the levels of this neurotrophin influence the behavioural response in mammalian models. Recently it has been demonstrated that the exposure to 34° C for 21 days alters the brain proteome and behaviour in zebrafish. The aim of this work was to investigate the role of BDNF in the nervous system of adult zebrafish under control and heat treatment conditions. For this purpose, wild type (WT), heterozygous BDNF+/- (HT) and knock out BDNF-/- (KO) zebrafish were kept for 21 days at 26° C or 34° C and then euthanized for brain analysis or subjected to behavioural tests (Y-maze test, novel tank test, light and dark test, social preference test, mirror biting test) used for assessing behavioural aspects such as boldness, anxiety, social preference, aggressivity, interest for the novel environment and exploration. RTqPCR analysis showed the reduction of gene expression of BDNF and its receptors after heat treatment. Moreover, proteomic analysis and behavioural tests showed genotype- and temperature-dependent effects on brain proteome and behaviour of zebrafish.

Project description:We performed high-throughput profiling of gene expression in aPVT-projectors in the mPFC of DBA2/J mice subjected to subchronic and mild social defeat stress. DBA mice were subjected to 5-day of social defeat stress and then tested their social interaction and anhedonic state. After behavioral tests, mice were divided into five subtypes: SA, social interaction deficit, ANH: anhedonia, SA:ANH, both social dificit and anhedonia, RES, resilience and non-stressed (NS) control. Mice were euthanized for tissue collection. We then conducted the Immunoprecipitation and RNA-seq. Our study provided insights into the understanding of the molecular mechanisms underlying behavioral heterogeneity.

Effects of Platycodins Folium on Depression in Mice Based on a UPLC-Q/TOF-MS Serum Assay and Hippocampus Metabolomics

Project description:Major depressive disorder (MDD), also known as depression, is a state characterized by low mood and aversion to activity. Platycodins Folium (PF) is the dried leaf of Platycodon grandiflorum, with anti-inflammatory and antioxidative activities. Our previous research suggested that PF was rich in flavonoids, phenols, organic acids, triterpenoid saponins, coumarins and terpenoids. This study aimed to investigate the antidepressant effect of PF using lipopolysaccharide (LPS)-induced depressive mice. Several behavior tests (sucrose preference test (SPT), forced swimming test (FST) and tail suspension test (TST)) and biochemical parameters (IL-6, TNF-α and SOD levels) were used to evaluate the antidepressive effect of PF on LPS-induced depression model. Furthermore, a UPLC-Q/TOF-MS-based metabolomics approach was applied to explore the latent mechanism of PF in attenuating depression. As a result, a total of 21 and 11 metabolites that potentially contribute to MDD progress and PF treatment were identified in serum and hippocampus, respectively. The analysis of metabolic pathways revealed that lipid metabolism, amino acid metabolism, energy metabolism, arachidonic acid metabolism, glutathione metabolism and inositol phosphate metabolism were disturbed in a model of mice undergoing MDD and PF treatment. These results help us to understand the pathogenesis of depression in depth, and to discover targets for clinical diagnosis and treatment. They also provide the possibility of developing PF into an anti-depressantive agent.

After behavioral tests, mice were euthanized for tissue collection. We then conducted the transcriptomics analysis. Our study provided insights into the understanding of the molecular mechanisms underlying stress susceptibility and provide the role of KDM5C in stress-induced behavioral changes.

Molecular mechanism for stress-induced depression assessed by sequencing miRNA and mRNA in medial prefrontal cortex (miRNA)

Project description:Major depressive disorder is a common mood disorder. Chronic stressful life is presumably main etiology that leads to the neuron and synapse atrophies in the limbic system. However, the intermediate molecules from stress to neural atrophy remain elusive. Mice were treated by chronic unpredictable mild stress (CUMS) until demonstrating depression-like behaviors confirmed by the tests of sucrose preference, forced swimming and Y-maze. The sequencings of microRNA and mRNA from the medial prefrontal cortices were performed in CUMS-induced depression mice versus control mice to assess the molecular profiles of major depressive disorder. In the medial prefrontal cortices of depression-like mice, the levels of mRNAs that translated the proteins for the GABAergic synapses, dopaminergic synapses, myelination, synaptic vesicle cycle and neuronal growth were downregulated. miRNAs of regulating these mRNAs are upregulated. The deterioration of GABAergic and dopaminergic synapses as well as axonal growth is associated to CUMS-induced depression.

wdr13 knockout mice develops MD like phenotype on chronic stress

Project description:WDR13 expresses from the X chromosome and has a highly conserved coding sequence. There have been multiple associations of WDR13 with memory and psychiatric disorders. However, its detailed function in context of brain and behavior remains unknown. We characterized the behavioral phenotype of male mice lacking the homologue of WDR13 gene (Wdr13-/0). Mice were evaluated for changes in anxiety, learning and memory, and chronic stress induced behavioral deficits. Taking cue from analysis of its expression in the brain, we chose hippocampus and pre-frontal cortex for molecular studies to delineate its function. In the absence of Wdr13, there was a significant upregulation of synaptic proteins, viz., SYN1, RAB3A, CAMK2A etc accompanied with better memory retention in mice. Interestingly, three weeks social isolation stress resulted in downregulation of aforementioned synaptic proteins, decreased exploration time in central area of Open field test, increased immobility in Forced swim test, and reduced dendritic arborations of hippocampal CA1 neurons- hallmarks of Major Depressive Disorder (MDD) in Wdr13-/0 mice. This phenotype could be rescued by short imipramine treatment. We hypothesise that one of the key factors behind the MDD like phenotype in these mice was upregulation of transcription factor GATA1. Parallel work from our lab has indicated WDR13 to interact with multiple nuclear receptors and acting majorly as a co-repressor. Absence of Wdr13 resulted in de-regulated expression of a number of genes including multiple synaptic genes leading to observed phenotypes. Summarily, our data provides functional evidence for the role of Wdr13 in memory retention and in withstanding chronic stress induced depression.

Individual variation in preference behavior in sailfin fish refines the neurotranscriptomic pathway for mate preference

Project description:Social interactions can drive distinct gene expression profiles which may vary by social context. Here we use female sailfin molly fish (Poecilia latipinna) to identify genomic profiles associated with preference behavior in distinct social contexts: male-interactions (mate choice) versus female-interactions (shoaling partner preference). We measured behavior of 15 females interacting in a non-contact environment with either two males or two females for 30 minutes followed by whole brain transcriptomic profiling by RNA sequencing. We profiled females that exhibited high levels of social affiliation and great variation in preference behavior to identify an order of magnitude more differentially expressed genes associated with behavioral variation than by differences in social context. Using linear modeling (limma), we took advantage of the individual variation in preference behavior to identify unique gene sets that exhibited distinct correlational patterns of expression with preference behavior in each social context. By combining limma and weighted gene co-expression network analyses (WGCNA) approaches we identify a refined set of 401 genes robustly associated with mate preference that is independent of shoaling partner preference or general social affiliation. While our refined gene set confirmed neural plasticity pathways involved in moderating female preference behavior, we also identified a significant proportion of discovered that our preference-associated genes were enriched for ‘immune system’ gene ontology categories. We hypothesize that the association between mate preference and transcriptomic immune function is driven by the less well-known role of these genes in neural plasticity which is likely involved in higher-order learning and processing during mate choice decisions.

GPR158 in pyramidal neurons mediates social novelty behavior via modulating synaptic transmission in male mice

Project description:Impairment in social communication skills is a hallmark feature of autism spectrum disorder (ASD). The role of G-protein coupled receptor 158 (GPR158) in ASD remains largely unexplored. In this study, we observed both constitutive and cell/tissue-specific knockouts of Gpr158 in pyramidal neurons or medial prefrontal cortex (mPFC) result in impaired novelty preference, while sociability remains unaffected in male mice. Notably, loss of GPR158 leads to a significant decline in excitatory synaptic transmission, characterized by the reduction in glutamate vesicles, as well as the expression and phosphorylation of GluN2B in the mPFC. We successfully rescue the phenotype of social novelty deficits either by reintroducing GPR158 in the mPFC of Gpr158 deficient mice or by chemogenetic activation of pyramidal neurons where Gpr158 is specifically ablated. Our findings indicate that GPR158 in pyramidal neurons plays a specific role in modulating social novelty, and may represent a potential target for treating social disorder.
