Proteomic study of extracellular vesicles from kefir-derived lactic acid bacteria

ABSTRACT: This project analyzed the proteomics of extracellular vesicles of kefir derived lactic acid bacteria. Samples were analyzed on timsTOF Pro (Bruker Daltonics), a hybrid trapped ion mobility spectrometer (TIMS) quadrupole time-of-flight mass spectrometer. The functional characteristics of extracellular vesicles of kefir derived lactic acid bacteria were obtained through bioinformatics analysis

However, the effect of lactic acid on Treg induction is not known. We therfore evaluated the effect of lactic acid on Treg induction and observed an increased induction of Tregs in the presence of lactic acid. This increase occurred in a glycolysis-independent, acidity-dependent manner.

Project description:Proteins were precipitated using chloroform and methanol, by the protocol described by Wessel and Flügge (1984). Following precipitation, the proteins were reduced and alkylated before being digested, cleaned and desalted using STrap tips (Zougman A, Selby PJ, Banks RE. 2014). The peptide samples were analyzed by a nano UPLC (nanoElute, Bruker) coupled to a trapped ion mobility spectrometry and a quadrupole time of flight mass spectrometer (timsTOF Pro, Bruker).
