Prognosis prediction model for Alzheimer’s disease conversion from mild cognitive impairment by integrative analysis of multi-omics data

Project description:Mild cognitive impairment (MCI) is a clinical precursor of Alzheimer’s disease (AD). Many MCI subjects convert to AD, although others remain stable as MCI or sometimes return to cognitive normal. Currently, there are no curative treatments for patients who are already in AD, and therefore biomarkers for early detection of high risk MCI-to-AD conversion subjects are rapidly required. Here, we investigated potential biomarkers from blood-based miRNA (miR) expression profiles and genomic data of 197 Japanese MCI patients. Using the candidate biomarkers, we constructed a prognosis prediction model based on a Cox proportional hazard model. The final prediction model, composed of 24 miR-eQTLs (i.e. SNP-miRNA pairs) and three clinical factors (age, sex and ApoE ε4 carriers), successfully classified MCI patients into two groups, low and high, in terms of risk of MCI-to-AD conversion (log-rank trend test P=3.44×10^(-4)), and achieved a concordance index of 0.702 on an independent test set. Network-based meta-analysis using target genes of the miR-eQTLs further revealed four important hub genes (SHC1, FOXO1, GSK3B, and PTEN) associated with the pathogenesis of AD. Statistically significant differences were observed in PTEN expression between MCI and AD and SHC1 expression between cognitively normal elder subjects (CN) and AD when examining RNA-seq data from 610 blood samples (PTEN, P=0.023; SHC1, P=0.049), although FOXO1 and GSK3B showed low levels of expression in blood. Accurate prediction of MCI-to-AD conversion enables earlier appropriate intervention for those MCI patients and can lead to a reduction of MCI patients that convert to AD with high risk. We believe that further investigation with larger sample sizes will contribute to practical clinical use of our approach in MCI-to-AD conversion in the near future.

Transcriptomic profiling of mouse subgranular zone-derived neural stem cells exposed to cadmium

Project description:We performed RNA-seq to investigate the effects of cadmium (Cd) exposure on the transcriptome of primary mouse neural stem cells (NSCs) isolated from the subgranular zone (SGZ) of the hippocampus. NSCs at passage 3 were exposed to 0.8 μM CdCl₂ for 72 hours, and total RNA was extracted for sequencing on an Illumina platform. Differential expression analysis identified 684 differentially expressed genes (334 downregulated, 350 upregulated). Gene Ontology enrichment analysis of downregulated genes revealed impairment of neurogenesis-related processes, and WikiPathways analysis indicated enrichment of Wnt signaling pathway components. These transcriptomic data support the finding that Cd disrupts non-canonical Wnt5a signaling and impairs neuronal differentiation of SGZ-derived NSCs.

Project description:A mixed-aerosol pH1N1 (Cal04) challenge of rhesus macaques was establised to serve as a pre-clinical model for the evaluation of candidate vaccines. After characterizing the clinical signs and immune responses associated with pH1N1 challenge in naïve rhesus macaques, a follow-up study assessing 2 candidate vaccines was performed. This study has 2 phases: 1) Model Establisment consisting of 3 groups: Unvaccinated Live Challenge (n=3, Unvaccinated UV-inactivated Challenge (n=3), Previously Vaccinated Live Challenge (n=3) which were sampled at 2 baseline timepoints Day -7 and Day 0. Following the H1N1 Challenge, samples were collected at day 1,2,5,8,14,20. 2) Candidate Vaccine Assessment consisting of four groups: Previously Vaccinted with anti-CD40-NP5+PolyICLC (n=4), Previously vaccinated with CD40-HA+PolyICLC (n=4), Previously vaccinated with commercial mismatched Fluzone (n=4), Previously vaccinated with Media+PolyICLC alone (n=4). Daseline samples were collected at Day -7 and 0 (baseline) and Day 1,3,6,14,20 post-challenge.

Long-Term Benefits of TUDCA Supplement in ARSACS Zebrafish Model

Project description:Our work demonstrated an impairment in cognitive, social and emotional behavior in aged sacs knockout zebrafish similar to that observed in human patients. Transcriptomic and proteomic data from the adult brains revealing alterations in several genes related to circadian rhythms and neuroinflammation in adult mutants. t Interestingly, alterations in sleep and circadian rhythms are common in individuals with cerebellar ataxia and may contribute to cognitive function and behavior. Long term treatment with TUDCA , a neuroprotective molecule,partially rescued social and cognitive deficits. These findings suggest the potential for further optimization of this neuroprotective compound in subsequent preclinical and clinical studies.

Assessment and Prediction of Cetuximab-Induced Hypersensitivity Reactions Using Cetuximab Specific IgE Detection

Project description:1. Background Cetuximab (trade name Erbitux) is a murine-human chimeric monoclonal antibody to human epidermal growth factor receptor (EGFR). This drug has been used as a treatment for colorectal cancer and head and neck cancer. It is known that allergic reactions can occur in more than 5% of the patients, although the side effects are relatively low compared with other chemotherapeutic agents. It is known that cetuximab can induce hypersensitivity even at the first administration, unlike other anticancer drugs. In this study, we aimed to establish a model to predict patients with hypersensitivity reaction before administration of cetuximab and to provide safe chemotherapy. 2. Recruitment method and consent procedure The study is designed for analysis patients scheduled for administration of cetuximab for the first time. Patients matching the selection and exclusion criteria with voluntary agreement to the study will be enrolled. Enrolled patients will be tested for skin prick test and serum sIgE before cetuximab administration.
