Cryo-EM Structures of Biopsy-derived TTR Filaments in Hereditary Transthyretin Amyloidosis.

ABSTRACT: We examined the structure of transthyretin filaments extracted from the vitreous humor of an individual with ATTRv-F64S by cryo-electron microscopy. Mass spectrometry was used to confirmed the presence of both mutant and wild-type transthyretin in the filaments.

Cryo-EM Structures of Prion protein filaments from Gerstmann-Sträussler-Scheinker disease

Project description:Aggregation of the prion protein (PrP) and formation of PrP amyloid (APrP) is severe in some Prion diseases. In the dominantly inherited prion protein amyloidosis known as Gerstmann-Sträussler-Scheinker (GSS) disease, plaques made of PrP amyloid are a distinct feature. The TTC to TCC mutation in Prion protein gene (PRNP), resulting in a phenylalanine to serine amino acid substitution at PrP residue 198, causes a severe amyloidosis in a well-studied GSS variant. The neuropathologic phenotype of this neurodegenerative disease is characterized by the presence of numerous extracellular APrP plaques and intracytoplasmic tau neuronal inclusions which are identical to neurofibrillary tangles of Alzheimer disease. Using cryogenic electron microscopy (cryo-EM), we determined for the first time the structures of filaments of human APrP, isolated post-mortem from the brain of a symptomatic PRNP F198S mutation carrier. We report that in GSS (F198S) APrP filaments are composed of dimeric, trimeric and tetrameric left-handed protofilaments with their protomers sharing a common protein fold. The protomers in the cross-β spines consist of sixty-two amino acids and span from glycine 80 to phenylalanine 141, adopting a previously unseen spiral fold with a thicker outer layer and a thinner inner layer. Each protomer comprises nine short β-strands. The β1 and β8 strands as well as the β4 and β9 strands are engaged in a steric zipper. The new data, obtained by Cryo-EM, provide insights into the complexity of the structure of pathogenic PrP, and reveal the difference of PrP’s structure in brain disease versus that of recombinant PrP, highlighting the urgency of extending our knowledge of the structure of the amyloid filaments involved in human neurodegenerative diseases.

The molecular basis of lamin- chromatin interactions [SAMMY-Seq]

Project description:Lamins are the main constituents of the nuclear lamina, a fibrillar layer underlying the inner nuclear membrane. The nuclear lamina governs chromatinorganization through lamina-associated domainswithin the densely packed heterochromatin regions. Employing cryo-focused ion beam (cryo-FIB) milling in conjunction withcryo-electron tomography (cryo-ET), we mappedthe concentration of nucleosomesat the lamin-chromatin interface. Depletion of lamin A/C induces nanometer-scale chromatin decompaction, macroscopic-scale alterations in gene expression and chromatin-wide alterations of chromatin properties, revealed by 4f-genome-wide analysis. Cryo-electron microscopy (cryo-EM) structural analysis revealed a specific interaction between nucleosomes and the tail domain of lamin A. A unique motif of lamin A that distinguishes it from other lamin isoforms. These findings illuminatethe dynamic interplay between specific lamins and chromatin, shaping chromatin architecture and epigenetic regulation.

Structural basis of nucleosomal H4K20 recognition and methylation by SUV420H1 methyltransferase

Project description:Here we report the cryo-electron microscopy (cryo-EM) structures of SUV420H1 associated with canonical nucleosome core particles (NCPs) or H2AZ containing NCPs. We find that SUV420H1 shows conformational change after bound to a nucleosome and make extensive site-specific contacts with histone and DNA regions, thus enabling H4K20 insertion for catalysis specifically. Through in vivo functional analysis, we validated special residues of SUV420H1 that are critical for its catalytic activity and function in chromatin state regulation.Thus, our study provides molecular insights into the nucleosome-based recognition and histone- methylation mechanisms of SUV420H1.
