Project description:Late blight caused by Phytophthora infestans seriously threatens world’s potato production. Understanding the responsive mechanism of potato to P. infestans infection and exploring key regulators would be advantageous for improving the late blight resistance. Here, we assess the late blight resistance of various potato cultivars from Liangshan Yi Autonomous Prefecture and identified Chuan Liang Shu 10 (CLS) with strong resistant, while Bu Wu Yu (BWY) is highly susceptible. Comparative transcriptomics between these two cultivars upon P. infestans inoculation reveal that CLS activates defense pathways rapidly, while BWY prioritizes energy towards growth and development pathways, resulting in distinct late blight resistances. Specifically, the resistance of CLS might be contributed by enhanced oxidation-reduction process and SA signaling. Clustering analysis of the expression patterns for transcription factors (TFs) demonstrates that the WRKY family members play important roles in late blight resistance of CLS, among which StWRKY26 is identified as a positive regulator of late blight resistance. Silencing of StWRKY26 in CLS significantly compromise its intrinsic resistance to P. infestans. Our study enriches the knowledge of transcriptional responses of potato under P. infestans infection and provide candidate genes for breeding potato cultivars with strong late blight resistance.

Project description:Potato Late blight is one the most important crop diseases worldwide. Even though potato has been studied for many years, the potato disease late blight still has a huge negative effect on the potato production. A total of three commercially available field potato cultivars of different resistance to late blight infection: Kuras (moderate), Sarpo Mira (highly resistant) and Bintje (very suseptable) under controlled green house growing conditions innoculated with a diversity of P. infestans populations. We used label-free quantitative proteomics to investigate the infection with P. infestans in a time-course study over 258 hours. Several key issues limits proteome analysis of potato leaf tissue4–6. Firstly, the immense complexity of the plant proteome which is further complicated by the presence of highly abundant proteins, such as ribulose bisphosphate carboxylase/oxygenase (RuBisCO). Secondly, plant leaf and potato in particular contain abundant levels amounts of phenols and polyphenols which hinder or, unless precautions are taken, completely prevent a successful protein extraction.

Project description:<p><strong>INTRODUCTION:</strong> Grafting is a traditional research and plant production technique used to study the long-distance movement of molecules and reduce disease susceptibility, respectively. Tomato/potato grafts are rare examples of successful interspecies grafting, even resulting in commercially available products. Nevertheless, information on the effect of tomato on the quality parameters of potato tubers is scarce.</p><p><strong>MATERIALS AND METHODS:</strong> The tomato cultivar ‘Mobil’ was grafted with the potato cultivars ‘White Lady’, ‘Hópehely’ and ‘Désirée’, and the phenotype, metabolite composition, and starch and protein contents of tubers were analysed. Anthocyanins were isolated from the tuber skins, and the expression level of the transcription factor ANTHOCYANIN1 (StAN1) was evaluated.</p><p><strong>RESULTS:</strong> Out of the 112 identified metabolites, the concentrations of twelve compounds were altered in the same direction in all three cultivars. The starch content of tubers was increased, while the protein level remained unaltered in ‘White Lady’ and ‘Hópehely’. The oval tubers became roundish. The tomato scion increased the anthocyanin content of ‘Hópehely’ and ‘Désirée’ tuber skins, which was correlated with the upregulation of StAN1 expression.</p><p><strong>CONCLUSION:</strong> Tomato has a significant impact on the quality parameters of potato tubers and reduces the asparagine concentration and risk of acrylamide formation.</p>

Project description:<p><strong>INTRODUCTION: </strong>Earliness of tuberisation and the quality of potato tubers are important traits in potato breeding. The qualitative traits rely on the metabolite profile of tubers, which are storage organs and net importers of assimilates. Thus, the quality of tubers largely depends on the metabolites transported from leaves to developing tubers.</p><p><strong>OBJECTIVES: </strong>To test the influence of canopy on the quality of tubers by metabolite profiling of tubers of an early- and a late-maturing potato line and their grafts.</p><p><strong>METHODS:</strong> Potatoes were grown under greenhouse conditions, grafted and the tubers harvested at the end of the scions' vegetation period. Metabolite profiling of freshly harvested tubers was performed using gas chromatography coupled with mass spectrometry. Statistical analyses were applied to determine the significant differences between the different tubers.</p><p><strong>RESULTS:</strong> 99 metabolites were identified and an additional 181 peaks detected in chromatograms, out of which 186 were polar and 94 non-polar compounds. The concentrations of 113 metabolites were significantly different in the tubers from the early-maturing CE3130 and the late-maturing CE3027 line. Hetero-grafting resulted in considerable changes in the metabolite content of tubers. Especially, the effect of CE3027 on the metabolite composition of tubers formed on CE3130 rootstocks was readily apparent. Nevertheless, many compounds were present at similar levels in the tubers of hetero-grafted plants as was found in the tubers of their scion counterparts.</p><p><strong>CONCLUSION:</strong> Hetero-grafting resulted in many compounds at similar concentrations in rootstock tubers as in scion tubers suggesting that these are transported from the source leaves to tubers.</p>

Project description:Posttranscriptional regulator RsmA is part of a regulatory system that controls production of plant cell wall degrading enzymes (PCWDEs), motility and other virulence factors among the Pectobacterium spp of plant pathogens. We constructed the knockout mutation of rsmA gene in Pectobacterium wasabiae strain SCC3193. The rsmA- strain has three times lower growth rate but increased virulence when compared to wild type. To explain this phenotype we compared wild type and rsmA- mRNA levels using genome wide microarray. We found that the synthesis of all known virulence factors and flagella is upregulated, while cell division and peptidoglycan synthesis is down regulated in rsmA- strain. The TCA cycle is directed fully to energy production, indicating low energetic status of the cell that is a probable reason for the low growth rate of the mutant. Growth experiments showed that the enzymatic activities of extracellular virulence factors polygalacturonase and pectate lyase are very high throughout the growth curve even without induction by plant component. Wild type cells are rod shaped in liquid media, but elongated and hyperflagellated in swarm plate containing potato tuber extract and in potato tuber. The rsmA- strain exhibits the same conditional phenotype, but differentiation into swarmer cells is faster and cell elongation more pronounced. Virulence experiments in potato tuber showed that although rsmA- strain causes bigger tissue maceration, its ability to compete with the natural bacterial community in the host is decreased. We conclude that the lack of RsmA switches the cells to infective phenotype characterized by expression of virulence factors and swarming. The lack of control over these energy costly processes is probably the reason for decreased growth rate and fitness.
