Proteomic identification of Amuc_1100-interacting proteins by GST pull-down LC–MS/MS

ABSTRACT: To identify potential interacting proteins of the outer membrane protein Amuc_1100 from Akkermansia muciniphila, GST pull-down assays were performed using lysates from HTR-8/SVneo cells, mouse placenta, and small intestine tissues. The bound proteins were analyzed by LC–MS/MS on an Orbitrap Exploris 480 mass spectrometer

Project description:We propose here a workflow based on an automated immunocapture followed by top-down and middle-down LC-MS/MS approaches to characterize mAb proteoforms spiked in mouse plasma. We demonstrate the applicability of our workflow on a large concentration range using pembrolizumab as a model. We also compare the performance of two state-of-the-art Orbitrap platforms (Tribrid Eclipse and Exploris 480) for these studies. The added value of our workflow for an accurate and sensitive characterization of mAb proteoforms in mouse plasma is highlighted.

Project description:GST-TbCSBβC1 and GST proteins were expressed in Escherichia coli cells, and then incubated with total N.benthamiana proteins. The protein band that was specifically pulled-down by GST-TbCSB βC1 is indicated with a red arrow. GST is around 26 kDa and GST-IbCSB βC1 is around 40 kDa.

Project description:For the GST pull-down of DRG ganglion, the GST- or Tmc7-GST-associated beads were further incubated with cell lysates derived from DRG ganglion. The pulled-down proteins were then separated by SDS-PAGE gel electrophoresis and subsequently stained with silver staining Kit. Finally, the target molecular weight bands and undenatured beads were analyzed by proteomic analysis, respectively.

Project description:The Chikungunya virus (CHIKV) is a single positive-stranded RNA virus transmitted by mosquitoes. More than 40 countries have reported cases of CHIKV infection to varying degrees. Currently, there is no licensed specific antiviral drug or vaccine for CHIKV in the world. In this study, we have identified host protein interactors of CHIKV nsP4 proteins in 293T cells and the major cellular pathways involved in the interaction between host factors and CHIKV nsp4 using Co-Immunoprecipitation and GST Pull-down followed by high-throughput quantitative mass spectrometry.

The differentially expressed proteins were screened in this quantitative proteomics.
