Project description:We have developed an oligoGEArray with 434 genes from Ae. aegypti salivary gland. OligoGEArray was customized with the salivary gland genes chosen from the transcriptome source published by Ribeiro et al (2007). Oligonucleotides (60bp) were designed and array were manufactured by SABiosciences. Using this OligoGEArray, we analyzed the differential expression of salivary trancritptome upon blood feeding. Salivary glands were dissected from 1, 3, 24 and 48 hours post fed (hpf) and unfed Ae. aegypti. Total RNA were extracted and the differential expression of transcriptome were analysed. Quantitative Real-Time PCR was performed on selected genes to validate the OligoGEArray data.

Project description:Mosquito saliva facilitates blood feeding through the anti-haemostatic, anti-inflammatory and immunomodulatory properties of its proteins. However, the potential contribution of non-coding RNAs to host manipulation is still poorly understood. We analysed small RNAs from Aedes aegypti saliva and salivary glands and show here that chikungunya virus-infection triggers both the siRNA and piRNA antiviral pathways with limited effects on miRNA expression profiles. Saliva appears enriched in specific miRNA subsets and its miRNA content is well conserved among mosquitoes and ticks, clearly pointing to a non-random sorting and occurrence. Finally, we provide evidence that miRNAs from Ae. aegypti saliva may target human immune and inflammatory pathways, as indicated by prediction analysis and searching for experimentally validated targets of identical human miRNAs. Overall, we believe these observations convincingly support a scenario where both proteins and miRNAs from mosquito saliva are injected into vertebrates during blood feeding and contribute to the complex vector-host-pathogen interactions.

Project description:A differential display experiment was done to compare genes expressed in salivary glands from unfed males, males fed in the presence of females and males fed in the absence of females. Possible differential bands were excised, cloned and sequenced. A macroarray study was done on these clones as probes. RNA from the salivary glands of ticks from the three conditions was labelled and hybridized. Two sets of primers were used in the differential display, and these same two were used in the labelling, so there are two platforms, corresponding to what primer pair was used. Two independent RNA isolations were done, Experiment 1 used one, 2 and 3 the other. Experiment 2 used exposure to X-ray film, while 1 and 3 used a phosphorimager plate. Two exposures of the X-ray film were found to give acceptable results, 1 and 5 min. These were averaged in the analysis. Project description:Aedes aegypti is a major vector for dengue, chikungunya and yellow fever. Though salivary gland plays a crucial role in transmission of virus and vector life cycle, there is no global and unbiased published report on salivary gland proteome of Ae. aegypti. In this study, we have carried out mass spectrometry based proteomic analysis of salivary gland from adult female Ae. aegypti mosquitoes. By fractionating the proteins isolated from salivary gland on SDS-PAGE and analyzing the in-gel digested bands on high resolution mass spectrometry, we identified 1,205 proteins. This is by far the largest catalogue of salivary gland proteome of Ae. aegypti. These proteins were then assigned molecular functions and biological processes. Several immunity related pathways were found to be enriched in salivary gland. In addition, subset of proteins was predicted to secretory in nature and may play an important role during blood feeding. This study provides a useful resource of proteins expressed in salivary gland of Ae. aegypti female mosquitoes and will aid in biomedical research focused on development of transmission blocking vaccine.

Results: We have performed a global transcriptome analysis of the A. gambiae salivary gland response to blood-feeding, to identify candidate genes that are involved in hematophagy. A total of 4,978 genes were found to be transcribed in this tissue. A comparison of salivary gland transcriptomes prior to and after blood-feeding identified 52 and 41 transcripts that were significantly up-regulated and down-regulated, respectively. Ten genes were further selected to assess their role in the blood-feeding process using RNAi-mediated gene silencing methodology. Depletion of the salivary gland genes encoding D7L2, anophelin, peroxidase, the SG2 precursor, and a 5'nucleotidase gene significantly increased probing time of A. gambiae mosquitoes and thereby their capacity to blood-feed. Conclusions: The salivary gland transcriptome comprises approximately 38% of the total mosquito transcriptome and a small proportion of it is dynamically changing already at two hours in response to blood feeding. A better understanding of the salivary gland transcriptome and its function can contribute to the development of pathogen transmission control strategies and the identification of medically relevant bioactive compounds. Project description:Triatoma sordida salivary glands secrete a number of complex bioactive proteins during feeding that affect hemostatic and immunological systems from vertebrate hosts. In this study, salivary glands and saliva were collected from 5th instar nymphs and adult triatomines. Liquid chromatography tandem-mass spectrometry (LC-MS/MS) was used to identify T. sordida salivary proteins. A total of 133 proteins was identified. Lipocalins, Hemiptera specific families, CRiSP/Antigen-5 and Kazal-type protein inhibitors proteins match to 42.01%, 14.32%, 5.86% and 1.91% of the total spectrum count, respectively. These proteins are involved in several physiological roles, including anti-coagulant, immunity and anti-microorganism. Salivary proteins contribute to successful feeding.
