Project description:Metabolic dysfunction-associated fatty liver disease (MASLD), the hepatic manifestation of obesity and type 2 diabetes (T2D), can progress to metabolic dysfunction-associated steatohepatitis (MASH) and fibrosis. MASLD is characterized by elevated hepatic lipid accumulation (steatosis) and insulin resistance. Ketogenic diet (KD), a high-fat, low-carbohydrate diet, induces hepatic insulin resistance and steatosis in animal models through unknown mechanisms. Our studies demonstrate the importance of adipose tissue-liver crosstalk in mediating MASLD progression and identify adipocyte IL-6-gp130 as a potential therapeutic target.

Project description:Metabolic dysfunction-associated steatotic liver disease (MASLD) is a growing health concern. Better understanding of factors that prevent hepatic lipid accumulation may help design new strategies to prevent or treat MASLD. We have previously shown that adipocyte microsomal triglyceride transfer protein (MTP) regulates intracellular lipolysis by inhibiting adipose triglyceride lipase (ATGL) activity. Here, we show that adipose-specific MTP knockout mice (A-Mttp−/−) fed an obesogenic diet exhibit moderately high plasma triglyceride levels and less hepatic steatosis compared to their wild-type counterparts. Mechanistic studies revealed that high plasma triglycerides are due to increased hepatic triglyceride synthesis and production of triglyceride-rich lipoproteins from the livers of A-Mttp−/− mice. In addition, hepatocytes from these mice showed increased fatty acid oxidation and this could be a contributing factor for less hepatic steatosis. Studies aimed at understanding how adipose MTP regulated hepatic triglyceride-rich lipoprotein production and fatty acid oxidation revealed that adipose MTP modulates the release of agonists that activate hepatic PPARα activity. Our study highlights the significance of the regulated movement of fatty acid from adipose tissue to the liver in maintaining a healthy liver. It is likely that adipocyte fatty acid release can be modulated to reduce hepatic steatosis.

Project description:Adipose tissue dysfunction drives hepatic lipid overload in metabolic dysfunction-associated steatotic liver disease (MASLD), yet the involvement of adipose tissue-derived small extracellular vesicles (sEVs) remains unclear. Herein, we showed that transplanting adipose tissue from high‑fat diet-fed male mice exacerbated insulin resistance and hepatic steatosis in lean recipients. Adipose‑specific Sirt3 overexpression (Sirt3AKI) alleviated insulin resistance and liver steatosis in high fat diet-fed male mice, whereas adipose‑specific Sirt3 knockdown aggravated these phenotypes. Moreover, adipose‑derived sEVs play key roles in hepatic lipid metabolism by delivering miRNAs in Sirt3AKI male mice. MicroRNA sequencing identified miR-30a-3p was increased in the circulating sEVs from high fat diet-fed male mice, while decreased in sEVs from Sirt3OE adipocytes and Sirt3AKI male mice. Mechanistically, miR‑30a‑3p promoted hepatic steatosis by targeting Becn1; this process was suppressed when Sirt3 downregulated miR‑30a‑3p transcription via deacetylation of H3K56. These findings highlight the critical role of adipose sEVs microRNAs in driving hepatocyte lipotoxicity, and suggest miR-30a-3p inhibition could be a therapeutic intervention for MASLD.

Project description:Emerging evidence suggests that the peptide hormone kisspeptin, signaling via the kisspeptin 1 receptor (KISS1R), decreases hepatic steatosis and protects against metabolic dysfunction-associated steatotic liver disease (MASLD). De novo lipogenesis (DNL) is a key contributor to the pathogenesis of MASLD. This study aimed to determine whether kisspeptin treatment of obese, diabetic mice directly attenuates DNL. DNL was assessed in kisspeptin-treated (KPA) or phosphate-buffer saline (PBS)-treated mouse livers, using a mouse model of MASLD employing metabolic tracing using 2H2O-enriched water and mass spectrometry. Kisspeptin-treated steatotic livers demonstrated a decrease in DNL of free fatty acids (FFA), known to be associated with type 2 diabetes, steatosis, and hepatocellular carcinoma. Enhancement of KISS1R signaling has a therapeutic effect in limiting DNL, suggesting a crucial role in restricting the pathogenesis and progression of MASLD.

Project description:In our study, we constructed a Simple steatosis (NAFL) mouse model with a high-fructose and high-cholesterol (HFHC diet) for 12 weeks. Next, we performed RNA Sequencing analysis to reveal molecular features of Simple steatosis. As a result, we obtained hepatic genome-wide mRNA expression in the livers from two groups of mice (NAFL versus Normal).Using FDR < 0.01 and fold change > 2 as the threshold, we identified 176 differentially expressed genes (DEGs) in the livers of NAFL mice relative to the normal control mice, of which 118 up-regulated and 58 down-regulated. The top 5 up-regulated genes in NAFL mice included the genes encoding for Themis, Cfd, Aqp8, Osbpi3 and Scd1.

Project description:Co-agonists at the glucagon-like peptide-1/glucagon receptors (GLP1R/GCGR) show promise as treatments for metabolic dysfunction-associated steatotic liver disease (MASLD). Unlike GLP1, glucagon directly acts on the liver to reduce fat content. To date most metabolic studies have looked at heavily GLP1R-biased co-agonists and have not distinguished weight-loss versus weight loss-independent effects. We demonstrate that 24 days’ treatment with Dicretin, a GLP1/GCGR co-agonist with high potency at the GCGR, in mice with hepatic steatosis secondary to diet-induced obesity leads to superior reduction of hepatic lipid content when compared to Semaglutide or equivalent weight loss by calorie restriction. Hepatic transcriptomic and metabolomic profiling demonstrated many changes that were unique to Dicretin-treated mice: some known targets of glucagon signalling and others with as yet unclear physiological significance. Our study supports the development of GLP1/GCGR co-agonists for treatment of MASLD and related conditions.

Project description:To test whether NDGA attenuate dyslipidemia and hepatic steatosis by enhancing fatty acid oxidation through activation of PPAR-α. Using wild type (WT, C57BL/6) fed with chow diet as control, WT mice were either fed with high-fat diet or high-fat diet with NDGA (2.5g/kg food); ob/ob mice were fed with either chow or chow with NDGA (2.5 g/kg food), and maintained on the respective diets for 16 weeks. The expression of lipid metabolism related genes in the liver of these mice were analyzed using Phalanx GPL6845 platform (Mouse OneArray V1). Together with other biochemical/physiological data, our results suggest that the beneficial actions of NDGA on dyslipidemia and hepatic steatosis in ob/ob mice are exerted primarily through enhanced fatty acid oxidation and energy utilization via the activation of PPAR- α receptor activity. To examine the changes in gene expression in liver of WT and ob/ob mice with different NDGA diet treatment, RNA isolated from 3 animals of each group were used for studies of gene expression profiles. Phalanx GPL6845 platform (Mouse OneArray V1) was used for the microarrays analysis.
