Repurposing salmon calcitonin for Glioblastoma Treatment: Targeting YAP/TAZ via Hippo pathway activation

Project description:The calcitonin receptor (CTR) signaling axis has been implicated in growth regulation in Glioblastoma (GBM). This study examines the molecular and signaling effects of salmon calcitonin (sCT) treatment in GBM using proteomic, cellular, in vivo, and computational approaches. Reverse Phase Protein Array (RPPA) analysis was performed to assess changes in protein expression in glioma cells following sCT treatment and to identify signaling pathways associated with CTR activation. Complementary biochemical and functional assays were used to characterize CTR-mediated regulation of the Hippo pathway. CTR expression was found to be elevated in human and murine glioma stem-like cells in comparison to their differentiated counterparts, and intranasal sCT treatment lead to decrease in the GSC-initiated tumor growth in orthotopic mouse glioma model. In parallel, microsecond-scale all-atom molecular dynamics simulations were conducted to examine structural features of wild-type and patient-derived mutant CTR variants and their interactions with calcitonin and Gα subunits. Together, this work reveals mechanism of CT/CTR axis, therapeutic implications of sCT, and impact of structural alterations in patient-derived CTR variants in GBM.

Project description:Here, we successfully used NO as the direct electron acceptor for the enrichment of a microbial community in a continuous bioreactor. The enrichment culture, mainly comprised of two new organisms from the Sterolibacteriaceae family, grew on NO reduction to N2 and formate oxidation, with virtually no accumulation of N2O. The microbial growth kinetics of the enrichment culture as well as its affinity for different N-oxides were determined. In parallel, using metagenomics, metatranscriptomics, and metaproteomics, the biochemical reactions underlying the growth of these microorganisms on NO were investigated. This study demonstrates that microorganisms thrive and can be enriched on NO, and presents new opportunities to study microbial growth on this highly energetic and climate-active molecule that may have been pivotal in the evolution of aerobic respiration.

Molecular basis of NFIB-Mediated Regulation of Oncogenic Transcription

Project description:The Nuclear Factor I (NFI) family of transcription factors orchestrates key regulatory programs in development, differentiation, and metabolism, with dysregulation implicated in various pathological conditions, including cancer. Among them, NFIB displays significant context-dependent roles, functioning either as an oncogene or a tumor suppressor depending on cancer type. Despite its emerging clinical relevance, the molecular basis of NFIB-mediated DNA recognition and transcriptional regulation in cancer remains poorly understood. Using HeLa cells as a model, we demonstrate that NFIB promotes oncogenic phenotypes, as CRISPR-Cas9-mediated knockout significantly impairs cell proliferation, migration, and invasion. Transcriptomic analysis reveals that NFIB regulates a cohort of cancer-associated genes, including FGFR3 and PDGFRB. To elucidate the underlying mechanism, we performed biochemical characterization and found that both full-length NFIB and its DNA-binding domain (DBD) exist as monomers in solution and bind DNA with 1:1 stoichiometry, challenging long-standing models proposing DBD-mediated dimerization. High-resolution crystal structures of NFIBDBD bound to ChIP-seq-derived DNA motifs reveal a monomeric binding mode driven by base-specific recognition of the TGGCA sequence. Mutational disruption of key DNA-contacting residues abolishes both DNA binding and transcriptional activation in luciferase reporter assays. Together, these findings define the molecular mechanism underlying NFIB-dependent gene regulation and establish a structural framework for its oncogenic activity, providing insights with potential therapeutic implications for targeting NFIB-driven cancers.
