Investigation of therapeutic response markers for acupuncture in Parkinson's disease: An exploratory pilot study

Project description:Introduction: In our previous study, we demonstrated that acupuncture improved gait disturbances, such as hypometric gait, in patients with Parkinson’s disease (PD). To investigate specific genes that indicate a therapeutic response to acupuncture, we analyzed transcriptome alterations following acupuncture using blood samples collected in our previous clinical trial. Methods: We reanalyzed the clinical data of patients and selected a representative patient with PD for transcriptomic analysis following acupuncture. We examined the expression changes of PD biomarker genes known to be consistently dysregulated in both the brain and blood in patients with PD. We validated these gene expression changes using quantitative real-time polymerase chain reaction (qPCR) in the blood of five other patients with PD who received acupuncture treatment. Results: After acupuncture treatment, the transcriptomic alterations in the representative patient were similar to those induced by dopaminergic therapy. Among PD biomarkers, ankyrin repeat domain 22 (ANKRD22), which is upregulated after dopaminergic therapy, and synapsin 1 (SYN1), which is the common gene to indicate synaptic dysfunction in PD, were upregulated following acupuncture. These alterations correlated with changes of gait parameters in patients with PD.alterations following acupuncture using blood samples collected in our previous clinical trial. Conclusion: This is the first report on gene expression changes and improvement of gait disturbance in patients with PD following acupuncture. Twenty-one LS participants with a confirmed pathogenic mutation in one of four DNA mismatch repair (MMR) genes were enrolled between April 2018 and January 2019. Follow-up was conducted 1-year from the start of the intervention. Statistical analysis by both per-protocol and intention-to-treat was performed from March to May 2021.

Transcriptome analysis and differentially expressed gene screening for hypoxic-ischemic brain damage in rats treated with acupuncture

Project description:Purpose: Hypoxic-ischemic brain damage (HIBD) is one of the most common critical diseases in neonates and has high mortality and disability rates. Increasing evidence shows that long non-coding RNAs(lncRNA) plays an important role in the development of HIBD. Recently, acupuncture therapy has been found to be effective in the treatment of HIBD. However, the mechanism of lncRNA in acupuncture treatment of HIBD is still unclear. Method: In this study, a whole transcriptometric RNA-sequencing technique was applied to investigate transcriptome changes caused by acupuncture in PFC. Results: A total of 48 mRNAs (7 up-regulated and 41 down-regulated) and 65 lncRNAs (17 up-regulated and 48 down-regulated) was identified relate to acupuncture group and model group. According to Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) analysis, we found several lncRNAs and their target mRNAs were related to PI3K-Akt signaling pathway, TNF signaling pathway and NOD-like receptor signaling pathway, etc. Conclusion: we found several significant lncRNAs whose corresponding target genes might be the targets of acupuncture therapy for HIBD. The results of our study may provide new perspectives on the mechanism of acupuncture and affect the diagnosis and therapy of HIBD

Acupuncture potentiates anti-PD-1 efficacy by promoting CD5+ dentritic cells and T cell-mediated tumor immunity in breast cancer

Project description:Acupuncture has been considered as an available complement therapy against various malignancy cancer by regulating immunity. However, it is not clear whether acupuncture influences the antitumor immune efficacy of PD-1 inhibitor. This study evaluates the efficacy of acupuncture combined with PD-1 inhibitor on the antitumor immune response against breast cancer and revealed the potential molecular mechanisms based on RNA-Seq transcriptome analysis. The results depicted that acupuncture combined with PD-1 inhibitor significantly hindered tumor development by inducing tumor cell apoptosis and inhibiting tumor growth, angiogenesis and metastasis. RNA-Seq analysis indicated that the different expression genes upon acupuncture intervention mainly enriched in immune response, T cell activation and cytokine interaction etc. items containing immune cell-related CD genes (CD5, CD4 and CD8 etc.). Acupuncture stimulation enhanced CD5 expression that had a positive correlation with overall survival of breast cancer patients. The improved immunity including the enhancement of CD5+ dendritic cell, CD4+ and CD8+ T cell content as well as various cytokine (IL-2, IL-6, TNF-α, IFN-γ) secretion in serum was found upon acupuncture combination with PD-1 inhibitor treatment. Taken together, present work reveals that acupuncture intervention promotes the antitumor immune response of PD-1 inhibitor, providing a prospective therapy for ICI therapy against breast cancer.

Remote ischemic preconditioning in on-pump coronary artery bypass graft surgery

Project description:In this placebo-controlled randomized controlled trial, we tested whether remote ischemic preconditioning (RIPC) elicited by four 5-minute cycles of 300 mmHg of cuff inflation/deflation of the lower limb would reduce myocardial necrosis in isoflurane-anesthetized patients undergoing on-pump coronary artery bypass graft surgery. Secondary outcomes were the perioperative release of the biomarkers NTproBNP, hsCRP, S100, atrial transcriptional profiles, and short- and long-term clinical outcomes. RIPC with concomitantly applied isoflurane did not affect the release of biomarkers or clinical outcome. NTproBNP release correlated with isoflurane- but not RIPC-induced transcriptional changes.
