Research on the progression and treatment resistance mechanism of liver cancer

ABSTRACT: Hepatocellular carcinoma (HCC) is a highly aggressive malignancy characterized by rapid progression, therapeutic resistance, and poor prognosis. This project aims to investigate the molecular mechanisms underlying HCC development and progression, with a particular focus on signaling pathways that regulate tumor growth, invasion, and treatment response. By integrating transcriptomic profiling with functional and prognostic analyses, the study seeks to identify key regulatory factors associated with disease progression and therapeutic efficacy. The findings are expected to provide mechanistic insights into HCC pathogenesis and offer a scientific basis for evaluating potential therapeutic strategies and prognostic biomarkers.

Project description:Hepatocellular carcinoma (HCC) is a formidable malignancy with limited effective therapeutic avenues. This study was designed to investigate the role of transglutaminase 2 (TGM2) in promoting HCC progression and assess its potential as a target for therapeutic intervention in HCC treatment.TMG2 expression was positively related to a higher AFP level, poor differentiation, and a later BCLC stage. Tgm2 deficiency or H3Q5ser inhibition notably restrained HCC progression. Mechanism research revealed that TGM2-mediated H3Q5ser modifications promote HCC progression via MYC pathway signaling. Furthermore, transcriptional intermediary factor 1 beta (TIF1-β/TRIM28) mediated the recruitment of TGM2 by MYC to facilitate H3Q5ser modifications on MYC targets. Finally, targeting the TGM2 transglutaminase activity significantly suppressed HCC progression in preclinical models.

RNA biotype-associated molecular classification in hepatitis B-related hepatocellular carcinoma

Project description:Recent advance of RNA-seq technology enabled us to profile the diverse variations of RNA transcripts precisely, including the variants from alternative splicing as well as non-coding transcripts. Here, by performing transcriptome profiling including coding and noncoding transcripts, we identify four molecular subtypes of hepatitis B-related hepatocellular carcinoma (HCC) patients that are characterized by enriched expression of the RNA biotypes of noncoding (NC) and immune-related transcripts (IM) (i.e., IM+NC+, IM-NC+, IM+NC-, IM-NC-). The subtype IM+NC+ shows better prognostic outcome, while the subtype IM+NC- shows the worst prognostic outcome, respectively. Further interrogation of the subtypes identifies long noncoding transcripts (i.e., LINC00844, C3P1, and TRPG1-AS1) as well as an alternatively spliced event of USO1 that play pivotal roles in HCC progression. In addition, we report an oncogenic fusion transcript SLC39A14-PIWIL2 that promotes an aggressive phenotype of HCC. Our comprehensive and systematic analysis of HCC transcriptome identify RNA biotype-based molecular classification, revealing novel driver transcriptome variants that can be potential biomarkers and/or therapeutic targets for precision medicine.

Transcriptome analysis in tumor budding positive hepatocellular carcinoma tissue samples relative to tumor budding negative hepatocellular carcinoma tissue samples

Project description:Tumor budding (TB) has become a crucial factor for predicting the malignancy grade and prognostic outcome for multiple solid cancers.Studies have investigated the prognostic value of TB within hepatocellular carcinoma (HCC). However, its molecular mechanism within HCC remains unclear. This is the first study to discover and compare the expression of differentially expressed genes (DEGs) between TB-positive and TB-negative HCC tissues. Total RNA was extracted from 40 fresh HCC tissue samples with different TB statuses and then sequenced. This study provides insights into the possible mechanisms of TB in HCC and revealed potential predictive diagnosis markers and treatment targets for HCC.

Study of gene expression of human liver Hepatocellular carcinoma

Project description:Hepatocellular carcinoma (HCC) affects millions of people worldwide and is a lethal malignancy for which there are no effective therapies. To identify prognostic gene markers for liver cancer, we conducted transcriptome profiling of frozen tissues (tumor and non-tumor) from 300 early-to-advanced stage HCCs plus 40 cirrhotic and 6 normal livers. We have profiles 268 HCC tumor, 243 adjacent non-tumor, 40 cirrhotic and 6 healthy liver samples.

Microarray of SIRT6 KO in hepatocellular carcinoma

Project description:We identified the specific role of Sirtuin 6 (SIRT6) in the progression of hepatocellular carcinoma (HCC) and evaluated its therapeutic and prognostic potential. SIRT6 knockdown by shRNA inhibited the proliferation of HCC cells; moreover, depletion of SIRT6 suppressed HCC tumor growth in vivo. SIRT6 silencing significantly down regulated the growth of HCC cell lines via induction of cellular senescence, which was attributed by DNA damage in p16/Rb and p53/p21-independent manners. We also showed that SIRT6 knockdown increased the number of G2/M phase arrested cells by increasing cyclin B1 and 14-3-3-∂. Moreover, SIRT6 levels were significantly higher in HCC cell lines than a normal hepatic cell line, and were higher in HCC than non-tumor tissues. Aberrant SIRT6 expression correlated with poor prognostic features of HCC. Taken together, our findings suggest SIRT6 may act as a tumor promoter by preventing cellular senescence attributed by DNA damage and suggests that targeting SIRT6 could be a promising therapeutic approach for cancer treatment.

Plectin-mediated cytoskeletal crosstalk as a target for inhibition of hepatocellular carcinoma growth and metastasis – subcellular in vitro profiling

Project description:The most common primary malignancy of the liver, hepatocellular carcinoma (HCC), is a heterogeneous tumor entity with high metastatic potential and complex pathophysiology. Increasing evidence suggests that tissue mechanics plays a critical role in tumor onset and progression. Here we show that plectin, a major cytoskeletal crosslinker protein, plays a crucial role in mechanical homeostasis and mechanosensitive oncogenic signaling that drives hepatocarcinogenesis. Our expression analyses revealed elevated plectin levels in liver tumors, which correlated with poor prognosis for HCC patients. Using autochthonous and orthotopic mouse models we demonstrated that genetic and pharmacological inactivation of plectin potently suppressed the initiation and growth of HCC. Moreover, plectin targeting potently inhibited the invasion potential of human HCC cells and reduced their metastatic outgrowth in the lung. Proteomic and phosphoproteomic profiling linked plectin-dependent disruption of cytoskeletal networks to attenuation of oncogenic FAK, MAPK/Erk, and PI3K/AKT signatures. Importantly, by combining cell line-based and murine HCC models, we show that plectin inhibitor plecstatin-1 (PST) is well-tolerated and potently inhibits HCC progression. In conclusion, our study demonstrates that plectin-controlled cytoarchitecture is a key determinant of HCC development and suggests that pharmacologically induced disruption of mechanical homeostasis may represent a new therapeutic strategy for HCC treatment.

Streptococcus anginosus promotes hepatocellular carcinoma progression and BET inhibitor resistance by inducing BRD4 lactylation

Project description:<p>Hepatocellular carcinoma (HCC) is a highly aggressive form of carcinoma with a poor prognosis. Diagnostic markers and potential therapeutic targets for HCC are urgently needed. Here, we demonstrate that fecal microbiota transplantation (FMT) using stool samples from HCC patients significantly promote the growth of patient-derived xenografts (PDXs). 16s-rRNA sequencing combined with qPCR reveal that Streptococcus anginosus (S. anginosus) is enriched in the stool samples of HCC patients and HCC-FMT mice. S. anginosus monocolonization recapitulates the tumor-promoting effect of HCC-FMT in germ-free (GF) mice. Mechanistically, S. anginosus plays a critical role in regulating the downstream pathways of BRD4 via its metabolites, thereby promoting the activation of oncogenic Myc and E2F signaling pathways, HCC progression and BET inhibitor (BETi) resistance. S. anginosus-derived lactate promotes BRD4 lactylation at the K317 site via AARS1, thereby inhibiting BRD4 ubiquitination and degradation. Notably, blockade of the S. anginosus-BRD4 lactylation axis improves the BETi responsiveness in both syngeneic mouse models and PDXs. Overall, our study highlights the oncogenic role of the S. anginosus-BRD4 signaling in the gut-liver axis and suggests its potential as a non-invasive biomarker and therapeutic target for HCC.</p>
