Proteomic Sequencing of Extracellular Vesicles Derived from Momordica charantia

ABSTRACT: This project aims to conduct a comprehensive proteomic sequencing analysis of extracellular vesicles (EVs) derived from Momordica charantia, systematically identify the protein composition and abundance characteristics of these EVs, and further explore the potential biological functions and molecular mechanisms of the key differential proteins in regulating cell physiological activities, which lays a theoretical foundation for the development and utilization of bioactive substances derived from Momordica charantia.

Osteoblasts secrete miRNA-containing extracellular vesicles that enhance expansion of human umbilical cord blood cells

Project description:Osteolineage cells represent one of the critical bone marrow niche components that support maintenance of hematopoietic stem and progenitor cells (HSPCs). Recent studies demonstrate that extracellular vesicles (EVs) regulate stem cell development via horizontal transfer of bioactive cargo, including microRNAs (miRNAs). Here, we characterize the miRNA profile of EVs secreted by human osteoblasts and study their biological effect of on human umbilical cord blood-derived CD34+ HSPCs by sequencing, gene expression and biochemical analyses. Using next-generation sequencing we show that osteoblast-derived EVs contain highly abundant miRNAs specifically enriched in EVs, including critical regulators of hematopoietic proliferation (e.g., miR-29a). EV treatment of CD34+ HSPCs alters the expression of candidate miRNA targets, such as HBP1, BCL2 and PTEN. Furthermore, EVs enhance proliferation of CD34+ cells and their immature subsets in growth factor-driven ex vivo expansion cultures. Importantly, EV-expanded cells retain their differentiation capacity in vitro and show successful engraftment in NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ (NSG) mice in vivo. These discoveries reveal a novel osteoblast-derived EV-mediated mechanism for regulation of HSPC proliferation and warrant consideration of EV-miRNAs for the development of expansion strategies to treat hematological disorders.

Multiple roles of extracellular vesicles in promoting microbial driven elemental cycle

Project description:<p>Microbial driven manganese (Mn) reduction influences the geochemical cycling of Mn and the environmental fate of various organic and inorganic substances. Extracellular vesicles (EVs) are known to impact microbial metabolic activities, but their specific role in Mn reduction remains arcane. Here, we investigated the potential and mechanistic roles of EVs in this process by integrating metagenomic analysis of environmental EVs with functional assays in representative dissimilatory metal-reducing bacteria. Microbial physiological assays and in situ electrochemical analyses revealed that the addition of EVs enhanced cellular metabolism, promoted biofilm formation, and facilitated extracellular electron transfer. The presence of various redox enzymes and metabolites in EVs contributed to more efficient substrate utilization and energy conservation. The electron transfer-promoting effect of EVs is primarily attributed to OmcA/MtrC, CymA, and FMN. These findings highlight the multifaceted role of EVs in microbial Mn reduction and suggest their broader involvement in elemental cycling and pollutant degradation.</p>

Multi-omics analysis reveals Jianpi formula-derived bioactive peptide-YG-22 potentially inhibited colorectal cancer via regulating epigenetic reprogram and signal pathway regulation [ChIP-Seq]

Project description:Colorectal cancer (CRC) is a prevalent malignancy worldwide, often treated with chemotherapy despite its limitations, including adverse effects and resistance. Chemotherapy combined with the traditional Chinese medicine (TCM) Jianpi formula has been demonstrated to improve efficacy. In this study, we aim to screen bioactive peptides derived from the blood of CRC patients through peptidomics and explore the molecular mechanisms of the candidate peptides in HCT116 cells using multi-omics analysis. Differential peptides were identified in plasma samples from patients treated with chemotherapy alone and those receiving the combined therapy. Among these, YG-22 exhibited the strongest cytotoxic effect on HCT116 cells, reducing viability in a dose- and time-dependent manner. Transcriptome analysis highlighted the modulation of key pathways involved in lysosome-mediated degradation and apoptosis, while metabolomic profiling indicated disruptions in tumor-supportive metabolic pathways. Additionally, chromatin accessibility and histone modifications suggested epigenetic reprogramming induced by YG-22. These findings demonstrate that combining chemotherapy with TCM enriches the molecular landscape and generates bioactive peptides with strong antitumor activity. Furthermore, this study also lays the foundation for further development of peptide-based therapies and highlights the value of combining traditional and modern therapeutic strategies for CRC management.

Characterization of Sheep Milk Extracellular Vesicle-miRNA by Sequencing and Comparison with Cow milk

Project description:Milk can mediate maternal-neonatal signal transmission by the bioactive component-extracellular vesicles (EVs), which select specific types of miRNA to encapsulate. The miRNA profiling of sheep milk EVs was characterized by sequencing and compared with that of cow milk. Sheep milk EVs contained various small RNAs, including tRNA, Cis-regulatory element, rRNA, snRNA, other Rfam RNA, and miRNA, which held about 36% of all the small RNAs. Totally 84 types of miRNAs were annotated with Ovis aries by miRBase (version 22.0) in sheep milk EVs, with 75 shared types of miRNAs in all samples. Fourteen sheep milk EV-miRNAs in the top 20, occupying 98% of the total expression, were immune-related.
