Proteomic IP-MS analysis of Asprosin/PFKP in regulating cardiac hypertrophy model

ABSTRACT: This dataset contains two parts of proteomics data: a) the identification and analysis results of co-immunoprecipitation (Co-IP: Flag-asprosin/HA-PFKP) in cardiomyocytes overexpressing Flag-asprosin/HA-PFKP; b) the identification and analysis results of co-immunoprecipitation (Co-IP) using IgG as a control.

Project description:To illustrate EMP1-interacting proteins, we performed co-immunoprecipitation (co-IP) followed by mass spectrometry using HSPCs (Lin-c-Kit+) to characterize protein interactomes of EMP1. Murine HSPCs (Lin-c-Kit+) were infected with retrovirus to force EMP1 overexpression (with HA tag). Then IgG or HA antibodies were used to conducted co-immunoprecipitation (co-IP), followed by and mass spectrometry analysis.

We identified P4HA2 binding proteins with anti-HA IP followed by (MS)-based proteomic analysis in 293T cells overexpressing HA-tagged P4HA2. To speculate that mTOR can be hydroxylated by P4HA2. We used 293T cells overexpressing Flag-mTOR, Flag-mTOR/HA-P4HA2, and Flag-mTOR/HA-P4HA2-overexpressing cells treated with EDHB, we conducted an anti-Flag IP to capture and enrich mTOR protein, and the immunoprecipitates were subjected to MS analysis.
