Project description:Chronic Pseudomas aeruginosa infection in the lung is a common in people with cystic fibrosis (CF). Current therapies for CF fail to eliminate persistent bacterial infections, chronic inflammation, or irreversible lung damage. Our group engineered mesenchymal stromal cell (MSC)-derived extracellular vesicles (EVs) to carry the microRNA let-7b-5p as a dual anti-infective and anti-inflammatory treatment. In a preclinical CF mice model, we found that let-7b-5p-loaded MSC EVs reduced P. aeruginosa burden, immune cells and proinflammatory cytokines in the lungs. This research hypothesized two mechanisms of the observed effects in the mouse model: anti-inflammatory properties of the let-7b-5p-loaded MSC EVs and inhibition of antibiotic-resistant P. aeruginosa biofilm formation in CF airways. Primary human broncial epithelial cells (pHBECs) were exposed to P. aeruginosa and treated with differet MSC EV conditions. The results demonstrated that MSC EVs engineered to contain let-7b-5p effectively blocked the formation of P. aeruginosa biofilms on pHBECs while also reducing P. aeruginosa-induced inflammation by CF-pHBECs.

Project description:Perineural invasion (PNI) represents a unique biological feature causing the poor prognosis of pancreatic ductal adenocarcinoma (PDAC), especially with KRAS mutation. Extracellular vesicle (EVs)-packaged circRNAs function as essential mediator for tumor microenviroment communication to trigger PDAC cells invading into the surrounding regions and lead to distant metastasis. However, the regulatory mechanism of exosomal circRNA in PNI of KRAS-mutated PDAC has not been elucidated. Herein, we identified an KRASG12D-mutation responsive exosomal circRNA-circPNIT, which was positively correlated with PNI of PDAC. Functionally, KRASG12D PDAC-derived EV-packaged circPNIT promotes axonogenesis and PNI both in vitro and in vivo. Mechanistically, circPNIT-mediated Rab5B-CD109 interplay bypassed traditional endosomal trafficking to anchor Rab5B in the lipid rafts of multivescular body and fostered the secretion of circPNIT as a cargo of CD109+EVs. Subsequently, CD109+EVs achieved the targeted delivery of circPNIT to neurons by recognizing their surface receptor TRPV1 and facilitated DSCAML1 transcription-induced axonogenesis, which in turn enhanced the PNI by activating GFRα1/RET pathway. Importantly, we established an engineered si-cirPNIT-loaded CD109+EVs to dramatically inhibit PNI of PDAC in a KRASG12D/+ Trp53R172H/+ Pdx-1-Cre mice model. Collectively, Our findings highlight the mechanism underlying EV-packaed circRNA mediating PNI of KRAS mutated-PDAC dependent of a Rab5B endosomal bypass, supporting circPINT as an effective target for the treatment of neuro-metastatic PDAC

Project description:Mesenchymal stromal cells (MSCs) are multipotent stromal cells with regenerative, immunomodulatory, and anti-inflammatory properties, making them a promising tool in cell-based therapies. Increasing evidence suggests the therapeutic effects of MSCs are largely mediated by secreted paracrine factors and extracellular vesicles (EVs), which facilitate extracellular communication by transferring bioactive molecules such as proteins, lipids, and nucleic acids to target cells. The lipid composition of EVs is critical for their stability, uptake, and functional activity. Sphingomyelinase (SMase), an enzyme that hydrolyzes sphingomyelin into ceramide, plays a key role in EV biogenesis by influencing membrane curvature allowing for invagination and budding of multivesicular bodies (MVBs). However, the impact of SMase treatment on the characteristics and therapeutic potential of MSC-derived EVs remains largely unexplored. In this study, human MSCs were cultured with SMase for 24 hours and EVs were isolated via ultracentrifugation. EV size distribution was analyzed using nanoparticle tracking analysis (NTA), microRNA (miRNA) profile was determined using RNA-sequencing, while lipidomic and proteomic profiling were assessed using mass spectrometry. Functional assays were conducted to assess changes in bioactivity. NTA revealed a shift in EV production, with a higher vesicle concentration in SMase induced MSCs compared to untreated cells. SMase treatment significantly altered MSC-EV composition, leading to increased ceramide and glycosphingolipids. In addition, the modifications in protein and miRNA EV cargo were found to be associated with pathways involving TNF-α signaling, vesicle trafficking, wound healing, and angiogenesis. Functional assays indicated greater suppression of TNF-α in activated macrophages and greater tubular network formation in HUVECS. These findings provide new insights into distinct biophysical and biochemical changes in MSC-derived EVs following SMase stimulation, potentially enhancing their therapeutic properties and may inform strategies for optimizing future EV-based therapies.

Project description:Sperm maturation depends on exposure to specific microenvironments within the different segments of the epididymis, but mechanisms underlying how these microenvironments are produced or maintained are not well understood. We hypothesize that epididymal extracellular vesicles (EVs) could play a role in this process, and therefore set out to test whether the EVs from different regions of the epididymis can serve as a form of paracrine communication between epithelial cells. Domestic cat tissues were used to develop a reproducible in vitro culture system for corpus epididymis explants that were then exposed to EVs collected from upstream (i.e., caput) segments. The impacts of different culture or exposure conditions were compared by analyzing the morphology, apoptosis, transcriptional activity, and gene expression in the explants. Here, we report the development of the first in vitro culture system for epididymal tissue explants in the domestic cat model. Using this system, we found that EVs from the caput segment have a significant effect on the transcriptional profile of tissue from the corpus segment (1,233 differentially expressed genes due to EV supplementation). Of note, expression of genes associated with regulation of epithelial cell differentiation and cytokine signaling in the epididymis were regulated by the presence of EVs. Together, our findings comprise the first report of paracrine control of segmental gene regulation by epididymal EVs in any species. These results contribute to a better understanding of epididymis biology and could lead to techniques to enhance or suppress male fertility.

Project description:Cyclic dinucleotide (CDN) agonists of the STimulator of InterferoN Genes (STING) pathway have shown immune activation and tumor clearance in pre-clinical models. However, CDNs administered intratumorally also promote STING activation leading to direct cytotoxicity of many cell types in the tumor microenvironment (TME), systemic inflammation due to rapid tumor extravasation of the CDN, and immune ablation in the TME. These result in a failure to establish immunological memory. ExoSTING, an engineered extracellular vesicle (EV) exogenously loaded with CDN, enhances the potency of CDN and preferentially activates antigen presenting cells in the TME. Following intratumoral injection, exoSTING was retained within the tumor, enhanced local Th1 responses and recruitment of CD8+ T cells, and generated systemic anti-tumor immunity to the tumor. ExoSTING at therapeutically active doses did not induce systemic inflammatory cytokines, resulting in an enhanced therapeutic window. ExoSTING is a novel, differentiated therapeutic candidate that leverages the natural biology of EVs to enhance the activity of CDNs.

Project description:Mesenchymal stem cells (MSC) secrete naturally extracellular vesicles (EV) with a regenerative profile, and an increasing number of studies have focused on the employment of EV for therapeutic drug delivery. However, EV are usually produced by the cells at low quantities and packed with unnecessary cytoplasmic components that are unfavorable for further drug loading. In this study, we developed a simple vesicle generation process from the cell membranes directly, after which they are called synthetic eukaryotic vesicles (SyEV or reNV). We hypothesized that MSC-derived SyEV can be efficiently loaded with an anti-inflammatory drug and the loaded vesicles can strongly suppress bacterial outer membrane vesicle (OMV)-induced sepsis. SyEV were generated from MSC membranes through serial extrusion, ionic stress and subsequent revesiculation, leading to high yield and purity with very few cytosolic contaminants. When these SyEV were given to macrophages and mice exposed to OMV, the release of pro-inflammatory cytokines was significantly inhibited comparable to natural EV. Then, we encapsulated SyEV with a peptide targeting Myd88 and showed the enhanced therapeutic potential of the loaded vesicles in OMV-induced macrophages. Further, in vivo study indicated that the peptide-encapsulated SyEV could dramatically suppress cytokine release into the systemic circulation via a synergistic way. Taken together, these data suggest that SyEV-based therapeutics is a highly interesting platform to deliver an advanced therapeutic drug for the treatment of inflammatory diseases without severe side effects.

Project description:Extracellular vesicles (EVs) are membrane-encapsulated particles that vary greatly in size and content. Although both normal and tumor cells produce an array of small EVs (S-EVs), only tumor cells have been found to release a population of atypically large EVs (L-EVs), also known as large oncosomes (LO). Little is known about the cargo of L-EVs, and how it differs from the S-EVs. Given the potential for tumor-derived L-EVs to provide insights into disease mechanisms and course, we performed proteomic analysis of L- and S-EVs derived from three cancer models followed by comparative proteomic and transcriptomic analysis of EVs from the prostate cancer model. 1) We identified both common and model-specific protein signatures for L- and S-EVs. 2) A subset of the proteins enriched in prostate cancer cell-derived L-EVs were also identified in L-EVs isolated from plasma of patients with metastatic prostate cancer. 3) Proteins enriched in L-EV that were also identified at the transcript level were mostly mitochondrial in origin, as confirmed by single vesicle RNA-Seq. Additionally, the L-EV mitochondrial signature was detected in plasma-derived L-EVs and distinguished patients with prostate cancer from cancer-free individuals as well as patients with metastatic prostate cancer from those with localized disease, corroborating the relevance of an integrative multi-analyte approach focused on L-EVs for liquid biopsy.

Project description:Reactive protein cysteine thiolates are instrumental in redox regulation. Aging associated to oxidative stress, can impair redox signaling by damaging essential cysteine thiolates. Our initial study found that cardiac-specific overexpression of catalase, can protect from oxidative stress and delay cardiac aging in mice. The Project aimed to investigate differential reversible cysteine thiol oxidation in cardiac proteome of wild type and Cat transgenic (Tg) mice, using “Tandem Mass Tag” (TMT) labeling strategy and mass spectrometry. Our results show globally decreased cysteine thiol occupancy in cardiac proteins in Cat Tg mice. The majority of proteins with differentially modified thiols may be involved in pathways of aging and cardiac disease including cellular stress response, proteostasis and apoptosis. Overexpressed catalase may modulate these protein cysteine thiol oxidations resulting in delayed cardiac aging and related pathologies.
