Defective O-fucosylation of EDIL3 attenuates invasion and vascular remodeling of trophoblasts via LIFR signaling pathway in preeclampsia

Project description:Preeclampsia (PE) is a hypertensive disorder, which affects up to 10% of pregnancies worldwide. The primary etiology is considered to be abnormal development and function of placental cells called trophoblasts. We previously developed a two-step protocol for differentiation of human pluripotent stem cells, first into cytotrophoblast (CTB) progenitor-like cells, and then into both syncytiotrophoblast (STB)- and extravillous trophoblast (EVT)-like cells, and showed that this protocol can be used to model both normal and abnormal trophoblast differentiation. We have now applied this protocol to differentiate induced pluripotent stem cells (iPSC) derived from placentas of pregnancies with or without PE. While we did not note any differences in CTB induction, PE-iPSC-derived CTB showed a defect in syncytialization, and had a >180-fold reduction in expression of PSG4, a mature STB marker (p<0.02). While EVT formation did not appear to be altered, the invasive capacity of PE-iPSC-derived EVT was non-responsive to a decrease in oxygen tension, while that of non-PE-iPSC-derived EVT was enhanced 3.8-fold. RNA sequencing analysis showed gene expression changes consistent with defects in STB formation and response to hypoxia in PE-iPSC derived trophoblast. However, differences in DNA methylation were minimal, with only documented differences consistent with part of the response to hypoxia. Overall, PE-associated iPSC recapitulated multiple defects associated with placental dysfunction, with their lack of response to decreased oxygen tension emphasizing the importance of the interface with the maternal microenvironment in normal placentation.

Project description:Introduction: To determine the miRNA expression profile in pregnancies complicated by Preeclampsia (PE) and compare it to uncomplicated pregnancies. Methods: Seventeen placentas from women with PE, [all of them being late onset PE (LOPE)], as well as 17 placentas from uncomplicated pregnancies were analyzed using miRNA NGS. For statistical analyses the MATLAB® simulation environment was applied. The expression of miR-99b and miR-23b were verified using Quantitative Real-Time Polymerase Chain Reaction. Results: Two miRNAs were found dysregulated in PE complicated placentas. Statistical analysis revealed that miR-99b and miR-23b were under-expressed in complicated placentas as compared to controls. Discussion: Since specific miRNAs can differentiate complicated from uncomplicated pregnancies, they may be considered as putative PE-specific biomarkers.

Project description:Preeclampsia (PE) is a common pregnancy disorder, and it complicates 5~7% of all pregnancies. At present, termination of pregnancy is the only curative strategy for PE. To explore a potential target for PE treatment, we collected placental tissue samples from patients with preeclampsia (PE) and normal pregnant women (N), and performed high-throughput RNA sequencing. We found that FOXP2 was significantly downregulated in placental samples of patients with PE compared with the controls.

Previous studies have suggested that fatty acid binding protein 4 (FABP4) is involved in lipid metabolism and oxidative stress regulation in various metabolic and cardiovascular diseases. However, its specific role and underlying mechanisms in PE, particularly in trophoblast function and mitochondrial homeostasis, remain unclear. This study aims to investigate the role of FABP4 in the pathogenesis of PE by exploring its effects on trophoblast cell function and mitochondrial oxidative stress through regulating lipid metabolism.

Project description:Preeclampsia, a life threatening hypertensive disorder of pregnancy, is a leading cause of maternal and perinatal morbidity and mortality. Its early onset form (EO PE), requiring delivery before 34 weeks of gestation, is particularly severe and closely linked to defective trophoblast differentiation. Here, we identify BRCA1-associated protein 1 (BAP1) and its cofactors ASXL2 and ASXL3 are upregulated in EO PE placentas. Enforced BAP1 expression in human trophoblast stem cells reinforced epithelial identity, enhanced adhesion, and impaired both extravillous trophoblast differentiation and syncytiotrophoblast formation. Integrated transcriptomic and proteomic analyses revealed suppression of lineage-specific pathways alongside maintenance of progenitor-like and pro-inflammatory signatures. In trophoblast organoids, excess of BAP1 disrupted syncytial maturation and induced interferon-driven pathways overlapping with EO PE transcriptomes. Together, these findings establish BAP1 as a key regulator of human trophoblast differentiation and implicate its dysregulation in the pathogenesis of EO PE, providing mechanistic insight into the cellular basis of placental dysfunction.

Project description:Placentation requires the proper regulation of extravillous trophoblast (EVT) migration and invasion into the decidua and maternal vasculature, processes which are initiated in physiologic hypoxic conditions. Abnormal EVT migration and/or invasion have been suggested to lead to pregnancy complications, such as preeclampsia. The objectives of this study are to determine how exposure to hypoxia impacts gene expression and cellular motility of first trimester trophoblasts, and to assess if expression of migration-associated genes is dysregulated in 2nd trimester chorionic villous samples (CVS) from preeclampsia pregnancies relative to CVS from healthy pregnancies. The 1st trimester trophoblast cell line, HTR8/SVneo, was used to investigate the relationship between hypoxia and Notch signaling in trophoblast migration and invasion. RNA sequencing and quantitative RT-PCR analyses show that exposure to hypoxia (2.5% O2) activates Notch signaling in HTR-8/SVneo. We demonstrate that exposure of HTR-8/SVneo to hypoxia induces expression of genes associated with cellular migration and invasion and increases HTR-8/SVneo cellular migration and invasion, whereas inhibition of gamma-secretase decreases Notch signaling and decreases HTR-8/SVneo migration and invasion. Analysis of RNA sequencing data from CVS of preeclampsia and uncomplicated pregnancies identified significant differentially expressed genes that are involved in cellular migration and invasion. Decreased expression of migration and invasion genes in CVS from preeclampsia pregnancies, may impair trophoblast migration and invasion in the 2nd trimester of pregnancy, resulting in the development of preeclampsia.

Project description:The role of long noncoding RNA (LncRNA) in numerous diseases was well-established. However, its pathological significance in preeclampsia (PE) remains obscure. In this investigation, a novel low-expression LncRNA (LncRNA-LPE) was identified in PE, and the regulatory mechanism in the progression of PE was examined. We found that LncRNA-LPE was downregulated in PE compared with normal plancenta, suggesting its potential relation with the development of PE. Modulation of LncRNA-LPE expression in trophoblast cells revealed its capacity to influence the migration and invasion abilities. To identify proteins that interact with LncRNA-LPE, RNA pull-down assays, mass spectrometry, and RNA immunoprecipitation (RIP) assays were employed. And we found LncRNA-LPE could interact with the RNA-binding protein T-complex protein 1 (TCP1), which might subsequently influence the PI3K/AKT signaling pathway and lead to autophagy changing. Our results demonstrated that LncRNA-LPE facilitates the development of preeclampsia by binding to TCP1 and activating PI3K/AKT signaling pathway, indicating its potential as a morbidity marker and therapeutic target for preeclampsia.
