Project description:Long term use of titanium implants generates wearing particles off. Titanium nanoparticles (TiNP) favor a pro-inflammatory macrophage polarization (M1) and lower the tolerogenic activation (M2). Our previous Affymetrix data showed that TiNP upregulate GDF15 and downregulate STAB1 expression in macrophages. GDF15 is a cytokine that regulates energy expenditure and fibrosis, and is endocytosed by the scavenger receptor Stabilin-1. Our study aim was to investigate the effect of TiNP on GDF15 and STAB1 expression, on GDF15 secretion and on endocytosis efficiency in macrophages. We used the model system of primary human macrophages derived from CD14+ monocytes. After 6 days of incubation with TiNP, we quantified GDF15 and STAB1 expression, and GDF15 secretion in INF-γ stimulated (M1) and IL-4 stimulated (M2) macrophages using RT-PCR and ELISA, respectively. Flow cytometry assessed the effect of TiNP on endocytosis based on the uptake of fluorescently labelled acLDL, a known ligand of stabilin-1, and MS-1, an antibody directed against the extracellular portion of stabilin-1 in M1 and M2. Our results revealed a strong up-modulating effect of TiNP on GDF15 expression and secretion in all subtypes of macrophages. In contrast, stabilin-1 expression was significantly suppressed by TiNP. acLDL and MS-1 uptake by macrophages was also significantly suppressed under the treatment with TiNP. Our findings highlight a consistent ex vivo stimulatory effect of TiNP on GDF15 production and inhibitory effect on STAB1 expression and Stabilin-1-mediated endocytosis. Our results suggest an important implication of TiNP on macrophage healing functions.

Project description:T-cell receptor (TCR) signaling is essential for the function of T cells and negatively regulated by the E3 ubiquitin-protein ligases CBL and CBLB. Here we combine mouse genetics and affinity purification coupled to quantitative mass spectrometry to monitor the dynamics of the CBL and CBLB signaling complexes that assemble in normal T cells over 600 seconds of TCR stimulation. We identify many previously known CBL and CBLB interacting partners, as well as a majority of proteins that have not yet been implicated in those signaling complexes. We exploit correlations in protein association with CBL and CBLB as a function of time of TCR stimulation for predicting the occurrence of direct physical association between them. The dataset is divided in two distinct subsets corresponding to the Cbl and to the Cblb interactomes. Project description:In this study, we combined mouse genetics and quantitative proteomics to characterize in a comprehensive manner the VAV1 interactome. A line of knock-in mouse expressing endogenous VAV1 molecules with a genetic tag permitting affinity purification was generated and combined with affinity purification and mass spectrometry (AP-MS) to analyze the VAV1 signaling complex that assembles in primary CD4+ T cells over 300 s of activation. This allowed us to describe in a time-resolved manner the interactome of VAV1 in primary CD4+ T cells and to identify 44 previously unknown bindings partners of VAV1. The dataset contains mass spectrometry results from the analysis of AP-MS purifications (based on affinity purification on Streptactin beads of One-Strep-tagged proteins) starting from CD4+ T cells which were either non stimulated or stimulated with pervanadate for different time lengths. Project description:Loss of the seed-specific WRKY transcription factor WRKY43 confers enhanced tolerance towards high salt, high osmolarity and low temperature with respect to seed germination. wrky43 loss of function lines display increased inhibition of seed germination in response to exogenous ABA, while WRKY43 overexpression lines are more tolerant towards exogenous ABA. The opposing effect of the wrky43 mutant on salt and ABA tolerance is reminiscent of fatty acid desaturase mutants. Loss of WRKY43 enhances polyunsaturated fatty acid content, particularly 18:2 and 18:3 in TAGs and Phospholipids. Gene chip arrays show that ABA-induced regulation of FUSCA3, ZAT10 and seed storage proteins are absent in the wrky43 mutant. Promoter-Luciferase studies confirm direct regulation of ZAT10 by WRKY43 and suggest indirect regulation of FUS3 and SSPs. In summary WRKY43 acts as a positive regulator of ABA-dependent gene regulation and of fatty acid desaturation that finally results in enhanced tolerance to abiotic stress. 2 biological replicates of Arabidopsis thaliana Ler-0 wildtype and wrky43 muatnt seeds were compared after incubation in liquid 0.5 MS media with 2 µM ABA for 4 days

Project description:MES cells were treated with vehicle or 250 nM extracellular alpha-synuclein and plasma membrane proteins were labeled with biotin, followed by affinity purification. The biotinylated enriched membranous proteins were incubated with an alpha-synuclein peptide array membrane, washed extensively with TBS-T, followed by incubation with avidin-HRP. The positive spots were excised and the bound proteins were eluted. The elutions from consecutive spots were combined before trypsin digestion and analysis of peptides by LC-MS/MS.

Project description:NAADP interactome data obtained by affinity precipitation with immobilized NAADP from HEK293 cells expressing exogenous TPC1-eGFP-FLAG. Heavily labeled proteins were pulled-down by immobilized NAADP and lightly labeled proteins were pulled-down by empty beads (no NAADP). Proteins were eluted from beads by NAADP first (raw data file names contain NAADP) and then by SDS (file names contain SDS). The suffix 1-3 for file names indicates order of excised gel bands from top.
