The deletion of a major facilitator superfamily gene VdMFS2 results in enhanced pathogenicity of Verticillium dahliae to cotton

ABSTRACT: The knockout of VdMFS2 can inhibit the proliferation of mycelia and the germination of spores of V. dahliae, while enhancing its pathogenicity to cotton. This study provides new insights into the role of MFS transporters in V. dahliae and offers more precise clues for elucidating the impact of MFS transporters on this fungus.

RNA-Sequencing, Physiological and RNAi Analyses Provide Insights into the Response Mechanism of the ABC-Mediated Resistance to Verticillium Dahliae Infection In cotton

Project description:Verticillium wilt which is caused by Verticillium dahliae causes massive annual losses of cotton yield. Control by conventional mechanisms is not possible due to wide host range and longevity of dormant fungi in the soil in case of absence of a suitable host. Plants have developed various mechanisms to boost their immunity against various diseases, and one of which is through the induction of various genes. In this research work, carried out of RNA sequencing and identified the members of the ABC genes are critical in enhancing resistance to V. dahliae infection. A total of 166 ABC genes were identified in Gossypium raimondii with varying physiochemical properties. A novel ABC gene, Gorai.007G244600 was found to be highly upregulated, its homolog in the tetraploid cotton Gh_D11G3432, was then silenced through virus induced gene silencing (VIGS) in tetraploid cotton, the mutant cotton seedlings that have the ability to tolerate V. dahliae infection were significantly reduced. Evaluation of oxidant, hydrogen peroxide (H2O2) and malondialdehyde (MDA) were found to have increased levels in the leaves of the mutant compared to the wild types. In addition, antioxidant enzymes, peroxidase (POD), catalase (CAT) and superoxide dismutase (SOD) concentration levels were significantly reduced in the mutant cotton compared to the wild types. Moreover, expression levels of the biotic stress genes, cotton polyamine oxidase (GhPAO), cotton ribosomal protein L18(GhRPL18) and cotton polygalacturonase-inhibiting protein-1 (GhPGIP1) were all downregulated in the mutant but highly upregulated in the wild cotton tissues. The outcome of this research has shown that ABC genes could be playing an important role in enhancing immunity of cotton to V. dahliae infection and can be explored in developing more resilient cotton genotypes with improved resistance to V. dahliae infection.

Transcriptome sequencing of Verticillium dahliae with different virulence

Project description:The V. dahliae could cause host obvious disease symptoms. Moreover, it was difficult to control, because of the rapidly genetic variation. Due to the rapidly genetic variation and complexity of the genetic background, the mechanism of pathogenicity differentiation of V. dahliae has still been unknown. To solve the problem, we isolated 24 strains of V. dahliae from a farmland of Shihezi city, Xinjiang province. Through ITS sequences and VCGs assay, the strains of SHZ-4, SHZ-5, and SHZ-9 had closer kinship. Moreover, the large difference was presented on the pathogenicity of SHZ-4, SHZ-5, and SHZ-9. Furthermore, a number of differentially expressed genes was identified through comparative transcriptome sequencing method and the genes were mainly centered on CWDEs and transport relative proteins. It was well understand that the CWDEs determined the source of nutrients of V. dahliae and the organic substance of host cell degraded by the CWDEs would induce the hyperosmosis of intercellular liquid. According to these, we proposed a hypothesis that the hyperosmosis induced the dehydration and death of host cell and contributed to absorb the nutrition for V. dahliae. The hyperosmosis promoting the nutrients scramble indicated that the V. dahliae could resist the hyperosmosis and the pathogenicity was positive associating with the tolerance. With regards to this, the sugar-induced-hyperosmotic resistance of the three V. dahliae isolates and cotton leaf tissue were analyzed through changing the sugar concentration. The result was entirely consistent with our hypothesis. It was firstly found that the pathogenicity of V. dahliae was relative to the resistance of sugar-induced-hyperosmosis, which could provide a new idea for cultivating a new varieties to resistant to the V. dahliae.

Transcriptome and metabolome profiling reveals key pathways and metabolites involved in defense response of upland cotton against Verticillium dahliae

Project description:Cotton is the main source of natural fiber in the textile industry, making it one of the most economically important fiber crops in the world. Verticillium wilt, caused by the pathogenic fungus Verticillium dahlia, is one of the most damaging biotic factors limiting cotton production. Mechanistic details of cotton defense responses to verticillium wilt remain unclear. In this study, GFP-labeled strain of V. dahlia was used to track colonization in cotton roots, and clear conidial germination could be observed at 48 hours post-inoculation (hpi), marking this as a crucial time point during infection. Transcriptome analysis identified 1,523 and 8,270 differentially expressed genes (DEGs) at 24 hpi and 48 hpi, respectively. Metabolomic screening found 78 differentially accumulated metabolites (DAMs) at 48 hpi. Conjoint analysis indicated that the phenylpropanoid biosynthesis pathway was activated in cotton infected with V. dahliae. The five metabolites in the phenylpropanoid biosynthesis pathway, including caffeic acid, coniferyl alcohol, coniferin, scopoletin and scopolin, could significantly inhibit V. dahlia growth in vitro, implicating their roles in cotton resistance to Verticillium wilt. The findings expand our understanding of molecular mechanisms underlying the pathogen defense response against V. dahlia infection in upland cotton, which may lead to future insights into controlling Verticillium wilt disease.

Transcriptome sequencing of Verticillium dahliae and △VdBre1 mutant

Project description:The soil-borne ascomycete Verticillium dahliae which causes wilt disease in many important crops results a severe reduction in crop yield and quality. During infection, the V.dahliae will secrete a lot of secondary metabolites which act as toxic factors to promote the infection process. However, the mechanism underlying how V.dahliae secondary metabolites regulate cotton infection remains largely unexplored. BRE1 is a highly conserved ubiquitin ligase (E3) enzymes and regulate expression of specific genes related to the production of secondary metabolites. To get an insight into the genes regulated by VdBre1, the comparative transcriptomic analysis was conducted between VdBre1 deletion mutant and the wild type V592. The conidia of wilde type and mutant were collected and total RNA was extracted using fungal RNA kit, sequencing was performed on an BGISEQ-500 platform.

VdGGCT, a GGCT_like super family protein, is required for mycelial growth, conidiation and pathogenicity of Verticillium dahliae

Project description:Given the scarcity of known virulence factors in V. dahliae, which causes severe agricultural losses globally, identification of novel virulence genes is imperative for developing small-molecule inhibitor-based strategies against Verticillium wilt. Although gamma-glutamyl cyclotransferase (GGCT) holds promise as a therapeutic target in cancer, its functions and regulatory mechanisms in plant-pathogenic fungi remain largely elusive. In this study, we demonstrate that deletion of the VdGGCT gene severely attenuates the development and virulence of V. dahliae. Interestingly, despite belonging to the GGCT subfamily of the GGCT superfamily, VdGGCT exhibits no detectable GGCT activity towards GSH, γ-Glu-Met, or γ-Glu-Cys. Further Transcriptomic and metabolomic analyses revealed that VdGGCT is critically involved in regulating the transcription of genes and the abundance of metabolites closely associated with pathogenicity, particularly those implicated in amino acid metabolism, glutathione metabolism, and the biosynthesis of secondary metabolites. Collectively, our findings revealed the importance of VdGGCT in the development and virulence of V. dahliae, thereby identifying a novel target for the design of innovative fungicides.

The transcriptional landscape of Nicotiana benthamiana and Verticillium dahliae interaction

The plant immunity can be activated by Vd-derived patterns while V. dahliae secreted effectors. We report here the RNAseq analyses of samples from N. benthamiana infected by V. dahliae strains 592 and 171 at different time points. The data showed dynamic expression patterns of genes not only from plant defense signaling but also plant developmental signaling.
