Triple Negative Breast Cancer Metastasis Marker

ABSTRACT: Triple-Negative Breast Cancer (TNBC) is known for its high recurrence and metastasis rates, posing treatment difficulties and a poor prognosis post-metastasis. Exosomes, linked to metastasis and immune evasion, could be early biomarkers for tumor spread. This study seeks to identify and preliminarily validate serum markers for diagnosing TNBC metastasis using high-throughput sequencing.

Downregulation of antigen presentation-associated pathway proteins in poor outcome triple-negative breast cancer patient tumors

Project description:Triple-negative breast cancer (TNBC) is a heterogeneous subtype with varying disease outcomes. Tumor infiltrating lymphocytes (TILs) are frequent in TNBC and have been shown to correlate with outcome, suggesting an immunogenic component in this subtype. However, other factors, intrinsic to the cancer cells, may also influence outcome. To identify proteins and molecular pathways associated with recurrence in TNBC, 34 formalin-fixed paraffin-embedded (FFPE) primary TNBC tumors were investigated by global proteomic profiling using TMT-HILIC-LC-MS/MS. Approximately half of the patients were lymph node-negative and remained free of local or distant metastasis within 10 years follow-up, while the other half developed distant metastasis. Proteomic profiling identified >4000 proteins, of which 63 exhibited altered expression in primary tumors of recurrence versus recurrence-free patients. Importantly, down-regulation of proteins in the major histocompatibility complex (MHC) class I antigen presentation pathways were enriched, including TAP1, TAP2, CALR, HLA-A, ERAP1 and TAPBP, and were associated with significantly shorter recurrence-free and overall survival. In addition, proteins involved in cancer cell proliferation and growth, including GBP1, RAD23B, WARS and STAT1, also exhibited altered expression in primary tumors of recurrence versus recurrence-free patients. The association between the antigen-presentation pathway and outcome were validated in a second sample set of 10 primary TNBC tumors and corresponding metastases using proteomics and in a large public gene expression database of 249 TNBC and 580 basal-like breast cancer cases. Our study demonstrates that down-regulation of antigen presentation is a key mechanism for TNBC cells to avoid immune surveillance, allowing continued growth and spread.

Gene expression profile at single cell level of bone metastases from triple-negative breast cancer in mice.

Project description:Triple-negative breast cancer (TNBC) bone metastasis has been lacking targeted therapy options. Our research has identified the androgen receptor (AR) as a potential driver of TNBC bone metastasis. To investigate the role of AR in TNBC bone metastasis, we utilized the iliac artery injection method to establish mouse models of bone metastasis. Subsequently, we administered PBS/DHT injections to explore the effects of AR on TNBC bone metastasis in mice.

Sphingosine-kinase-1 pathway offers positive therapeutic targets in triple-negative breast cancer for effective inhibition of metastasis

Project description:We identified sphingosine kinase 1 (SPHK1) as a top candidate. SPHK1-overexpression in human TNBC cell-line promotes spontaneous metastasis to lungs in nude mice. Moreover, genetic knockdown of SPHK1 in TNBC patient-derived xenograft (PDX) cells and TNBC cell-line decreases spontaneous metastasis to lungs in nude mice. SPHK1 regulates the FSCN1 gene expression in order to drive metastasis. Mechanistically, SPHK1 transcriptionally upregulates metastasis-promoting FSCN1 gene expression via NFκB activation to promote metastasis. SPHK1/NFκB/FSCN1 signaling pathway activation is associated with distance metastasis and poor clinical outcome in TNBC patients.

Connective Tissue Growth Factor, a Potential Prognostic Marker of Triple Negative Breast Cancer treatment

Project description:Triple negative breast cancer (TNBC) patient with high level of connective tissue growth factor (CTGF) show poor survival and prognosis. Identification of novel target contributes to the development of TNBC treatment. Kahweol, a coffee diterpene molecule, is a natural compound with pharmacological activities such as anti-inflammatory, anti-angiogenetic, and apoptotic effect in several cancer cells. In this study, we demonstrated the involvement of CTGF expression level in TNBC treatment. Experiments were performed on MDA-MB 231, a TNBC cell line. Firstly, functional annotation and gene set enrichment analysis found that the CTGF involves in migratory pathways in TNBC treatment. We also investigated that kahweol regulates CTGF expression and functions as antitumor compound resulting in inhibition of cell migration and colony formation in TNBC. Next, CTGF knockdown synergically suppressed cell motility with kahweol. Migratory ability and colony conformation were also performed in vitro to confirm the results from functional annotation assay. Overall, our results suggest that CTGF is a potential target as a prognostic marker predicting the prognosis of TNBC treatment, and kahweol is a potential antitumor compound to regulate CTGF expression for TNBC.

SOX2-OT induced by PAI-1 promotes triple-negative breast cancer cells metastasis

Our published study showed that PAI-1 could induce the migration and metastasis of TNBC cells. However, the underlying mechanism by which PAI-1 regulates TNBC metastasis has not been addressed. Using microarray analysis of lncRNA expression profiles, we identified a lncRNA SOX2-OT, which is by induced by PAI-1 and could function as an oncogenic lncRNA in TNBC.

Decreased expression of key tumour suppressor microRNAs is associated with lymph node metastases in triple negative breast cancer

Project description:Breast cancer is the most common malignancy that develops in women, responsible for the highest cancer-associated death rates. Triple negative breast cancers (TNBC) represent an important subtype that have an aggressive clinical phenotype, are associated with a higher likelihood of metastasis and are not responsive to current targeted therapies. miRNAs have emerged as an attractive candidate for molecular biomarkers and treatment targets in breast cancer, but their role in the progression of TNBC remains largely unexplored. This study has investigated miRNA expression profiles in 31 primary TNBC cases and in 13 lymph node metastases compared with 23 matched normal breast tissues to determine miRNAs associated with the initiation of this disease subtype and those associated with its metastasis. 71 miRNAs were differentially expressed in TNBC, the majority of which have previously been associated with breast cancer, including members of the miR-200 family and the miR-17-92 oncogenic cluster, suggesting that miRNAs involved in the initiation of TNBC are not subtype specific. However, the repertoire of miRNAs expressed in lymph node negative and lymph node positive TNBCs were largely distinct from one another. RAGE Inhibitor TTP488 (Azeliragon) Reduces Metastasis in Triple-Negative Breast Cancer

Project description:We tested the preclinical efficacy of two small molecule RAGE inhibitors (TTP488 (Azeliragon) and FPS-ZM1) against TNBC metastasis in the orthotopic xenograft MDA-MB-231/4175 - NSG model. Both TTP488 and FPS-ZM1 impaired TNBC metastasis in this lung metastatic breast cancer model, with TTP488 affecting metastasis to a greater degree than FPS-ZM1. Transcriptomic analysis of the primary tumors revealed that TTP488 and FPS-ZM1 displayed high concordance in gene expression changes affecting metastatic pathways.

Protein Tyrosine Phosphatase UBASH3B is Overexpressed in Triple-Negative Breast Cancer and Promotes Invasion and Metastasis

Project description:Efforts to improve the clinical outcome of highly aggressive triplenegative breast cancer (TNBC) have been hindered by the lack of effective targeted therapies. Hence, it is important to identify the specific gene targets/pathways driving the invasive phenotype to develop more effective therapeutics. Here we show that UBASH3B (ubiquitin associated and SH3 domain containing B), a protein tyrosine phosphatase, is overexpressed in TNBC, where it supports malignant growth, invasion and metastasis in large through modulating EGFR. We also show that UBASH3B is a functional target of anti-invasive miR-200a that is downregulated in TNBC. Importantly, the oncogenic potential of UBASH3B is dependent on its tyrosine phosphatase activity, which targets CBL ubiquitin ligase for dephosphorylation and inactivation, leading to EGFR upregulation. Thus, UBASH3B may function as a crucial node in bridging multiple invasion-promoting pathways, thus providing a potential new therapeutic target for TNBC. Marizomib eradicates primary and metastatic triple-negative breast cancer via proteasome and oxidative phosphorylation inhibition

Project description:Lacking effective targeted therapies, triple-negative breast cancer (TNBCs) is highly aggressive, metastatic, and clinically challenging breast cancer subtype with worst prognosis. Despite survival dependency on the proteasome pathway genes, the FDA-approved proteasome inhibitors induced minimal clinical response in TNBC patients due to weaker proteasome inhibition. Here, we show that a novel proteasome inhibitor Marizomib (Mzb), inhibited multiple proteasome catalytic activities and induced better anti-tumor response in TNBC cell line and patient-derived xenografts alone and in combination with a standard-of-care chemotherapy, doxorubicin. Mechanistically, Mzb inhibits oxidative phosphorylation (OXPHOS) via PGC-1α suppression in conjunction with proteasome inhibition in TNBC cells. Development of metastatic disease, especially brain metastasis, remains a reason for a greater mortality rate amongst TNBC patients. Mzb reduces lung and brain metastasis in vivo by reducing circulating tumor cells and the expression of multiple epithelial-to-mesenchymal genes. We also demonstrate that Mzb-induced OXPHOS inhibition upregulates glycolysis to fulfill the metabolic demand of TNBC cells and hence, combined inhibition of glycolysis with Mzb leads to a synergistic anti-cancer activity in vivo. Collectively, our data provide a strong rationale for the clinical evaluation of Mzb in primary and metastatic TNBC patients.
