The viability of storing platelets can be maintained in plasma without cryoprotectants at -80°C

ABSTRACT: To prolong the retention period of platelets, dimethyl sulfoxide (DMSO) was employed for platelet storage at -80℃ to prevent hypothermic lesions. Accordingly, we established an experimental group with no 5% DMSO supplementation and a control group supplemented with 5% DMSO for comparative analysis. All samples were cryopreserved at -80 °C for 28 days. Each group comprised 6 independent biological replicates derived from distinct donors, resulting in a total of 12 samples subjected to untargeted metabolomics analysis.

Functionally-relevant splicing of resident pre-mRNAs in anucleate platelets upon activation by physiological stimuli

Project description:Platelet activation is the key event triggering thrombus formation in physiological and pathological conditions, such as acute coronary syndromes. Current therapies using antiaggregants still fail to prevent thrombotic coronary events in a significant number of patients, indicating that the mechanisms modulating platelet response during activation need to be clarified. The evidence that platelets are capable of de novo protein synthesis in response to stimuli raised the issue of how the activity of megakaryocyte-derived mRNAs is regulated in these anucleate cell fragments. We applied a combined multi-omics approach to investigate this phenomenon in platelets from healthy donors activated in vitro with Collagen or Thrombin Receptor Activating Peptide. Combining HiRIEF LC-MS to transcriptome analysis by RNA-Seq allowed platelet proteome characterization at deep coverage, revealing a significant effect of either stimulus on proteome composition. In silico intron retention analysis was then applied to search for splicing events induced by platelet activation, coupled to unbiased proteogenomics, to correlate intron retention in resting platelets to intron removal by RNA splicing during activation. This allowed identification of a set of transcripts, specifically involved in platelet shape changes, showing reduced intron retention and high peptide representation at exon-exon junctions in activated vs resting platelets. These results indicate that RNA splicing events takes place in platelets during activation and that pre-mRNA maturation of specific transcripts is part of the activation cascade and could therefore provide novel molecular markers of platelet activation status in acute coronary syndromes and other pathological conditions.

In the present study, GeneChips were performed in human platelets from three normal donors infected with the EBV-containing supernatant of the B95.8 marmoset cell line in vitro.

Immunological role of platelets in Theiler’s virus-induced inflammatory demyelinating disease and myocarditis

Project description:Platelets have been immunologically associated with viral and immune-mediated diseases such as Severe acute respiratory syndrome coronavirus 2 (SARS‑CoV‑2) and Experimental autoim-mune encephalomyelitis (EAE). However, the role of platelets in Theiler’s murine encephalo-myelitis virus (TMEV)-induced myocarditis and inflammatory demyelination disease (IDD) is still unknown. We determine whether platelets were associated with TMEV infection and whether glycoprotein Ib alpha chain (GPIbα/CD42b) antibody injection could affect the pathogenicity of TMEV-induced myocarditis and TMEV-IDD. We conducted a mechanistic experiment by injecting TMEV-infected mice with the anti-GPIbα antibodies 0 and 5 days p.i. (“Early” group) or 18 and 22 days p.i. (“Late” group). Using in silico and in vivo approaches, we found platelets and platelet-related genes were associated with TMEV infection. Platelet depletion in Early and Late mice reduced fibrotic area in the heart, inflammatory demyelinating lesions with lower viral persistence in the CNS and altered antiviral antibody responses. Thus, platelets contribute to the pathogenesis of TMEV infection by promoting fibrosis and CNS inflammation. Platelet-depletion treatment might open new therapeutic approaches for TMEV-induced myocarditis and TMEV-IDD.

Next generation sequencing of blood platelets and PBMCs in early and late stage of melanoma in mice

Project description:We report the application of next generation sequencing of blood platelets in an early-early melanoma mice model for screening early cancer biomarkers. By inoculating C57BL/6 mice with different numbers of B16F10 cells, we establish an early-early melanoma model. We find that mice inoculated with lower number of B16F10 cells develop tumors at a later time. Therefore the blood samples collected at day 14 post-injection before the formation of visible tumors are early-early cancer samples. The mRNA profiles of platelets from early-early group are distinct from those of control group and large tumor group. However, the mRNA profiles of PBMCs from early-early group are not so distinct from those of control group and large tumor group. This study provides a proof of concept strategy for future cancer biomarker screening and cancer immunology research.
