ABSTRACT: Quantitative label-free proteomic analysis investigated the global proteomic response of S. aureus YUSA145 treated with 1/2 × MIC MLS0315771 during the exponential growth phase. The results were obtained after performing principal component analysis (PCA) on the raw data and excluding two replicates that showed substantial variation.

Staphylococcus aureus delays wound repair by harnessing the agr quorum sensing system to target keratinocyte lipid metabolism [bulkRNA-seq]

Project description:NHEKs wounded by scratch and incubated with CM for 24 hours were evaluated by bulk RNAseq. Principal component analysis of these data revealed wounded NHEKs exposed to S. hominis products clustered close to control keratinocytes, suggesting these groups have similar transcriptomes, while the presence of S. aureus products caused variation shift in the transcriptome to result in a distinct cluster away from control. This was supported by a heatmap of differential gene expression of S. aureus and S. hominis CM treatment showing broad S. aureus-mediated suppression of genes induced by S. hominis. Conversely, upregulation of many genes by S. aureus were downregulated by S. hominis.

Global transcriptome profile of S aureus treated with celecoxib and ampicillin in combination

Project description:We have previously shown that celecoxib, a selective COX-2 inhibitor, in combination with antibiotic sensitizes S aureus to antibiotic thereby decreasing the MIC of antibiotic. The present study is aimed at understanding the molecular mechanism of action of celecoxib on S aureus growth by microarray analysis. The results were analysed and the data clearly shows global change in the expression levels of S aureus genes in presence of celecoxib. Many virulence genes, essential genes and nonessential genes are down-regulated in presence of celecoxib in combination with ampicillin when compared to drug treatment alone.

Gene expression profile analysis of Staphylococcus aureus treated with nisin

Project description:Staphylococcus aureus (S. aureus) has already to be one of the most commonly identified bacteria that cause food poisoning. S. aureus colonization in humans can cause serious infections, toxinoses and life threatening diseases. The bacteriocin nisin has been extensively used as potential natural preservative in the food industry, but the overall transcriptional response mechanisms of S. aureus to nisin are still poorly understood. To detect the possible molecular mechanism of nisin against S. aureus, Affymetrix GeneChips were used to determine the global comparative transcription of S. aureus cells triggered by treatment with sub-inhibitory concentrations of nisin. Staphylococcus aureus planktonic cells were exposed for 60 minutes to nisin at concentration of 4 M-NM-<g/ml (1/2M-CM-^W MIC). 2 samples including 2 control samples are analyzed.

Staphylococcus aureus gene expression in a rat model of infective endocarditis

Project description:S. aureus has a propensity to cause endocarditis; diabetes mellitus is a frequent underlying comorbitity in patents with S. aureus endocarditis. S. aureus Affymetrix GeneChips were used to compare S. aureus expression properties in cardiac vegatations isolated from diabetic and nondiabetic rats. S. aureus Affymetrix GeneChips were also used to compare the S. aureus expression properties of cardiac vegatations (both diabetic and nondiabetic) in comparsions to planktonic cells. Few differences were observed between the expression properties of S. aureus harvested from diabetic vs. nondiabetic cardiac vegatations. Significant differences were observed between the expression properties of S. aureus harvested from cardiac vegetations in comparison to exponential and/or stationary phase planktonically grown cells.
