Project description:nevaeh: The PARP inhibitor olaparib is widely used in cancer but is underused in treating lung adenocarcinoma (LUAD). Our initial analysis revealed that RIG-I, which is typically found in the cytoplasm and activates innate immunity, is linked to LUAD outcome. Here, we discovered that RIG-I undergoes lactylation, with the acetyltransferase PCAF serving as the lactylation writer. Treatment with the lactate transporter inhibitor syrosingopine prevents the efflux of lactic acid from cancer cells, increasing intracellular lactic acid, promoting RIG-I lactylation, and enhancing its interaction with the nuclear transport protein importin 8. The augmented interaction facilitates the nuclear translocation of RIG-I, diminishing PARP1 activity and enhancing the therapeutic effects when combined with olaparib. This synergy disrupts PARP1-mediated DNA repair mechanisms, increasing drug sensitivity and inhibiting tumor growth. The combination of olaparib and syrosingopine demonstrates better therapeutic efficacy than olaparib monotherapy in LUAD, providing a promising strategy for future LUAD treatments. nevaeh: HEK293T cells were transfected with Flag-RIG-I for 48 h, lysed in NP-40 buffer, and sonicated using a JY92-II sonicator. The supernatant was first incubated overnight with an anti-Flag conjugated with beads (Sigma, A2220) after being denatured. Following centrifugation at 3,000 rpm (4°C for 5 min), the supernatants were completely removed, and 60 μL of Flag–peptide was added to competitively bind to the beads. RIG-I protein was released, precipitated using Trichloroacetic acid (TCA), and subjected to mass spectrometry analysis for lactylation.

Project description:A new global post-translational modification (PTM) discovery strategy, G-PTM-D, is described. A proteomics database containing UniProt-curated PTM information is supplemented with potential new modification types and sites discovered from a first-round search of mass spectrometry data with ultrawide precursor mass tolerance. A second-round search employing the supplemented database conducted with standard narrow mass tolerances yields deep coverage and a rich variety of peptide modifications with high confidence in complex unenriched samples. The G-PTM-D strategy represents a major advance to the previously reported G-PTM strategy and provides a powerful new capability to the proteomics research community.

Project description:CSR-1 is an essential Argonaute protein that binds to a subclass of 22G-RNAs targeting most germline-expressed genes. Here we show that the two isoforms of CSR-1 have distinct expression patterns; CSR-1B is ubiquitously expressed throughout the germline and during all stages of development while CSR-1A expression is restricted to germ cells undergoing spermatogenesis. Furthermore, CSR-1A associates preferentially with 22G-RNAs mapping to spermatogenesis-specific genes whereas CSR-1B-bound small RNAs map predominantly to oogenesis-specific genes. Interestingly, the exon unique to CSR-1A contains multiple dimethylarginine modifications, which are necessary for the preferential binding of CSR-1A to spermatogenesis-specific 22G-RNAs. Thus, we have discovered a regulatory mechanism for C. elegans Argonaute proteins that allows for specificity of small RNA binding between similar Argonaute proteins with overlapping temporal and spatial localization.

Project description:The implementations of targeted molecular therapies and immunotherapy in melanoma vastly improved the therapeutic outcome in patients with limited efficacy of surgical intervention. Nevertheless, a large fraction of melanoma patients still remains refractory or acquires resistance to these new forms of treatment, illustrating a need for improvement. Here we report that the clinically relevant combination of MAP kinase inhibitors Dabrafenib and Trametinib synergizes with RIG-I agonist-induced immunotherapy to kill BRAF-mutated human and mouse melanoma cells. Kinase inhibition did not compromise the agonist-induced innate immune response of the RIG-I pathway in host immune cells. In a melanoma transplantation mouse model, the triple therapy outperformed the individual therapies. Our study suggests that targeted activation of RIG-I with its synthetic ligand 3pRNA could vastly improve tumor control in a substantial fraction of melanoma patients receiving MAP kinase inhibitors.

Project description:Many of the best-studied actin regulatory proteins use non-covalent means to modulate the properties of actin. Yet, actin is also susceptible to covalent modifications of its amino acids. Recent work is increasingly revealing that actin processing and its covalent modifications regulate important cellular events. In addition, numerous pathogens express enzymes that specifically use actin as a substrate to regulate their hosts' cells. Actin post-translational alterations have been linked to different normal and disease processes and the effects associated with metabolic and environmental stressors. Herein, we highlight specific co-translational and post-translational modifications of actin and discuss the current understanding of the role that these modifications play in regulating actin.
