A core pattern-induced transcribed enhancer regulates the immune response to fungi in Arabidopsis by regulating the synthesis of camalexin

Project description:Effective plant defense against pathogens relies on highly coordinated regulation of immune gene expression. Enhancers, as cis-regulatory elements, are indispensable determinants of the dynamic gene regulation, but the molecular functions in plant immunity are not well understood. In this study, we identified a novel enhancer, CORE PATTERN-INDUCED ENHANCER 35 (CPIE35), is rapidly activated upon pathogenic elicitation and negatively regulates camalexin synthesis and anti-fungal resistance through modulating WRKY15 expression. During immune activation, CPIE35 activates the transcription of WRKY15 by forming chromatin loops with the promoter of WRKY15 in a WRKY18/40/60-, WRKY33-, and MYC2-dependent manner. WRKY15 directly binds to the promoters of GSTU4 and PAD3 and suppresses their expression, leading to the reduced camalexin synthesis and resistance. Interestingly, the CPIE35 region is evolutionally conserved among distantly related dicots, and the CPIE35-WRKY15 module exerts similar functions in Brassica napus to negatively regulates antifungal resistance. Our work reveals the “TFs-enhancer-promoter” regulatory mechanism in maintenance of immune hemostasis and highlights the importance and conservation of enhancers in fine-tuning immune gene expression in plants.

Effect of Plant and Animal Proteins on Biomarkers of Colorectal Cancer and Type 2 Diabetes in Healthy Adults (ScenoProt)

Project description:The aim of this study is to investigate the effects of dietary plant and animal proteins on gut metabolism and markers for colorectal cancer as well as blood protein metabolites and markers for type 2 diabetes in healthy adults. The study participants will be stratified into three groups with different protein composition in diets: 1) animal 70%/plant 30%; 2) animal 50%/plant 50% and 3) animal 30%/plant 70%. The participants will get part of their diet as ready foods or raw material to promote their compliance. The participants will also get personal advice for their diets. Blood, stool and urine samples will be collected in the beginning and in the end of the 12 week intervention, as well as phenotype measures like BMI, blood pressure and body composition. The participants will also fill food diary before and in the end of the intervention.

Global transcriptome of the fungal pathogen Sclerotinia sclerotiorum (strain 1980) during the colonization of 23 Accessions of Arabidopsis thaliana

Project description:Quantitative disease resistance (QDR) is an almost universal and often broad-spectrum process in plants, serving to limit the damage caused by pathogen infections. It represents the primary form of plant immunity that reduces disease symptoms induced by numerous pathogens actively killing host cells during infection, including the necrotrophic pathogen Sclerotinia sclerotiorum. Investigating the evolutionary origins of QDR against necrotrophic fungi is crucial for comprehending how plant resistance evolves. To explore the diversity of local responses to S. sclerotiorum within a plant species level, we conducted a comprehensive analysis of the entire transcriptomes from 23 accessions of Arabidopsis thaliana, mainly distributed across Europe. More than half on the pan-transcriptome displayed local responses toS. sclerotiorum, including similar transcriptome patterns. Notably, core S. sclerotiorum-responsive genes exhibited a clear gene age pattern, dominated by older genes forming protein-protein networks that continuously acquiring new hubs. Comparative transcriptome analyses revealed QDR is associated with quantitative expression variations specific to accession subsets. By comparison of promoter sequences, we have shown evidence that accession subsets independently evolved and acquired specific cis-regulatory elements, confering Sclerotinia resistance. This scenario suggests multiple exaptation trajectories of novel QDR genes through species-level cis-regulation. This study sheds light on the regulation of QDR-associated genes within a species, contributing to our understanding of the molecular mechanisms of plant fungal resistance.

Project description:To determine the mechanism of abnormal activity of RA FLS, we have done some research and found out that lncRNA HASML and protein HuR have some effects on migration and invasion of RA FLS. To more deeply explore how HASML and HuR regulate the functions of RA FLS, RNA sequencing analysis was used to evaluate the transcriptome of siHASML and siHuR treatment and negative control (NC) group. A P-value less than 0.05 was considered significant.

A role of PSK-CPK28-GS2 signalling in optimizing growth and defence in tomato

Project description:Phytosulfokine (PSK) is a plant pentapeptide hormone that fulfills a wide range of functions. Although it has been frequently reported to function in the inverse regulation of growth and defence in response to (hemi)biotrophic pathogens, the mechanism involved remains largely unknown. Using tomato (Solanum lycopersicum) and Pseudomonas syringae pv. tomato (Pst) DC3000 pathogen as a system, we present compelling evidence that the tomato PSK receptor PSKR1 interacts with the calcium-dependent protein kinase CPK28, which phosphorylates the key enzyme of nitrogen assimilation glutamine synthetase GS2 at two sites (S334 and S360 sites). These post translational modifications uncouple the two PSK-induced effects on defence responses and on growth regulation, respectively. The discovery of these sites will inform breeding strategies designed to optimize the growth-defence balance. The concept that such peptide hormones can regulate plant growth and resistance in a compatible manner is attractive, not least because crop yields and fitness can be optimized simultaneously in global climate-change era.

BUI1, an actin cytoskeleton protein that confers durable and broad-spectrum resistance in rice

Project description:The cytoskeleton functions as a dynamic intracellular structural scaffold, enabling cells to rapidly perceive biotic and abiotic stresses. During plant innate immune responses, the cytoskeleton undergoes actin remodeling. Using the rice-Magnaporthe oryzae pathosystem, we investigated changes in actin organization within host plant cells. Our results demonstrate that pattern-triggered immunity (PTI), induced by pathogen-associated molecular patterns (PAMPs), triggers a rapid but transient increase in actin filament abundance. This process depends on BUI1, a type-II formin protein. Disruption of actin filaments with latrunculin B not only suppresses this increase but also enhances host susceptibility. Furthermore, we show that BUI1 positively regulates broad-spectrum resistance against multiple pathogens by suppressing reactive oxygen species (ROS) scavenging. Together, these findings uncover a key mechanism by which receptor-mediated signaling reorganizes the host cytoskeleton during plant-pathogen interactions.

To explore whether GM-CSF mediates its effects via monocytes, we analyzed effects of GM-CSF on monocytes in vitro and assessed the immunomodulatory potential of GM-CSF-activated monocytes (GMaM). We used microarray technology and functional assays to characterize GMaM in vitro and used a mouse model of colitis to study GMaM functions in vivo.
