Paternal cold exposure enhances brown adipose tissue sensitivity in offspring and ameliorates diet induced obesity

Project description:Research in recent years has focused on environmental effects that control tissue functionality and systemic metabolism. We show here in a retrospective analysis that presence of brown adipocytes and date of birth are correlated and linked to BMI. In a mouse model system, we demonstrate that pre-conception cold exposure leads to improved systemic metabolism and protects from diet induced obesity, through the sperm via the paternal lineage. Using a brown fat ablation model, we show that this effect is mediated by brown adipose tissue. Furthermore, through RNAseq and DNA methylation analysis we identify Adrb3 as a putative signaling mechanism which could mediate the effects of pre-conception cold exposure and could account for the heterogeneity of brown adipose tissue responsiveness. Taken together, our results identify paternal cold exposure as a stimulus which induces a hyper-active state in brown adipose tissue leading to improved adaptation to overnutrition and hypothermia in the offspring.

Cysteine redoxome landscape in mouse brown adipose tissue under acute cold exposure

Project description:Reversible cysteine post-translational modifications serve as a "switch" for protein structure-function dynamics. While reversible cysteine oxidation in uncoupling protein 1 is known to play a role in brown fat thermogenesis, the full cysteine redoxome affected by cold exposure remains unexplored. We established a strategy for comprehensively mapping the cysteine redoxome by pinpointing oxidized and reduced cysteine residues in the brown adipose tissue of mice under room temperature and acute cold exposure. We identified over 8000 labeled cysteine residues under room and cold temperatures. Cold exposure shifted the cysteine redox states toward oxidation. Cold-sensitive reactive cysteine residues were enriched in biological processes and molecular functions associated with thermogenesis pathways. The presence of proximal positively charged and negatively charged amino acids determined the highly reactive and non-reactive cysteine residues, respectively, under cold exposure. Our findings broaden the landscape of cold-sensitive proteome and identify potential therapeutic targets to fine-tune thermogenesis.

Long non-coding RNA expression profiles in cold-induced injury of mouse livers

Project description:Hepatic injury provoked by cold storage is a major problem affecting liver transplantation, as exposure to cold induces apoptosis in hepatic tissues. Long noncoding RNAs (lncRNAs) are increasingly understood to regulate apoptosis, but the contribution of lncRNAs to cold-induced liver injury remains unknown. Using RNA-seq, we determined the differential lncRNA expression profile in mouse livers after cold storage and found that expression of the lncRNA TUG1 was significantly down-regulated. Over-expression of TUG1 attenuated cold-induced apoptosis in mouse hepatocytes and liver sinusoidal endothelial cells LSECs, in part by blocking mitochondrial apoptosis and ER stress pathways. Moreover, TUG1 attenuated apoptosis, inflammation and oxidative stress in vivo in livers subjected to cold storage. Over-expression of TUG1 also improved hepatocyte function and prolonged hepatic graft survival rates in mice. These results suggest that the lncRNA TUG1 exerts a protective effect against cold-induced liver damage by inhibiting apoptosis in mice, and suggests a potential role for TUG1 as a target for the prevention of cold-induced liver damage in liver transplantation.

Transcriptome analysis of brown fat in young or old mice in cold exposure

Project description:Purpose: We investigated the transcriptomic change in brown fat of young and old mice (wild type) through high-throughput RNA-sequencing (RNA-Seq) analysis when the mice were exposed to cold room or room temperatur. Methods: We prepared 10 of young (3 months) mice and 9 of old (24 months) mice, and kept them in cold room (4°c) or room temperature (24°c) for 24 hours. Then, we sacrified mice and extracted RNA from brown fat tissue (BAT) for RNA-seq experiment. Results: BAT of Young mice showed increased carbohydrate metabolism and glycolytic flux during cold exposure Conclusions: The thermogenesis function of BAT is accelerated on cold exposure.
