Ubiquitination site mapping of NICD-interacting proteins in mouse by IP-MS

ABSTRACT: Primary murine γδ T cells were isolated and polarized toward a pro-inflammatory γδT17 phenotype by stimulation with recombinant cytokines IL-1β (20 ng/mL), IL-23 (10 ng/mL), and IL-7 (10 ng/mL) for 72 hours. The Notch intracellular domain (NICD)-associated protein complex was then immunoprecipitated from these activated γδ T cells using an anti-NICD antibody. The immunoprecipitated material was subjected to ubiquitin remnant motif (K-ε-GG) enrichment and analyzed by high-resolution LC-MS/MS on a Q Exactive HF-X mass spectrometer to identify ubiquitination sites on NICD-interacting proteins in the context of γδT17 cell activation.

Project description:Human CD14 positive monocytes were purified from healthy volunteers' blood and were differentiated to immature dendritic cells in vitro by culturing for five days in the presence of interleukin-4 (IL-4 100 ng/ml) and GM-CSF (75 ng/ml). Immature dendritic cells were activated three different ways for 24 hours: 1. Double-stranded DNA poly(dA:dT) (2.5 μg/ml) complexed with LyoVec transfection reagent. 2. LPS (500 ng/ml) 3. Inflammatory cocktail containing 10 ng/ml TNF, 5 ng/ml IL-1β, 20 ng/ml IL-6, 75 ng/ml GM-CSF and 1 μg/ml PGE2. We used SA Biosciences Antigen Presenting and Toll-like Receptor Pathway PCR Arrays to quantitate gene expression of immunologically relevant genes from the immature and the differently activated cells.

Project description:The hPDLSCs were cultured in the tenogenic medium treated with 0.1 ng/ml IL-1β or 10 ng/ml TNF-α for 7 days. Total proteins were isolated from the collected cell samples. Subsequently, the LFQ-based proteomic analysis was conducted for the extracted proteins in each group as established procedures [59]. Differential expression analysis was performed by comparing the expression levels of the identified proteins in the hPDLSCs treated with IL-1β or TNF-α with those in the Ctrl group.

Transcriptome of cytokines-polarized mouse bone marrow-derived macrophages

Project description:To understand the molecular signature of the IL-10/IL-18 polarized macrophages, we performed transcriptome analysis for mouse BMDMs polarized with PBS (Naïve), IFN-γ (classic M1 stimulator, M1, 20ng/ml), IL-4 (alternative M2 stimulator, M2, 20ng/ml), IL-10 (M10, 2000ng/ml), IL-18 (M18, 2000ng/ml) or IL-10 and IL-18 (M1018, 2000ng/ml each). These data suggest that IL-10 and IL-18 cooperatively modulate a set of gene expressions and pathway activities in macrophages and contribute to a distinct polarization state.

Project description:Human monocyte THP-1 cells obtained from ATCC were cultured in RPMI 1640 (Invitrogen, Carlsbad, CA) containing 10% FBS and supplemented with 10 mM Hepes (Gibco BRL). THP-1 was differentiated into macrophages by 24-h incubation with 160 nM phorbol 12-myristate 13-acetate (PMA; Sigma, St. Louis, MO) followed by 24-h incubation in RPMI medium. Macrophages were further polarized to M1 macrophages by incubation with 10 pg/ml of lipopolysaccharide (LPS; Sigma) and 20 ng/ml of interferon (IFN)-γ (R&D Systems, MN) and are referred to as M(LPS+IFN-γ) cells. M2 macrophages were obtained by incubation with 20 ng/ml of interleukin (IL)-4 (R&D Systems) and are referred to as M(IL4) cells. To test the represented polarization marker of PMA differentiated-THP-1 macrophages stimulated with 20 ng ml(-1) IFNγ + 10 pg ml(-1) LPS and 20 ng ml(-1) IL-4, which are known to influence macrophage polarization in vetro into the M1 and M2 state, respectively. We used microarrays to detail the gene expression pools to identify distinct M1 and M2 state during this process.

Project description:The conditioned media from Bifidobacterium infantis (BCM) and Lactobacillus acidophilus (LCM) were reported to promote maturation of innate immune response gene expression, which explained the protective effects of probiotics in clinical necrotizing enterocolitis. We used microarray analysis to investigate the expression of genes involved in regulation of BCM and LCM in IL-1β stimulated immature human enterocytes. The H4 cells (a human nontransformed primary intestinal epithelial cell line) were pretreated with BCM or LCM (15%) for 30 minutes, and then stimulated with or without IL-1β (10 ng/mL) for 4 h. Cell media and IL-1β stimulation were negative and positive control, respectively. RNA was extracted for mcroarray analysis.

Gene expression profiling of IL-18 stimulated, in vitro differentiated mouse mucosal mast cells (MMC)

Project description:The aim of the present study was to clarify the role of IL-18 in mouse mast cell functions. Mouse mucosal mast cells (Balb/c) were differentiated in the presence of IL-3 (5 ng/ml), SCF (40 ng/ml), IL-9 (10 ng/ml) and human TGFbetaI (2 ng/ml) for >4 weeks. Cells were then stimulated by 100 ng/ml IL-18 for 2 hours to monitor the direct effect of IL-18. Treated samples were compared to the untreated ones in a paired experimental setting.
