Quantitative Proteomics of mPFC Tissue in a Vascular Cognitive Impairment Mouse Model

ABSTRACT: Deep quantitative proteomic profiling of bilateral medial prefrontal cortex (mPFC) from control (ctrl), vascular cognitive impairment (VCI, model) and EVs treatment (test) mice (3 biological replicates each). Proteins extracted with RIPA + inhibitors, quantified by BCA, processed via FASP, digested with trypsin, labeled with TMT-16plex, fractionated by high-pH RP, and analyzed on nanoLC-Q Exactive HF-X (DDA, 120k/15k resolution). Raw files searched against Mus musculus UniProt in PD 3.0/Mascot (FDR ≤1%). Differentiation criteria: fold-change >1.2, p<0.05 (Perseus).
