Project description:To investigate the function of Opa1 in the regulation of adult neurogenesis, we created a conditional Opa1-knockout mouse line by cross-breeding Nestin-CreERT2;ROSA26YFP mice with Opa1-Flox mice. Using Tamoxifen, we induced Opa1 knockout on 8-weeks-old Nestin-CreERT2;ROSA26YFP;Opa1-Flox mice. We then sorted YFP-positive cells from the hippocampus using flow cytometry at 10 weeks. RNA was extracted from sorted cells and subjected to next generation sequencing.

Project description:We isolated splenic NK cells and cultured them for 3 days in 100 ng/ml IL-15. Next, we stimulated the cells with plate-bound anti-NK1.1 for 6 hours and sorted YFP+ (IFNg+) and YFP- (IFNg-) populations. We then performed gene expression profiling analysis using data obtained from RNA-seq from unstimulated, YFP+ or YFP- NK cells.

Project description:To identify a common differentially expressed gene signature between metastatic tumors and primary tumors, we utilized both p48-Cre/p53F/+KrasL/+/ROSA-LSL-YFP and SIRT6/p48-Cre/p53F/+KrasL/+/ROSA-LSL-YFP as well as orthotopically injected 4T1-eGFP Balb/c mice to generate spontaneous. metastases We then sorted YFP and GFP positive cells from primary and metastatic sites in mice with evidence of metastatic disease. Comparative gene expression profiling on sorted cells was then performed using RNA-Seq.

Flexible and scalable control of T cell memory by a reversible epigenetic switch (RNA-seq)

Project description:In this study, we find that the Tcf7+ CD8 T cells present following acute challenge can form either from cells that maintain Tcf7 expression throughout activation or from cells that silence Tcf7 and reactivate it after stimulation removal. In this experiment, we asked whether both pathways can give rise to cells with genomic characteristics of memory. To do this, we activated naive CD8 T cells ex vivo for 2 days followed by 1 day of culture without TCR stimulation. On day 3, we sorted Tcf7-YFP high and low populations, all from a single CellTrace peak representing cells that had undergone the same number of divisions. The sorted populations were cultured for an additional 6 days and sorted again on day 9 by Tcf7-YFP levels. These samples, as well as control naive, antigen-experienced, and effector samples, were processed for RNA-seq analysis.

Gene Expression in Bone-marrow derived Dendritic Cells stimulated with H. pylori lysate vs. E. coli lysate

Project description:Gene Expression changes in BMDCs stimulated with H. pylori vs. E. coli. The hypothesis tested was that the gene expression profile in BMDCs stimulated with H. pylori lysate will be less inflammatory than BMDCs stimulated with E. coli. BMDCs were generated from bone marrow of wild type female C57BL/6 mice using 100ng/mL Flt3L. DCs were harvested at day 8 and stimulated with either media alone, H. pylori antigen lysate or E. coli lysate. At 24 hours, the cells were harvested and RNA was isolated for microarray analysis.
