To this end, rat dams (Sprague-Dawley rats ) were fed diets with 0 or 1,000 IU vitamin D3 per kg diet 5 weeks prior to conception until the end of lactation. By using high-density DNA microarrays, we analyzed the gene-expression profile of mesenterial adipose tissue of both groups of newborn rats.

Characteristics of differentiated newly identified adipose precursor cells

Project description:We have identified proliferative cells in adipose tissue expressing adipocyte specific genes, which were named small proliferative adipocytes (SPA). Comparison of gene expression among stromal vascular cells (SVC), SPA and mature adipocyte (MA) revealed that adipocyte specific genes and several neuronal genes were increased in the order of SVC < SPA < MWA. However, proliferin 2 (PLF) was detected only in SPA. We found that SPA changed to lipid laden cells by incubation with differentiation medium containing insulin/dexamethasone/IBMX. In this study, we investigated the gene expression of differentiated SPA (SPAD) comparing with differentiated SVC (SVCD). Microarrays and real time PCR revealed that uncoupling protein 1 (UCP1) and PLF were strongly expressed in D-SPA. These data suggested that SPA might be able to differentiate into beige cells.

The white adipose tissue transcriptional response to withdrawal of vitamin B3

Project description:Distinct markers for early, mild vitamin B3 deficiency are lacking. To identify these, we examined the molecular responses of white adipose tissue to vitamin B3 withdrawal. We performed a dietary intervention in male C57Bl/6JRcc mice. A diet with a low but adequate level of tryptophan without nicotinamide riboside (NR) was compared to the same diet with NR at the recommended vitamin B3 (30 mg NR per kg diet). Physiological and circulating parameters were determined and global transcriptomics, qRT-PCR and histology of epididymal white adipose tissue (eWAT) were done. We observed a decreased insulin sensitivity and a shift from carbohydrate to fatty acid oxidation. This was consistent with molecular changes in eWAT, where we observed an altered MEK/ERK signalling, a lowering of glucose utilization markers and an increase in makers of fatty acid catabolism, which may be related to the consistent reduction of mitochondrial OXPHOS Complex I (mRNAs and protein). The synthesis pathway of tetrahydropteridine (BH4), an essential cofactor for neurotransmitter synthesis, was found to be increased. Based on our results, we propose the technically validated downregulation of Anp32a, Tnk2 and the upregulation of Mapk1, Map2k1, Mthfs, Mthfsl and Qdpr as a WAT transcriptional signature marker for mild vitamin B3 deficiency.

Maternal dietary antioxidants enrichment regulates weaned piglets’ adipose tissue functionality

Project description:Homeostasis during the perinatal period is critical for the correct development of mammals and unbalances in the redox potential are common in this stage. Thus, enrichment of maternal diets with antioxidants may be useful to improve piglet early development. We have tested the effects of sows’ diet enrichment with different antioxidants on the piglets’ adipose tissue functioning, in comparison to a control diet with a basal antioxidant level, by studying gene expression and cellularity. The maternal diet strongly influenced adipose tissue transcriptome of the offspring post-weaning. Piglets born to sows supplemented with either vitamin E or hydroxytyrosol show improved metabolic and antioxidant status of adipose tissue, while animals from control group show impaired homeostasis and activation of oxidative stress, immune signalling, and inflammation pathways. Moreover, vitamin E, when supplemented alone at a high dose, activated lipid metabolism and increased adipocyte size. When both vitamin E and hydroxytyrosol were combined, the gene expression profile was scarcely affected in comparison to the control, showing a prooxidant/proinflamatory adipose tissue, which is an unexpected result with different potential explanations. Findings deepen in the processes taking place in adipose tissue of genetically fat individuals and highlight the crucial role of antioxidants in fat cells metabolism

System level analysis of insulin action revels insight into whole body metabolism and insulin action in muscle and adipose tissue

Project description:Muscle and adipose tissue insulin resistance are major drivers of metabolic disease. To uncover pathways involved in insulin resistance specifically in these tissues, we leveraged the metabolic diversity of different dietary exposures and discrete inbred mouse strains. This revealed that muscle insulin resistance was driven by gene-by-environment interactions and was strongly correlated with hyperinsulinemia and decreased levels of ten key glycolytic enzymes. Remarkably, there was no relationship between muscle and adipose tissue insulin resistance. Adipocyte size profoundly varied across strains and diets, and this was strongly correlated with adipose tissue insulin action. The A/J strain in particular exhibited marked adipocyte insulin resistance and hypertrophy despite robust muscle insulin responsiveness, challenging the role of adipocyte hypertrophy per se in systemic insulin resistance. These data demonstrate that muscle and adipose tissue insulin resistance can occur independently and underscore the need for tissue-specific interrogation to understand metabolic disease.
