Project description:The cryopreservation of avian semen without significant loss of fertilizing ability is of practical interest to the poultry industry and for the conservation of genetic biodiversity. However, the unique physiological features make avian spermatozoa highly sensitive to damage caused by cryopreservation. The potential proteomic changes in turkey (Meleagris gallopavo) semen throughout the cryopreservation process have never been investigated previously. Therefore, the purpose of the present study was to evaluate the proteomic changes of spermatozoa during cryopreservation at three stages (fresh, equilibrated, and frozen/thawed semen) using 2-dimensional difference in-gel electrophoresis (2D-DIGE) coupled with matrix-assisted laser desorption/ionization mass spectrometry (MALDI TOF/TOF). No differentially expressed proteins (DEPs) were found in spermatozoa proteome of fresh and equilibrated semen. One hundred forty six protein spots differed significantly between fresh and frozen/thawed spermatozoa. One hundred three protein spots differed significantly between equilibrated and frozen/thawed spermatozoa. Most of the proteins that changed in response to cryopreservation were related to the generation of precursor metabolites and energy through aerobic respiration, flagellum-dependent cell motility, and binding of sperm to the zona pellucida. This suggests the complexity of the processes leading to decreases in the quality of cryopreserved semen.

Project description:Vitrification is not considered as ideal method for cryopreservation of human spermatozoa due to the presence of high concentrations of penetrating cryoprotectants in the vitrification medium. Several studies in the literature suggest that the outcome of semen cryopreservation is superior when cryopreserved using conventional rapid freezing protocol compared to vitrification. The present study was aimed at assessing the functional and proteomic changes in the human spermatozoa cryopreserved in vitrification medium without penetrating cryoprotectant. Leftover ejaculates (n = 71) from men visiting the andrology laboratory for routine semen analysis were used. The liquefied semen samples were cryopreserved using rapid freezing and vitrification for a minimum of seven days. Post-thaw motility, mitochondrial function, DNA integrity, and acrosome intactness of spermatozoa were similar between the cryoprotectant-free vitrification and rapid freezing methods. The head morphology of spermatozoa cryopreserved by the vitrification method was better preserved compared to rapid freezing method. Proteomic analysis led to the identification of a total of 4378 proteins in spermatozoa. However, differential proteomic profile observed in spermatozoa cryopreserved with vitrification and conventional rapid freezing methods compared to spermatozoa from the ejaculate indicates the possible differences in the functional competence of the spermatozoa cryopreserved using these two protocols. Even though vitrification of spermatozoa in medium without penetrating cryoprotectant holds promise as an attractive option owing to the simplicity and rapidity of the protocol, further studies are necessary to understand the safety of the protocol and pregnancy outcome upon using the vitrified-thawed spermatozoa for assisted conception and assisted reproductive technology.

Project description:<p>Cryopreservation of Chengde polled goat semen plays a critical role in conserving genetic resources, enhancing the utilization efficiency of superior breeding rams, and advancing artificial insemination techniques. However, spermatozoa are vulnerable to oxidative stress during the freezing process, which can significantly compromise sperm motility. This study demonstrated that supplementation of the semen cryoprotectant with 0.6% sericin significantly improved post-thaw sperm viability to 65.25% in Chengde hornless goats, while concurrently reducing both the sperm abnormality rate and intracellular ROS levels. Integrated TMT proteomics and LC/MS metabolomics identified 256 differentially expressed proteins and 109 differential metabolites between sericin supplementation and frozen control groups. Functional analysis revealed significant enrichment of differential metabolites, such as glutamine, in the alanine, aspartate, and glutamate metabolism pathway, concomitant with marked upregulation of antioxidant proteins including LRP8, GSTM3, and SIRT2. Thus, sericin enhances cryotolerance primarily by improving sperm viability, reducing oxidative damage, and sustaining energy metabolism. These findings establish the molecular foundation for optimizing goat semen cryopreservation and advancing artificial insemination technology.</p>

Project description:Our recent studies suggested that the freezability of carp semen is related to seminal plasma protein profiles. Here, we aimed to compare the spermatozoa proteomes of good (GF) and poor (PF) freezability semen of carp. To achieve this, we used two-dimensional difference in gel electrophoresis followed by MALDI-TOF/TOF mass spectrometry. The semen was classified as GF or PF based on sperm motility after freeze/thawing. We identified proteins enriched in spermatozoa of GF (22 proteins) and PF (18 proteins) semen. We also identified 12 proteins enriched in the supernatant after cryopreservation of PF semen. Good freezability is related to high concentrations of proteins involved in the maintenance of flagella structure, membrane fluidity, efficient control of Ca2+ and sperm motility, energy production, and antioxidative protection, which likely reflects the full maturation status of spermatozoa of GF semen. On the other hand poor freezability seems to be related to the presence of proteins identified as released in high quantities from cryopreserved sperm of PF. Thus, the identified proteins might be useful bioindicators of freezing resilience and could be used to screen carp males before cryopreservation, thus improve long-term sperm preservation in carp.

Twenty six protein spots were significantly enriched in ECF proteome after cryopreservation compared to equilibrated semen. The proteins enriched in ECF after cryopreservation were mainly related to NADH metabolic and glycolytic process.

Project description:In this study, two small RNA libraries were constructed using dry period and peak lactation dairy goat mammary gland tissues and sequenced by the Illumina Solexa high-throughput sequencing system. A total of 346 conserved and 95 novel miRNAs were identified in the dairy goat. The expression of miRNAs was confirmed by qRT-PCR in nine tissues and the mammary gland during development cycles. In addition, several candidate miRNAs that may be involved in mammary gland development and lactation were found by the comparison of miRNA expression profiles among different tissue and developmental stages of the mammary gland. This study provides the identification and profile of miRNAs related to the biology of the mammary gland in the dairy goat. The identification of these miRNAs could contribute to understanding the molecular mechanisms of lactation physiology and the development of the mammary gland in the dairy goat.

Project description:Cryopreservation causes significant lethal and sub-lethal damage to spermatozoa. In order to improve freezing outcomes, a comprehensive understanding of sub-lethal damage is required. Cryopreservation induced changes to sperm proteins have been investigated in several species, but few have employed currently available state of the art, data independent acquisition mass spectrometry (MS) methods. We used the SWATH LC-MS method to quantitatively profile proteomic changes to ram spermatozoa following exposure to egg yolk and cryopreservation. Egg yolk contributed 15 proteins to spermatozoa, including vitellogenins, apolipoproteins and complement component C3. Cryopreservation significantly altered the abundance of 51 proteins. Overall, 27 proteins increased (e.g. SERPINB1, FER) and 24 proteins decreased (e.g. CCT subunits, CSNK1G2, TOM1L1) in frozen thawed ram spermatozoa, compared to fresh spermatozoa. Chaperones constituted 20% of the proteins lost from spermatozoa following cryopreservation. These alterations may interfere with both normal cellular functioning and the ability of frozen thawed spermatozoa to appropriately respond to stress. This is the first study to apply SWATH mass spectrometry techniques to characterise proteins contributed by egg yolk based freezing media and to profile cryopreservation induced proteomic changes to ram spermatozoa.
