Targets identification of Biotin-Orf9b probe in HEK293T lysates

ABSTRACT: To identified Biotin- Orf9b probe interacting proteins, we applied SPIDER assay by using Biotin-Orf9b probe in HEK293T cell lysates, and the enriched protein was identified by mass spectrometry (MS) to quantitatively find the targets of Biotin-Orf9b interactors.

Project description:We engineering a G4-targeted biotin ligase probe (G4PID), which consists of three parts, RHAU23 at the C-terminus, miniTurbo at the N-terminus, and a flexible linker in the middle to prevent the disturbance of the two functional moieties. We used eCLIP to verified the G4s affinity of G4PID. We also used eCLIP method to verify the direct G4 interaction of Splicing factor 3B subunit 4 (SF3B4) in cellular transcriptome.

Effect of biotin deficiency on gene expression in Aravbidopsis leaves

Project description:In addition to its essential metabolic functions biotin is suggested a critical role in regulating gene expression. The first committed enzyme in biotin biosynthesis in Arabidopsis, 7-keto-8-aminopelargonic acid synthase is encoded by At5g04620 (BIO4). We isolated a novel T-DNA insertion mutant of BIO4 (bio4-1) showing a spontaneous cell death phenotype that could be rescued both by exogenous biotin and genetic complementation. The bio4-1 plants exhibited massive accumulation of hydrogen peroxide. The T-DNA insertion in the bio4-1 mutant contains a Methionine Sulfoxide Reductase B9 (AT4G21850) under a constitutive 35S CaMV promoter. To dissect the specific effects of biotin defiency on gene expression on a genome wide scale, we compared expression of wt Col-0 leaves with bio4-1 mutants and bio4-1 mutants complemented with D-biotin.

Project description:The CDKN2A/B locus at 9p21.3 contains crucial tumor suppressors (P16, P14, and P15) and oncogenic lncRNA ANRIL genes. This locus is most frequently inactivated in cancer genomes by deletion and DNA methylation. However, the mechanisms coordinately regulating their expression level are far from clear. In the present study, a novel lncRNA, P14AS, was characterized in the antisense strand of the fragment near CDKN2A in human cell lines using CDKN2A-specific probe captured RNA-sequencing (RNACap-Seq).
