Microglia coordinate activity-dependent protein synthesis in neurons through metabolic coupling

Project description:De novo protein synthesis is required for long-lasting synaptic plasticity and memory, but it comes with a great metabolic cost. In the mammalian brain, it remains unclear which cell types and biological mechanisms are critical for sensing and responding to increased metabolic demand. Here we demonstrate that microglia, resident macrophages of the brain, coordinate metabolic coupling between endothelial cells, astrocytes, and neurons to fuel protein synthesis in active neurons. Increasing metabolic demand via a motor task stimulates microglia to secrete the hypoxia-responsive protein CYR61, increasing glucose transporter expression in brain vasculature. Depleting microglia reduces training-induced metabolic fluxes and neuronal protein synthesis, which can be reproduced by blocking CYR61 signaling. Thus, we define a neuroimmune metabolic circuit required for on-demand protein synthesis in mouse motor cortex.

Identifying genes related to salt stress and analyzing their regulatory mechanisms in peanuts under salt stress

Project description:Peanut (Arachis hypogaea L.) is a prominent legume and oilseed crop cultivated globally for its economic significance and nutritional value, as it is rich in fat, protein, carbohydrates, and micronutrients. To meet the growing demand for edible oil and protein, it is necessary to further expand the cultivation area and improve the seed yield of peanut. Salinized land represents a potential resource for expanding peanut cultivation. However, most peanut genotypes are sensitive to salt stress, and soil salinity severely limits peanut productivity. Therefore, enhancing salt tolerance in peanut is essential. This study aimed to identify genes associated with salt stress through transcriptome analysis, thereby providing a genetic basis for improving salt tolerance in peanut. To identify salt stress-related genes, two cultivars—Huayu 33 (HY33, salt-tolerant) and Huayu 9115 (HY9115, salt-sensitive)—were subjected to salt stress treatment. Plants of HY33 and HY9115 were collected at 0 h, 3 h, 12 h, 24 h, and 48 h after salt treatment for transcriptome sequencing. Using the RNA sequencing data, numerous differentially expressed genes were identified as candidates for further functional study. This study provides extensive gene expression data for identifying key genes related to salt stress and establishes a basis for elucidating the underlying regulatory mechanisms. These findings can contribute to enhancing the salt tolerance of peanut through molecular breeding.

Deciphering the Sperm RNA Code of Aging with PANDORA-seq

Project description:Sperm aging impacts male fertility and offspring health, highlighting the need for reliable aging biomarkers to guide reproductive decisions. Using PANDORA-seq, a novel method to comprehensively profile small non-coding RNAs (sncRNAs), we identified an "aging cliff" in mouse sperm RNA profiles-a sharp age-specific transition marked by significant shifts in genomic and mitochondrial tRNA-derived small RNAs (tsRNAs) and rRNA-derived small RNAs (rsRNAs). Notably, rsRNAs embedded in sperm heads exhibited a transformative length shift, with longer rsRNAs increasing and shorter ones decreasing with age, suggesting altered biogenesis with age. Remarkably, this sperm rsRNA length shift was consistently observed in two independent human aging cohorts, highlighting its evolutionary significance. Moreover, a combination of tsRNAs and rsRNAs (tsRNA/rsRNA cocktails) resembling those in aged sperm induced transcriptomic changes in embryonic stem cells, impacting metabolism and neurodegeneration pathways, mirroring the phenotypes observed in offspring of aged sperm. These results reveal new insights of sperm aging, highlighting the conserved rsRNA length shift in mice and humans, with translational potential for fertility and intergenerational health
