Project description:Fulminant myocarditis (FM) is the most serious type of childhood myocarditis. However, the molecular mechanism underlying the pathogenesis of FM has not been fully elucidated. Studies have shown that small extracellular vesicles (sEVs) play important roles in many diseases, but any potential role of sEVs in pediatric FM has not been reported. Here, the differential expression profiles of lncRNAs in plasma sEVs were studied in five children with FM and five healthy children using whole transcriptome sequencing, followed by functional analysis and screening of immune-related target genes. A total of 68 up-regulated and 11 down-regulated differentially expressed sEVs-lncRNAs were identified. Functional analysis showed that the differentially-expressed sEVs-lncRNAs were mainly involved in immune processes, apoptosis, and protein efflux. Further analysis of immune-related target genes revealed that differentially expressed sEVs-lncRNAs were closely related to immune activation, immune cell migration and cytokine pathway signal transduction in FM. Thus, sEVs-lncRNAs may play an important role in the pathogenesis of FM in children

Project description:This study presents a scalable method for isolating small extracellular vesicles (sEVs) according to MISEV (minimal information for studies of extracellular vesicles) guidelines from 4 mL of bovine plasma while achieving a high purity suitable for proteomics analysis. The protocol combines ultracentrifugation (UC), size-exclusion chromatography (SEC), and ultrafiltration (UF) to minimize contamination by lipoproteins and free proteins. The purified sEVs were characterized through transmission electron microscopy (TEM), tunable resistive pulse sensing (TRPS), dynamic light scattering (DLS), and flow cytometry (FC), assessing particle size, morphology, and the presence of membrane markers such as CD9. The purity assessed by the particle-to-protein ratio (4.8 × 10⁸ particles/µg in average) was improved (× 10) compared to the previous protocol. Mass spectrometry (LC-MS/MS) and Parallel Reaction Monitoring (PRM) identified 417 proteins, 93% of which are referenced in sEVs databases. Specific markers of sEVs have been identified including cytosolic (HSPA8, SDCBP, ACT, TUB, and GAPDH) and membrane (CD9 and CD81) Markers. Gene Ontology (GO) analysis revealed an enrichment of sEVs proteins. This study demonstrates that this sEVs enrichment protocol is well-suited for small plasma volumes. These results pave the way for future studies of biomarker research on animal health and performance.

Project description:Chronic kidney disease (CKD) accelerates vascular calcification (VC) via phenotypic switching of vascular smooth muscle cells (VSMCs). We investigated the roles of circulating small extracellular vesicles (sEVs) between the kidneys and VSMCs and uncovered relevant sEV-propagated microRNAs (miRNAs) and their biological signaling pathways. We established CKD models in rats and mice by adenine-induced tubulointerstitial fibrosis. The miRNA transcriptome of sEVs revealed a depletion of several miRNAs in CKD. Their expression levels in sEVs from CKD patients were correlated to kidney function. This study revealed the transcriptomic landscape of miRNAs propagated in sEVs in CKD. We investigated the therapeutic potential of miRNAs in VC.

Project description:Human cytomegalovirus (hCMV) primo-infection, reinfection and/or reactivation is a major issue during pregnancy and affects 1% of live births in western countries, making hCMV the most frequently transmitted virus in utero. Despite the extensive research conducted so far, the pathophysiology of this congenital infection remains unclear. Recently, increasing evidence point out the role of small extracellular vesicles (sEVs) in cell-cell communication underlying the feto-placenta-maternal dialogue during pregnancy. In this study, we examined the impact of hCMV infection on the protein composition and function of placental sEVs. We observed that infection of placental cells led to an alteration of protein composition of their secreted sEVs, suggesting that placental sEVs may acquire a proviral phenotype. Functional studies performed on fetal recipient cells, notably neural stem cells, confirmed the ability of sEVs produced by infected cells to facilitate further infection of naive recipient cells. Altogether, our study demonstrates that placental sEVs are key players of hCMV pathophysiology during congenital infection, and may favor the transmission of the virus towards the fetus.

Project description:Extracellular vesicles (EVs) are important mediators of intercellular communication and promising sources of diagnostic and therapeutic biomarkers, yet effective EV isolation remains challenging due to trade-offs among yield, purity, and adaptability across biofluids. Here, we introduce divalent aptamer-mediated clustering (DAC), a streamlined affinity-based EV isolation strategy that induces controllable vesicle clustering and enables recovery by standard filtration. By exploiting multivalent aptamer binding to EV surface markers, DAC converts nanoscale vesicles into micron-scale clusters while preserving EV integrity and biological activity. We demonstrate robust EV isolation from plasma, urine, and cell culture media, and benchmark DAC against ultracentrifugation, density gradient ultracentrifugation, and size-exclusion chromatography. DAC achieves comparable or improved EV yield and purity with reduced processing time, cost, and operational complexity. Proteomic and metabolomic analyses show that DAC isolates affinity-defined EV subpopulations with cargo profiles distinct from those obtained using conventional methods. Moreover, DAC is readily adapted to alternative EV targets, exemplified by enrichment of EpCAM-positive EVs. Together, DAC provides a versatile and accessible platform for studying EV heterogeneity, function, and molecular composition.

Project description:Background. Ductal carcinoma in situ (DCIS) is a non-invasive precursor to breast cancer that increases lifetime risk for invasive disease and, in rare cases, causes distant metastases without local recurrence. Tumor-derived small extracellular vesicles (sEVs), or exosomes, mediate tumor progression, systemic signaling, and metastasis; however, the timing of release and biodistribution of sEVs from early preinvasive lesions remains unclear. Methods. Here, we investigated the release kinetics and systemic dissemination of sEVs secreted by DCIS cells confined to the mammary ducts using an orthotopic mouse mammary intraductal xenograft model with MCF10DCIS cells engineered to express NanoLuc (NLuc)-tagged CD63, an sEV marker. Tumor growth was monitored using bioluminescent imaging (BLI) in vivo and ex vivo; DCIS outgrowth and focal invasion were confirmed by fluorescent immunohistochemistry; and NLuc activity was assayed in tissue lysates. Next-generation RNA sequencing of brain tissue was used to evaluate gene expression changes at 28 days post-implantation (dpi). Results. Longitudinal BLI revealed CD63-NLuc signals in mammary glands (P < 0.05), and distant organs, including spleen (P < 0.05), liver (P < 0.05), lungs (P < 0.01) and brain (P < 0.01) by day 14 followed by detection in heart tissue (P < 0.001) and in the circulation by day 21, preceding histological evidence of invasion. Further, CD63-NLuc activity was significantly and positively correlated with BLI levels in mammary glands and plasma (P < 0.05). Immune, metabolic, and stress response pathways were downregulated in brain tissue at 28 dpi compared to 3 dpi. Conclusions. These findings demonstrate systemic dissemination of tumor-derived sEVs during DCIS progression, suggesting roles in early microenvironmental conditioning and metastatic preconditioning prior to overt invasion.

Project description:Tendon injuries are challenging to treat due to the limited blood supply and low cell density in tendon tissue. Tendon stem cells (TSCs) typically reside in a hypoxic environment (1%-5% oxygen), which may be crucial for their physiological function. In this study, we demonstrated that under hypoxic conditions, TSCs release small extracellular vesicles (sEVs) enriched with functional molecules (mainly non-coding RNAs and proteins). These sEVs significantly reduced fibrosis and inflammation in a rat model of Achilles tendon injury, while enhancing the expression of tendon-related factors, thus promoting tendon healing and improving tissue structure and function. In vitro experiments revealed that hypoxia-induced TSC-derived sEVs not only promoted TSC differentiation but also inhibited the conversion of tenocytes to myofibroblasts and reduced the expression of inflammatory markers in macrophages. These effects were mediated through the HIF-1ɑ/miR-145-3p and miR-504 pathways. Proteomic analysis revealed that hypoxia-induced changes in the protein expression profile of sEVs, with differentially expressed proteins closely associated with tendon injury repair. Our findings provide compelling evidence that hypoxia-induced sEVs from TSCs play a pivotal role in tendon repair, offering a robust theoretical basis for the development of novel therapeutic strategies targeting tendon injuries.

Project description:Adipose tissue dysfunction drives hepatic lipid overload in metabolic dysfunction-associated steatotic liver disease (MASLD), yet the involvement of adipose tissue-derived small extracellular vesicles (sEVs) remains unclear. Herein, we showed that transplanting adipose tissue from high‑fat diet-fed male mice exacerbated insulin resistance and hepatic steatosis in lean recipients. Adipose‑specific Sirt3 overexpression (Sirt3AKI) alleviated insulin resistance and liver steatosis in high fat diet-fed male mice, whereas adipose‑specific Sirt3 knockdown aggravated these phenotypes. Moreover, adipose‑derived sEVs play key roles in hepatic lipid metabolism by delivering miRNAs in Sirt3AKI male mice. MicroRNA sequencing identified miR-30a-3p was increased in the circulating sEVs from high fat diet-fed male mice, while decreased in sEVs from Sirt3OE adipocytes and Sirt3AKI male mice. Mechanistically, miR‑30a‑3p promoted hepatic steatosis by targeting Becn1; this process was suppressed when Sirt3 downregulated miR‑30a‑3p transcription via deacetylation of H3K56. These findings highlight the critical role of adipose sEVs microRNAs in driving hepatocyte lipotoxicity, and suggest miR-30a-3p inhibition could be a therapeutic intervention for MASLD.
