Project description:The testis is an important reproductive organ in male mammals, serving as the site for spermatogenesis, androgen synthesis, and secretion. Non-coding RNAs (ncRNAs) plays an important regulatory role in various biological processes. However, the regulatory role of ncRNAs on yak testicular development and spermatogenesis is still largely unclear. In this study, the expression profiles of circRNA, miRNA, and mRNA in testicular tissues were compared between 6 months (Y6M), 18 months (Y18M), and 4 years old (Y4Y), with a significant difference in testicular development using RNA-seq. A total of 22 differentially expressed circRNAs (DE circRNAs), 69 differentially expressed miRNAs (DE miRNAs), and 64 differentially expressed mRNA (DE mRNAs) were detected in testicular samples of Y6M, Y18M, and Y4Y, respectively. Furthermore, GO and KEGG analysis results showed that the source genes of DE circRNAs, predicted target genes of DE miRNAs, and DE mRNAs concentrated on signaling pathways and GO terms that were related to sperm synthesis, vitality, and testicular development, such as cell cycle, Wnt signaling pathway, MAPK signaling pathway, GnRH signaling pathway, and spermatogenesis process. A circRNA-miRNA-mRNA network analysis revealed that some differentially expressed non-coding RNAs (DE ncRNAs) may be involved in testicular spermatogenesis, including miR-574, miR-449a, CDC42, CYP11A1 and et al. At the same time, there are various circRNA-miRNA interaction pairs. This study provides a deep understanding of the regulatory network of ncRNA regulation in yak testicular development, providing data support for further elucidating the molecular mechanisms of yak testicular development.

Project description:Studying spermatogenesis is the key to understanding the development mechanism of the yak reproductive system. Although N6-methyladenosine (m6A) RNA modification has been reported to regulate spermatogenesis and reproductive function in mammals, the molecular mechanism of m6A in yak testis development and spermatogenesis remains largely unknown. Therefore, we collected testicular tissue from junile and adult yaks, and found that the m6A level significantly increased after sexual maturity in yak. In MeRIP-seq, 1,702 hypermethylated peaks and 724 hypomethylated peaks were identified. The hypermethylated differentially methylated RNAs (DMRs) (CIB2, AK1, FOXJ2, PKDREJ, SLC9A3, and TOPAZ1) mainly regulated spermatogenesis, while the hypomethylated DMRs (BCL6, USP25, CD96, EPHA2, and TAF12) mainly participated in immune response. Functional enrichment analysis showed that DMRs were significantly enriched in the adherens junction, gap junction, and Wnt, PI3K, and mTOR signaling pathways, regulating cell development, spermatogenesis, and testicular endocrine function. The functional analysis of differentially expressed genes showed that they were involved in the biological processes of mitosis, meiosis, and flagellated sperm motility during sexual maturity of yak testis. We also screened the key regulatory factors of testis development and spermatogenesis by combined analysis, which included BRCA1, CREBBP, STAT3, and SMAD4. This study has important research value and practical guiding significance for yak genetic improvement, particularly in understanding the molecular mechanism and evolutionary dynamics of the progression of testis developmental stages in yaks.

Project description:Establishment and maintenance of spermatogenesis need a complex process and vast regulatory network. There is growing evidence reveals that long noncoding RNAs (lncRNA) plays important role in regulating testicular development and spermatogenesis in a stage-specific way. We here report the identification of lncRNA LOC108635509 as key lncRNA regulator in black goat spermatogenesis. In the current study, we screened the transcriptomes (lncRNA and mRNA) of testicular from Guangxi black goats before puberty (3 days old, D3; 30 days old, D30), puberty (90 days old, D90) and postpuberty (180 days old, D180), and found there were 1211, 12180, 834 differential lncRNAs and 1196, 8838,269 differential mRNAs at the ages of D30 vs D3, D90 vs D30, and D180 vs D90. The expression pattern of differentially expressed (DE) lncRNAs indicated that D90 was a key node of lncRNAs participated in the regulation of testicular development and spermatogenesis in black goat. The of GO and KEGG analyses identified that DE lncRNAs and their target genes regulated spermatogenesis through MAPK, Ras, and PI3K-Akt signal pathways. Using cis- and trans-acting, 39 DE lncRNA-targeted genes were found to be enriched for male reproduction. Of these, LOC108635509, which specific expressed in testis and upregulated the expression levels at D90, was found participated in the regulation of testicular development through promoting the proliferation of SCs. Overall, this study provides new insight into the regulatory mechanisms that support spermatogenesis and testicular development in black goats.

Shotgun proteomics was used to compare proteomes of bull testicular, caput and cauda epididymal spermatozoa. This is crucial for understanding the physiological mechanisms that lead to sperm competence.

Project description:Hair follicles of the yak are in anagen and catagen , the hair follicles of healthy female yaks around 2 years old are collected for the preparation of single-cell suspension, the cells were sequenced by scRNA-seq on the 10x genome platform. A total of about 12000 single-cell transcriptome information were obtained. According to the reported marker genes, the main cell groups in anagen and catagen of yak were identified. Based on the analysis of pseudotime trajectory, the differentiation trajectory of epidermal cell lineage and dermal cell lineage during hair follicle development and the dynamic changes of genes during differentiation were described.

Project description:The project aimed to compare testicular proteomes from patients with obstructive and non-obstructive azoospermia in order to identify molecular signatures involved in spermatogenesis as well to identify candidate biomarkers for discriminating between different types of azoospermia. The samples used in this study were formalin fixed paraffin embedded (FFPE) testicular tissues obtained by biopsy from men with clinical diagnosis of azoospermia. Samples were grouped according to the histopathological report in 3 groups: 1) Spermatogenesis (obstructive azoospermia), 2) Hypospermatogenesis and 3) Sertoli cell only syndrome (SCO). Patients were aged 31–46 years with no differences among groups regarding age (Median age (Hypospermatogenesis) =35; Median age (SCO) =34 and Median age (Spermatogenesis) =39). Each group had 9 samples or 27 samples in total were used for the comparative proteomics analysis by label-free data-independent LC-MS/MS.

Here, we developed a formation-differentiation culture approach to generate optimized testicular organoids (O-Torgs) derived from neonatal mouse primary testicular cells. These O-Torgs could essentially recapitulate the seminiferous tubule morphogenesis, Sertoli cell maturation and testosterone secretion. Importantly, O-Torgs not only support spermatogenesis but also sustain proliferative undifferentiated spermatogonia and continuous generation of functional haploid cells for up to three months, with offspring derived from these haploid cells displaying normal growth and reproductive capability to F2 generation. Mechanistically, we found that the formation stage enhanced the reconstruction of tubule-like structures and subsequent spermatogenesis by providing a more conducive extracellular matrix niche. Finally, O-Torgs were proved to be effective in modeling male infertility and drug screening, with BTT-3033 acting as a potential drug in protecting busulfan-mediated germ cell loss. Overall, our work establishes a strategy to obtain functional testicular organoids, offering promising avenues for male infertility modeling and drug discovery.
