RNA-seq analysis of lenvatinib-sensitive and lenvatinib-resistant tumor tissues from a subcutaneous HCC mouse model

Project description:To investigate the molecular mechanisms underlying acquired resistance to lenvatinib in hepatocellular carcinoma (HCC), we established a lenvatinib-resistant mouse model using H22 cell-based subcutaneous xenografts. Tumor tissues were collected from lenvatinib-sensitive (H22-NR) and lenvatinib-resistant (H22-LR) mice. We performed RNA-sequencing to identify differentially expressed genes and signaling pathways that mediate lenvatinib resistance. Comparative gene expression profiling analysis between the NR and LR groups was conducted to screen for key drivers of drug resistance.

HIF-1 Signaling Contributes to Lenvatinib Resistance in Patient-Derived HCC Organoids

Project description:Resistance to lenvatinib remains an important limitation in the treatment of hepatocellular carcinoma (HCC). In this study, six patient-derived HCC organoid lines were established and classified as lenvatinib-sensitive or lenvatinib-resistant according to their ex vivo drug responses. The organoids retained histological and immunophenotypic features of their matched parental tumors. Lenvatinib-sensitive organoids exhibited more pronounced morphological changes and reduced ATP activity at higher lenvatinib concentrations, whereas ATP activity remained largely unchanged in resistant organoids. Transcriptome sequencing identified 408 upregulated and 269 downregulated genes in lenvatinib-resistant compared with lenvatinib-sensitive organoids, with HIF-1 signaling among the altered pathways. In resistant organoids, lenvatinib increased HIF-1alpha, ANGPT2, and HK3 mRNA expression, whereas comparable changes were not detected in sensitive organoids. KC7F2 reduced the lenvatinib-associated increases in these transcripts and further decreased ATP activity when combined with lenvatinib in resistant organoids. In organoid-derived xenografts, the combination of lenvatinib and KC7F2 resulted in the greatest reduction in tumor growth and lower HIF-1alpha, ANGPT2, and HK3 mRNA expression than lenvatinib alone. These findings indicate that HIF-1 signaling contributes to the lenvatinib-resistant phenotype in patient-derived HCC organoids and that its inhibition may enhance the response to lenvatinib.

Project description:Lenvatinib is the FDA-approved targeted drug for advanced hepatocellular carcinoma (HCC), but the efficacy is modest due to drug resistance. Tumor kinome re-wiring governs drug resistance in resistant cancer cells, which is an obstacle for efficient cancer therapy. Therefore, identification the kinases critical for this rewiring process in HCC is crucial.This study reveals the new role of CDK6 and its mechanistic insight in regulation of cancer stemness and drug resistance. These results support the combination treatment of lenvatinib with palbociclib for advanced HCC patients. Further studies will optimize patient target selection and identify the best treatment combinations.

Syntaxin-6 mediated autophagy confers lenvatinib resistance in hepatocellular carcinoma

Project description:Lenvatinib is the first-line therapy for inoperable HCC. However, intrinsic and acquired drug resistance occurs during the treatment period. Autophagy is an adaptive response that favors tumor survival under stress. In the present study, we aim to reveal the unknown autophagic engagement in lenvatinib resistance. Lenvatinib-resistant HCC cell lines and xenograft mouse HCC models were established to identify the key regulator of lenvatinib resistance in HCC. By in vitro functional restoration assays and autophagic flux detection, we demonstrated that the Syntaxin-6 (STX6) -mediated autophagy induced lenvatinib resistance of HCC cells. Mechanistically, Co-immunoprecipitation assay and mass spectrometry indicated that the interactions of STX6 with Beclin1, VTI1A, and VAMP3 facilitated autophagy, leading to the lenvatinib resistance. Additionally, STX6 enhanced the ability of proliferation, migration, and invasion of HCC in vitro and in vivo. Clinically, STX6 expression was significantly elevated in HCC tissues compared to it in para-tumor tissues. High STX6 expression predicted poor outcomes for patients following resection. Moreover, high expression of STX6 displayed low preventive efficacy of lenvatinib as a postoperative adjuvant treatment for HCC patients with a high risk of recurrence. Collectively, we identified that STX6-mediated autophagy was a pivotal determinant of lenvatinib resistance in HCC, providing a potential target to overcome lenvatinib resistance for HCC patients.

RNA Sequencing of HCC cells after lenvatinib, gefitinib, and combination treatment

Project description:Using a kinome-centred CRISPR/Cas9 genetic screen, we identify here that inhibition of the epidermal growth factor receptor (EGFR) is synthetic lethal with lenvatinib in liver cancer cells. We found that the combination of the EGFR inhibitor gefitinib and lenvatinib displays potent anti-proliferative effect in HCC cell lines that express EGFR in vitro and of xenografted HCC cell lines or patient-derived HCC tumours in mice. Herre, we analyzed the different transcriptome profiling of HCC cells treated with DMSO, lenvatinib, gefitinib, and lenvatinib plus gefitinib by RNA-sequencing.

GOLM1 dictates acquired Lenvatinib resistance by a GOLM1-CSN5 positive feedback loop upon EGFR signaling activation in hepatocellular carcinoma

Project description:Lenvatinib is a multiple receptor tyrosine kinases inhibitor (TKI) authorized for first-line treatment of hepatocellular carcinoma (HCC). However, Lenvatinib resistance is common in HCC clinical treatment, highlighting the urgent need to understand mechanisms of resistance. Here, we identified Golgi membrane protein 1 (GOLM1), a type II transmembrane protein originally located in the Golgi apparatus, as a novel regulator of Lenvatinib resistance. We established Lenvatinib resistance in human HCC cell lines PLC/PRF/5. Through RNA-seq, we found that GOLM1 was significantly upregulated, which was further validated in patients derived tumor tissue and peripheral blood. GOLM1 overexpression contributes to Lenvatinib resistance and HCC progression in vitro and in vivo. Mechanistically, GOLM1 upregulates CSN5 expression through EGFR-STAT3 pathway. Reversely, CSN5 deubiquitinates and stabilizes GOLM1 protein by inhibiting ubiquitin-proteasome pathway of GOLM1. Collectively, our data demonstrate that GOLM1 is one of the key driver of Lenvatinib resistance in HCC and depicts a molecular network in Lenvatininb resistant HCC cells.
