Project description:Purpose: To characterize exosomes derived from a mouse corneal progenitor cell line and investigate their role in delayed wound healing and sensory neuropathy in diabetic corneas. Methods: TKE2 cells were cultured in normal glucose (NG, 5 mM) or high glucose (HG, 25 mM) media for three passages. At passage 4, confluent cultures with or without scratch wounding were collected at 8 and 96 h. Exosomes were isolated by centrifugation and filtration and characterized by nanoparticle tracking analysis, cryo-TEM, and Western blotting. Their functions were tested by applying exosomes to unwounded and wounded corneas. The purified exosomes were analyzed by LC-MS/MS proteomics. The role of two identified proteins, HTRA1 and STAT3, in epithelial wound closure was assessed by siRNA and small molecular inhibitor, respectively. Results: The isolated extracellular vesicles were confirmed as exosomes, which were taken up by corneal epithelial cells and sensory nerves. While there were no effects on epithelial wound closure of NL corneas, exosomes from healing, but not quiescent, TKE2 cells accelerated delayed wound closure in DM corneas. In wounded corneas, siRNA knockdown of HTRA1—an abundant protein in healing TKE2 exosomes—impaired wound healing in normal but not diabetic corneas, while inhibition of exosomal Stat3 partially reduced the capacity of healing TK2 exosomes in promoting epithelial wound closure in DM corneas. Conclusions: Exosomes secreted by corneal epithelial progenitor cells exert distinct functions depending on culture conditions. Modulating exosomal contents may represent a therapeutic strategy to enhance wound healing in diabetic corneas.

Project description:Bone marrow (BM) niches provide an optimal substrate for multiple myeloma (MM) cell lodgement and growth. Nevertheless, little is known about the putative mechanisms by which the BM microenvironment can lead to initiation or progression of oncogenesis in this disease. We have demonstrated that BM mesenchymal stromal cell-derived exosomes transfer their miRNA and protein content to clonal plasma cells, thus acting as synaptic vesicles responsible for molding the microenvironment surrounding multiple myeloma (MM) cells, leading to MM growth, dissemination and, therefore, disease progression. We used microarray to detail the changes in microRNA expression in MM-BM mesenchymal stromal cell (MSC)-derived exosomes, compared to normal- and monoclonal gammopathy of undetermined significance- BM-MSC-derived exosomes. Exosomes have been isolated from cell culture supernatant of BM-MSCs (MM=7; MGUS=2; Normal=4), and subsequently evaluated at ultrastructural level by using electron microscopy and immunogolf labeling. RNA was extracted; and miRNA profiling has been assessed by using TaqMan human miRNA profiling. Mean miRNA expression value has been used for miRNA RT-qPCR data normalization, as described (Mestdagh et al., 2009).

Project description:We employed human HaCaT cells as a model system to identify cellular proteins that accompany SDS-induced toxicity based on a proteomic approach. HaCaT human keratinocyte cell line were treated with a non-cytotoxic dose of SDS (25 µg/ml, as determined by the MTT assay and microscopically examination) for 48 h. The altered abundance of proteins from HaCaT keratinocytes exposed to SDS was analyzed by LC-MS/MS approach and quantified using Progenesis LC software. The abundance of 217 proteins (which were identified by multiple peptides, ≥ 2) was altered in keratinocytes exposed to SDS; in which 131 proteins had increased abundance while 86 proteins was down regulated. The Pathview map of 131 up-regulated proteins was built and enhancement of glycolysis/gluconeogenesis was found.

Project description:Vitiligo is a skin disease characterized by the destruction of epidermal melanocytes due to oxidative stress and autoimmune response. As the main responder to oxidative stress, keratinocytes facilitate melanocyte loss under oxidative stress by inducing melanocyte death and recruiting antigen-specific CD8+ T cell to skin to destroy melanocytes. It has been proved that keratinocytes secrete abundant functional exosomes, but the role of exosomes secreted from keratinocytes under oxidative stress in vitiligo pathogenesis is unknown. We found that oxidative stress enhanced the secretion of exosomes from keratinocytes. These exosomes (OS-Exos) administration aggravated melanocyte loss and CD8+ T cell infiltration in the epidermis of tail in the vitiligo mouse model, thereby driving vitiligo progression. OS-Exos suppressed the survival of melanocytes while promoting the proliferation and activation of CD8+ T cells in vitro. As miRNAs contained in OS-Exos might be responsible for the functions of OS-Exos on melanocytes and CD8+ T cells, we employed small RNAs-seq analysis to screen miRNAs enriched in OS-Exos as compared to those in Exos (from untreated HaCaT cells). The hierarchical clustering analysis and the volcano plot showed the distinct signatures of known miRNAs as well as novel miRNAs between OS-Exos and Exos. In total, 276 differentially expressed miRNAs were discovered, with 134 and 142 miRNAs displaying significant up- or down-regulation, respectively, in OS-Exos compared with Exos (P<0.05; fold change≥2. The above data suggest that miRNAs enriched in OS-Exos might contribute to the facilitatory role of OS-Exos in vitiligo progression.

Project description:The purpose of this study is to search for aberrant genes in HaCaT keratinocytes after chronic exposure to arsenic trioxide. The objective of the investigation was to discover the mechanism of arsenic carcinogenicity in human epidermal keratinocytes. We hypothesize that a combined strategy of DNA microarray, qRT-PCR and gene function annotation will identify aberrantly expressed genes in HaCaT keratinocyte cell line after chronic treatment with arsenic trioxide. HaCaT cells were chronically exposed to 0.5M-BM-5g/mL arsenic trioxide (As2O3) up to 22 passages and RNA was extracted. Microarray data analysis identified 14 up-regulated genes and 21 down-regulated genes in response to arsenic trioxide Two experimental groups: 1. Project description:To investigate the regulatory effects of the ECM@SF-AGS-CS-LA hydrogel (HC), exosomes (HB), and their combination (HA) on the metabolic profiles of wound tissues in diabetic mice, HaCaT keratinocytes were employed as an in vitro cellular model. Untargeted metabolomics profiling was performed on HaCaT cells subjected to the respective treatments. Five pairwise comparisons were conducted: HC versus HD (diabetic control), HB versus HD, HA versus HD, HA versus HC, and HA versus HB. KEGG pathway enrichment analysis was applied to identify biologically relevant pathways significantly associated with differentially abundant metabolites (P < 0.05). This analysis revealed 8, 28, 30, 30, and 97 significantly enriched pathways in the HC vs HD, HB vs HD, HA vs HD, HA vs HC, and HA vs HB comparisons, respectively.

Project description:Small vesicles, known as exosomes, are secreted from various cell types. Exosomes secreted by mesenchymal stem cells have therapeutic effects against a variety of diseases, and may be able to partially replace stem cell therapies. Previously, we established and characterized human leukocyte antigen (HLA) haplotype homo (HHH) dental pulp cell (DPC) lines from human wisdom teeth. In this report, we purified the exosomes secreted from HHH-DPCs and evaluated their therapeutic potential in a periodontitis model. The exosomes purified from HHH-DPCs showed homogeneous and spherical membrane structures, and showed low but significant expression of HLA class I molecules. The exosomes further promoted proliferation and migration in DPCs. A comparison of miRNAs revealed that the HHH-DPC exosomes contained higher levels of multiple Let-7 family miRNAs compared to HHH-induced pluripotent stem cell (iPSC)-derived exosomes. Finally, the HHH-DPC exosomes showed preventive effects in a mouse model of periodontitis induced by lipopolysaccharides (LPS). In summary, HHH-DPC exosomes expressed HLA molecules which may induce an immune response in HLA-mismatched transplantations. However, they successfully stimulated the proliferation and migration of cells and showed suppressive effects on LPS-induced periodontitis. Therefore, HHH-DPC exosomes show great potential for applications in periodontal treatments.
