Project description:UV radiation is a ubiquitous component of solar radiation that affects plant growth and development. Analysis of natural variation in response to UV radiation revealed significant differences among natural accessions of Arabidopsis thaliana. However, the genetic basis of this is to a large extent unknown. Here, we analyzed the response of Arabidopsis accessions to UV radiation stress by performing RNA-sequencing of three UV sensitive and three UV resistant accessions. The genome-wide transcriptional analysis revealed a large number of genes significantly up- or down-regulated only in sensitive or only in resistant accessions, respectively. Mutant analysis of few selected candidate genes suggested by the RNA-sequencing results indicate a connection between UV radiation stress and plant-pathogen-like defense responses.

Project description:Clematis terniflora DC. is used as traditional herb medicine since ancient China. The active substances in C.terniflora have been confirmed to be effective in treating disease like prostatitis. The UV light radiation is a common environmental factor that damage the plants and has an influence on the primary metabolism and secondary metabolism. Previous researches have shown that ultraviolet B (UV-B) radiation followed by dark stress acting on C.terniflora leaves，resulting in the accumulation of secondary metabolites. An in-depth understanding of how C.terniflora leaves respond to UV-B stress is crucial for improving medicinal plants value. Here, we conducted label-free proteomic and phosphoproteomic to explore the protein changes under UV-B and UV-B combined with dark treatment, respectively. A total of 2839 proteins and 1638 phosphorylated proteins were identified. Integrative omics revealed the photosynthetic system and carbohydrate balance were modulated in both stresses. The phosphoproteomic data indicated that, mitogen-activated protein kinase signaling pathway was triggered, meanwhile, the abundance of phosphorylated proteins related to the osmotic stress were increased under UV-B stress. Differential abundant phosphoproteins from UV-B followed by dark treatment were mainly enriched in response to stimulus including calcium-mediated proteins. This study dissects the impact of UV-B stress on C.terniflora and the plant molecular resistant mechanisms through proteomic and phosphoproteomic in a new sight.

Project description:Purpose: UV-B radiation is a pivotal photomorphogenic signal and positively regulates plant growth and metabolite biosynthesis. In order to elucidate the transcriptional regulation mechanism underlying UV-B-induced artemisinin and flavonoid biosynthesis in Artemisia annua, the transcriptional response of A. annua leaves to UV-B radiation was analyzed using the Illumina transcriptome sequencing. Methods: For UV-B treatment, six-week-old A. annua seedlings growing under normal growth condition were supplemented with extra narrowband UV-B lamps (Philips TL20W/01RS; 1.5 μmol·m-2·s-1)(Yin et al. 2016). The most recently expanded leaf for each A. annua seedling was collected at 0, 2, 4 and 6 hours after exposure to UV-B radiation. Results: A total of 10706 differentially expressed genes including 533 transcription factors, were identified. Based on the expression trends of the differentially expressed factors as well as artemisinin and flavonoid biosynthesis genes, we speculated that transcription factors belonging to 6 clusters were most likely to be involved in the regulation of artemisinin and/or flavonoid biosynthesis. The regulatory relationship between transcription factors and artemisinin/flavonoid biosynthetic genes was further studied. Dual-LUC assays results showed that AaMYB6 is a positive regulator of AaLDOX, which belongs to flavonoid biosynthesis pathway. In addition, we identified a R2R3MYB transcription factor, AaMYB4 which positively mediated both artemisinin and flavonoid biosynthesis pathways by activating the expression of AaADS and AaDBR2 in artemisinin biosynthesis pathway and AaUFGT in flavonoid biosynthesis pathway. Conclusions: our findings provide fundamental knowledge for the further analysis of the parallel transcriptional regulation of artemisinin and flavonoid biosynthesis in A. annua L. under UV-B radiation.

Project description:Rice (Oryza staiva) is staple food for the human consumption and model plants for genetic and molecular studies. Only few studies have been conducted on ultraviolet-B (UV-B) stress on rice. UV-B stress effects different morphological and physiological changes in plants. However, underlying mechanisms governing these integrated responses are unknown. In this study, we conducted proteomic response of rice leaves to UV-B stress using two-dimensional gel electrophoresis and identified the selected proteins by mass spectrometry analysis. To identify changes in protein accumulation in response to UV-B stress, two cultivars, i.e., IR6 and REX were irradiated by artificial UV-B radiation. Four 4 levels of daily biologically effective UV-B radiation intensities, 0 (control), 5, 10, and 15 kJ m-2 d-1 were imposed. We conducted this experiment in Sunlit Soil-Plant-Atmosphere-Research (SPAR) chambers to mimic the natural environment.

Project description:Recently, we found a dioecious plant Populus cathayana males possess a greater tolerance to enhanced UV-B radiation than do females. To carry this work forward, comparative transcriptome analyses were carried out. Similar to previous studies, a set of conserved functions and pathways related to UV-B stress were detected in males and females, regardless of the sex. In addition, sex-specific responses via transcriptome remodeling were also detected as shown in the changes of sex-related gene expression occurred in some pathways. For example, a lot of differentially expressed genes (DEGs) involved in amino acid metabolism were mainly up-regulated in males, but down-regulated in females. Moreover, we found some DEGs expressed predominantly or exclusively in one sex, which may directly contribute to sex-related physiological responses. 4 samples examined: (i) males exposure to decreased solar UV-B radiation (MC); (ii) females exposure to decreased solar UV-B radiation (FC); (iii) males exposure to ambient solar UV-B radiation (MU); and (iv) females exposure to ambient solar UV-B radiation (FU). Nine plants of each sex were exposed to each treatment, and RNA samples from the 9 individuals were pooled with equal proportion.

Project description:In plants, ultraviolet (UV)-light is an important driver for their growth and natural distribution and is also a valuable tool for manipulating their productivity as well as their biotic interactions. Understanding plant responses to different UV radiation is sparse, especially from a systems biology perspective and particularly for conifers. Here, we evaluated the physiological and transcriptomic responses to the short-term application of high-irradiance UV-B and UV-C waves on Pinus tabuliformis Carr., a major conifer in Northern China. By undertaking time-ordered gene co-expression network analyses and network comparisons incorporating physiological traits and gene expression variation, we uncovered communalities but also differences in P. tabuliformis responses to UV-B and UV-C. Both types of spectral bands caused a significant inhibition of photosynthesis and conversely improvement of antioxidant capacity, flavonoid production and signaling pathways related to stress resistance, indicating a clear switch from predominantly primary metabolism to enhanced defensive metabolism in pine. We isolated distinct subnetworks for photoreceptor-mediated signal transduction, maximum quantum efficiency of photosystem II (Fv/Fm) regulation and flavonoid biosynthesis in response to UV-B and UV-C irradiation. From these subnetworks, we further identified phototropins as potential important elements in both UV-B and UV-C signaling and, for the first time, reveal peptide hormones possibly involved in promoting flavonoid biosynthesis against UV-B, while these hormones seem not to be implicated in defense against UV-C exposure. The present study employed an effective strategy for disentangling the complex physiological and genetic regulatory mechanisms in a non-model plant species, and thus, provides a suitable reference for future functional evaluations and artificial UV-light mediated growing strategies in plant production.

Project description:We have published results demonstrating that UV-C induces apoptotic-like changes in Arabidopsis (Danon and Gallois FEBS lett (1998) 437: 131-136). The Programmed Cell Death "phenotype" of nucleus shape, DNA laddering caspase-like activity is similar to what has been described in other plant PCD. This demonstrates that UV-C is an appropriate and controllable trigger to study PCD in plants. Using selected mutants and a set of chosen conditions we are aiming at identifying genes that are part of the PCD pathways in plants and filter out the general stress response. We are asking for a medium size experiment knowing that additional microarray analysis are likely to be required to refine the results obtained. We are currently awaiting the results of Affymetrix RNAs hybridisation of PCD-induced wild type that will define the appropriate single time point of the proposed analysis. The rationale is to vary treatments in order to distinguish changes in gene transcription arising from general cellular stress responses to the trigger used from those specific to PCD: 1. We will use wild-type Arabidopsis seedlings as a negative control to provide the basal gene expression pattern. 2. A dose of UV-C radiation of 1KJ/m2. We will irradiate wild type with this non PCD-inducing dose of UV-C radiation to identify sets of genes that respond to UV-induced damage but whose expression is not linked to cell death. 3. A dose of UV-C radiation of 50 KJ/m2. We will irradiate wild type with a PCD-inducing dose of UV-C radiation to identify sets of genes that respond to UV-induced damage and cell death. 4. We have shown that the cell death induced by UV is light dependant, the control, sub-inducing dose and inducing dose treatments will be repeated and the plants kept in the dark until RNA sampling. In those conditions there is no PCD.
