Project description:Transcriptional profiling of adult mouse liver tissue comparing offspring derived from sperm and seminal plasma of normal protein diet fed males (controls, NN), sperm and seminal plasma from males fed a low protein diet fed males (LL), sperm from normal protein fed males and seminal plasma from low protein fed males (NL) or sperm from low protein diet fed males and seminal plasma from normal protein diet males (NL). The first letter denotes the diet of the sperm donor and the second letter the diet of the seminal plasma donor. Three-condition experiment: NN vs. LL, NN vs. NL, NN vs. LN. Project description:Seminal fluid factors modulate the female immune response at conception to facilitate embryo implantation and reproductive success. Whether sperm affect this response has not been clear. We evaluated global gene expression by microarray in the mouse uterus after mating with intact or vasectomized males. Intact males induced greater changes in gene transcription, prominently affecting pro-inflammatory cytokine and immune regulatory genes, with TLR4 signaling identified as a top-ranked upstream driver. Recruitment of neutrophils and expansion of peripheral regulatory T cells were elevated by seminal fluid of intact males. In vitro, epididymal sperm induced IL6, CXCL2, and CSF3 in uterine epithelial cells of wild-type, but not Tlr4 null females. Collectively these experiments show that sperm assist in promoting female immune tolerance by eliciting uterine cytokine expression through TLR4-dependent signaling. The findings indicate a biological role for sperm beyond oocyte fertilization, in modulating immune mechanisms involved in female control of reproductive investment. Project description:Bisphenol A (BPA), a widely prevalent endocrine disruptor, has detrimental effects on human health and male reproduction. Elevated BPA levels have been linked to reduced sperm quality and production, while paternal exposure may result in poor reproductive outcomes and offspring health. Nevertheless, the impact of BPA on human sperm miRNA and protein profiles and its implications for fertilization and post-fertilization events remain unexplored. This study investigates the association between BPA concentration in seminal plasma and seminal quality and examines alterations in human sperm miRNA and protein expression among men with varying BPA levels in seminal plasma. BPA levels were measured in seminal plasma from 102 Portuguese men, and it was detected in 88% of the samples. No correlation was observed between BPA levels and age or seminal parameters. The small RNA content of 15 normozoospermic samples was evaluated using small RNA sequencing, revealing that 15 miRNAs exhibited a correlation with BPA levels. Gene ontology analysis of their target genes indicated their involvement in embryonic development and response to stress. The proteome of 20 normozoospermic human sperm samples, categorized into four groups based on BPA levels, was accessed by liquid chromatography-mass spectrometry. Sixty-two differentially expressed proteins were identified across groups. The biological processes most affected in samples with higher BPA levels were "protein sumoylation" and "cytoplasmic translation”. The altered miRNAs and proteins identified in spermatozoa from men environmentally exposed to BPA represent potential biological markers of exposure to this endocrine disruptor and may help elucidate cases of idiopathic male infertility, fertilization failure, and abnormal embryo development.

However, avoiding the local immune defense sperm can be stored for lenthy period in the oviductal sperm reservoir. It is currently unknown whether oviductal sperm reservoirs changes their gene expression to tolerate the spermatozoa after mating or sperm free seminal plasma infusion. Therefore this was tested using Swedish Landrace pigs in this study using cDNA microarray. We used 12 sows seperated into three groups- either oestrus sows were inseminated with 50 ml BTS (control, n=4) or mated with boars (treatment 1, n=4) or inseminated with sperm-free seminal plasma (treatment 2, n=4). The utero-tubal junction was retrieved within 24 h of treatment by operation.

Project description:A spectrum of reproductive strategies can manifest within the same species to help improve reproductive advantage when faced under the pressure of sexual competition, known as alternate reproductive tactics (ARTs). Chinook salmon are external fertilizing species that exhibit the sneak-guard strategy of ARTs. The larger hooknose males dominate for optimal mating position, while jack males rely on the sneak tactic to achieve fertilization in relation to their smaller body size. During copulation, a single female’s egg may be fertilized by more than one male. Therefore the threat of sperm competition is increased as the ejaculate from multiple males can interact during this time. As jack males establish a position further away from the female’s egg during fertilization attempts, it is proposed that jacks should invest more in spermatozoa quality. Although poorly understood, previous studies have suggested that differences in spermatozoa quality may play a critical role in sperm competition. Considering that the seminal plasma strongly regulates spermatozoa quality and other processes critical for fertilization success, we employed label free quantitative mass spectrometry utilizing ion mobility separation coupled to cross-species bioinformatics to examine the seminal plasma proteome of Chinook salmon.

Project description:Sperm small RNAs have emerged as important non-genetic contributors to embryogenesis and offspring health. A subset of sperm small RNAs are thought to be acquired during epididymal transit. However, the transfer of RNAs from the somatic epididymis to the sperm has been questioned, and the identity of the specific small RNAs transferred remains unclear. Here, we employ Cre/Lox genetics to generate germline- and epididymal-specific Dgcr8 conditional knockout mice to investigate the dynamics of sperm microRNAs and their function in the early embryo. Interestingly, sperm from germline specific Dgcr8 knockout males restored the levels of 58 of the 98 (59%) miRNAs that were lost in testicular sperm during epididymal transit. Conversely, sperm from epididymal Dgcr8 knockouts displayed a 5-fold reduction in 25 miRNAs. This substantial loss of epididymal miRNAs in sperm was accompanied by transcriptomic changes in the embryo which was rescued by microinjection of epididymal miRNAs. These findings ultimately demonstrate the acquisition of miRNAs by sperm during epididymal transit and their regulation of post-fertilization embryonic gene expression.

Project description:Seminal fluid proteins determine reproductive success in a wide range of animals. In the Drosophila male accessory gland, seminal fluid is mainly produced by two cell types, a vast majority of main cells and a small number of secondary cells that are characterized by a specialized secretory apparatus with unusually enlarged dense core granule vesicles. Loss of Abdominal-B expression from secondary cells in the enhancer mutant iab-6cocu disrupts their transcriptional and secretory identity. In consequence, mutant males fail to induce the long-term post-mating response, concomitant with loss of receptivity and sustained egg laying, in females. Here, we determine how secondary cells shape the seminal transferome and the female response by assessing iab-6cocu male accessory gland and female mate reproductive tract proteomes. We find downregulation of seminal fluid proteins constituting a signaling network that enables sperm binding and sustained action of key regulator Sex peptide and identify two new Sex peptide network proteins crucial for female fecundity, Cornutus (CG1701) and Hanrej (CG42564). Cornutus is required for dense core granule vesicle release, providing a link between the products of these compartments and the female long-term post-mating response. Our data highlights the importance of secondary cell signaling and secretion for overall seminal fluid composition and Sex peptide network function as well as the interdependence of main and secondary cells and their secretory products, advancing the general understanding of how seminal fluid signaling pathways modulate female physiology, sperm use and offspring production.
